Book excerpt: Excerpt from Memoirs of the Wistar Institute of Anatomy and Biology: The Development of the Lymphatic System in Fishes With Especial Reference to Its Development in the Trout My studies on the development of the lymphatic system in fishes have thus far been confined to the vessels of the head and pharynx. These vessels were chosen for the reason that they have not been dealt with by others to any extent and because they present a greater variety of conditions for study than those situated in the trunk and tail regions. The fishes thus far studied consist of ganoids and teleosts and include Amia calva, Lepidosteus osseous, Salvelinus fontinalis, Mitchill (brook trout), Salmo Gairdneri, Richardson (steelhead trout) and Salmo irideus, Gibbons (rainbow trout). The vascular system of between six and seven hundred trout embryos has been injected and the embryos studied during the process of injection, in transparent mounts after the method of Spalteholz, and in sections. Forty-two reconstructions have been made, after the method of Born, of the arteries, veins and lymphatics in the regions of the head and pharynx of Amia, Lepidosteus and the trout. These reconstructions form a fairly complete series which illustrate the development of the lymphatics from the time of their earliest appearance in the embryo, as a series of discontinuous anlagen or lymph vesicles, to the establishment of a condition in which a continuous system of lymph channels is present. Since the injection method has been employed in following the development of the lymphatics in the trout, I will deal, for the most part, with the conditions observed in this form. The lateral pharyngeal lymphatic drains the subocular lymph sac, the dorsal region of the head and pharynx and, at a slightly later stage, the operculum and the lower jaw. In fact, with the exception of one other channel (medial pharyngeal lymphatic), the lateral pharyngeal lym phatic and its tributaries, together with the subocular lymph sac, constitute, on each side of the body, the entire continuous lymphatic system found in the regions of the head and pharynx at this stage of development. The lymphatic of the lateral line of the trunk (10, fig. 10) is also completely developed on the twenty-second day and can be traced caudad into the tail region. It is a central feature of numerous post-embryonic processes, including tissue and organ growth and regeneration. It is also part of the pathology of tumour formation and certain inflammatory conditions. In recent years, comprehension of the molecular genetics of blood vessel formation has progressed enormously and studies in vertebrate model systems, especially the mouse and the zebrafish, have identified a common set of molecules and processes that are conserved throughout vertebrate embryogenesis while, in addition, highlighting aspects that may differ between different animal groups. The discovery in the past decade of the crucial role of new blood vessel formation for the development of cancers has generated great interest in angiogenesis (the formation of new blood vessels from pre-existing ones), with its major implications for potential cancer-control strategies. In addition, there are numerous situations where therapeutic treatments either require or would be assisted by vasculogenesis (the de novo formation of blood vessels). In particular, post-stroke therapies could include treatments that stimulate neovascularization of the affected tissues. The development of such treatments, however, requires thoroughly understanding the developmental properties of endothelial cells and the basic biology of blood vessel formation. While there are many books on angiogenesis, this unique book focuses on exactly this basic biology and explores blood vessel formation in connection with tissue development in a range of animal models. It includes detailed discussions of relevant cell biology, genetics and embryogenesis of blood vessel formation and presents insights into the cross-talk between developing blood vessels and other tissues. Book excerpt: When Budge in the eighth decade of the nineteenth centlll'Y, started his investigations into the origin and development of the lymphatic system, he probably did not expect that these would be the starting-point for disputes that would continue well into our time. But rather than by his results were these disputes intensified by the controversial conclusions of some papers published by Gulland (1894), Ranvier (1897) and Sala (1900), and above all by some new investigations by Sabin (1902-1913) and Huntington (1908-1914) during the first decades of the twentieth century. The most important controversial point was the difference in opinion about the origin of the lymphatic system. An extensive review of the relevant literature is found in O. F. Kampmeier's book: "Evolution and Comparative Morphology of the Lymphatic System"; Thomas, Springfield, U.S.A. (1969). Whereas some investigators were of the opinion that the lymphatic system was formed from the venous system, there were many others, who believed that this system originates from confluent spaces in the mesenchyme and secondarily communicates with the veins. The most prominent advocate of the first mentioned opinion was Sabin (1902-1913). She believed that the lymphatics are formed by a process of sprouting from the large central veins in a limited number of definitely localized areas, but that these sprouts demonstrate almost immediately their own lymphatic character. From these sprouts separate primordia develop at first; these enlarge, confluence and form new sprouts which grow out into more peripheral parts of the embryo.
