Book excerpt: In this work, use of immobilized luminol for the CL determination of hydrogen peroxide using flow injection analysis (FIA) was investigated. Nylon was initially chosen as the immobilization support because of its chemical inertness. Luminol was covalently immobilized on the inside of nylon tubing and used for FIA determination of peroxide using solution-phase hemin as catalyst (detection limit = 0.4 $mu$M, relative standard deviation (rsd) = 2.5%). Next, horseradish peroxidase (HRP) was immobilized on nylon membranes. Luminol was immobilized on to an anion exchange resin and packed in a column. When used in the FIA mode, stoichiometric amounts of luminol could be released from the column by injecting another anion with higher affinity towards the resin. The pH downstream of the column could also be controlled by the amount of anion injected upstream of the column. Such columns were used in a single channel FIA scheme for the determination of peroxide in conjunction with immobilized HRP as catalyst (detection limit = 8 $mu$M, mean rsd = 0.8%). The experiment was extended for the determination of glucose by using co-immobilized HRP and glucose oxidase (detection limit = 25 $mu$M). The problem presented by real samples was solved using a sample pre-treatment technique. The CL of luminol-Ru(bpy)$sb3sp{3+}$-peroxide system was investigated. It was found that CL is produced when the potential of a glassy carbon (GC) electrode coated with Nafion and loaded with Ru(bpy)$sb3sp{2+}$ is raised above 0.95 V in the presence of luminol and peroxide. The shape of the ECL vs applied potential curve obtained in this case was markedly different from that obtained using a bare GC electrode. The modified electrode (detection limit = 0.5 $mu$M) as well as the bare GC electrode (detection limit = 0.2 $mu$M) were used for peroxide determination at pH 8 in the presence of 10 $mu$M luminol. In the presence of 1 mM peroxide, luminol could be detected down to 0.5 nM with the modified electrode. The modified electrode was also used for amperometric detection of peroxide (detection limit = 0.08 $mu$M at pH 7.5).

Book excerpt: A flow injection analysis (FIA) method for the determination of hydrogen peroxide in reactor moderator water was developed and installed at the Savannah River Site (SRS) Water Quality Laboratory. The technique has an analytical range of 0.10 to 2.50 ppm (ug/mL) with a sampling rate of 40 samples per hour. The calibration curve is linear with a correlation coefficient of 0.999, and the precision is excellent with relative standard deviations at the 0.50% level for both 0.10 and 2.50 ppm standards. When the automated FIA procedure is compared to the manual method it demonstrates a twenty minute reduction in analysis time per sample, and the total liquid waste generated per sample analyzed is reduced by roughly 95 mL. 6 refs., 10 figs., 6 tabs.

Book excerpt: A hydrogen peroxide (H2O2) measurement scheme was developed using an inexpensive and microscale photodiode-based luminometer. The measurement of H2O2 was based on the chemiluminescence (CL) emitted by the Co(II)-catalyzed luminol-H2O2 system. The proposed technique employed two injection systems: batch-wise and flow injection methods. Reaction and instrument-centered parameters were systematically optimized. Under optimum conditions, the working ranges were linear from 7.08 x 10−6 to 1.91 x 10−4 M H2O2 (R = 0.989) and 1.18 x 10−4 to 9.56 x 10−3 M H2O2 (semi-logarithmic scaling) (R = 0.984) for the batch-wise and flow injection CL methods, respectively. The detection limit calculated using the 3 SD/m criterion for the batch-wise method is 5.8 x 10−7 M H2O2 (n = 11). For the flow injection method, the detection limit based on the lowest concentration in the calibration curve is 1.18 x 10−4 M H2O2 (n = 3). Excellent repeatability and reproducibility were recorded for both injection methods. A total of 180 and 120 samples/hour can be analyzed using the batch-wise and flow injection CL method, respectively. The measurement system was evaluated using pharmaceutical and cosmetic H2O2 preparations and the method was validated through the official method of analysis of the association of official analytical chemists (AOAC) for H2O2 assay, iodometric titration. Statistical tests revealed that there is no significant difference between the measurements obtained using the CL and titrimetry methods. The developed method can be an alternative technique for H2O2 quantification by virtue of its simplicity, short analysis time, sensitivity, excellent precision, cost-effectiveness as well as battery-operation capability and field-portability (batch-wise method) and semi-automation capacity (flow injection method).
