These approaches emerge from the recent work in tu mor virology, e. g. the control of vital cellular genes by viral regulatory signals; the implication of monoclonal antibodies as a vehicle for the targeted drug delivery and selective de struction of tumor cells; immunologic advances in the recog nition of some specific events during metastatic growth; the role of biological response modifiers in modifying or rever sing malignant growth; and biochemical advances, such as the role of gene amplification in drug resistance and the app roaches to the reversal of resistance in drug refractory cancer, the role of membranes in designing useful strategies, the identification of new enzymic targets in some types of cancer cells, and the characterization of metabolically active forms of cytostatic compounds. These are the important issues which were addressed in an international NATO Advanced Study Institute (ASI) attended by scientists from allover the world representing a wide spectrum of scientific disciplines. Although the acquired immune deficiency syndrome (AIDS) is not a malignant disease, but the etiological involvement of a retrovirus belonging to the family of human T-cell lympho tropic viruses which cause leukemia and lymphomas in man, justifies its inclusion in the agenda of this Study Institute. Furthermore, intensive investigations in progress were bringing sophisticated methodologies to bear on the problems in the field, clinical trials were yielding interesting results, and unique approaches to cancer therapy were being designed. Therefore, an organizing committee was formed and planning culminated in the symposium which was held in Burlington, Vermont, June 22-24, 1983.

Book excerpt: The observation in the 1950s that nitrogen mustard and other toxic chemicals could induce antitumor responses in patients with refractory lymphoma initiated a massive search for active chemotherapeutic agents. The initial observations stimulated a search for new chemotherapeutic agents which might have increased antitumor activity with less toxicity for normal tissues. To aid in the search for these new chemicals and to attempt to distinguish among the many toxic chemicals which might be candidates for clinical studies, the National Cancer Institute, the pharmaceutical industry, and the cancer research laboratories of most Western nations developed systems for "screening" drugs for antitumor activity. Perhaps the most extensive screening program was established by the National Cancer Institute (1). This screening program has evolved over the last two decades, an evolution which has been repeatedly reviewed (2-5). Various screening programs in use have examined over 500,000 compounds as potential anticancer agents. From these, there are now approximately forty anticancer drugs in clinical use. The utiliy of these compounds and their toxicities have been reviewed on many occasions. It is now apparent that more active and less toxic anticancer drugs are needed. It is also clear that the current screening programs are identifying compounds with similar levels of activity and with continuing moderate to severe toxicity (6).
