Available in PDF, EPUB and Kindle. Book excerpt: The Series on Antibiotics produced by Springer-Verlag began more than a decade ago with the nearly simultaneous appearance of two volumes, one dealing with the mode of action of antibiotics and the other concerning the biosynthesis of them. The standards set by the original Editors were high, and these books have proved useful to many. The rapid advances in our knowl edge of the mode of action of antibiotics and other antitumor agents has stimu lated two further works in the same series (Volume III, 1975; and Volumes Vj1 and Vj2, 1979). For some time it had appeared to Dr. Konrad Springer that the time might' be ripe' for bringing the subject of the biosynthesis of antibiotics up-to-date. This Editor agreed to survey the literature and discuss this possibility with his colleagues who are active in research on antibiotics. In spite of the appearance of numerous review articles, both of a highly special ized and general nature, on the biosynthesis of antibiotics, it was agreed generally that it would be extremely useful to add a new volume on biosynthesis to the Series. Such a work should focus on collecting a group of contributions dealing with those antibiotics whose biosynthesis is understood in much greater detail now than it was in the middle 1960's. Since Volume II on biosynthesis continues to be available, this addition to the series has not dealt with each and every antibiotic whose biosynthesis was studied long ago.

This book was released on 2009-04-11 with total page 644 pages. Available in PDF, EPUB and Kindle. Book excerpt: Microbial natural products have been an important traditional source of valuable antibiotics and other drugs but interest in them waned in the 1990s when big pharma decided that their discovery was no longer cost-effective and concentrated instead on synthetic chemistry as a source of novel compounds, often with disappointing results. Moreover understanding the biosynthesis of complex natural products was frustratingly difficult. With the development of molecular genetic methods to isolate and manipulate the complex microbial enzymes that make natural products, unexpected chemistry has been revealed and interest in the compounds has again flowered. This two-volume treatment of the subject will showcase the most important chemical classes of complex natural products: the peptides, made by the assembly of short chains of amino acid subunits, and the polyketides, assembled from the joining of small carboxylic acids such as acetate and malonate. In both classes, variation in sub-unit structure, number and chemical modification leads to an almost infinite variety of final structures, accounting for the huge importance of the compounds in nature and medicine. Gathers tried and tested methods and techniques from top players in the field Provides an extremely useful reference for the experienced research scientist Covers biosynthesis of Polyketides, Tarpenoids, Aminocoumarins and Crabohydrates

Book excerpt: This book was developed from the proceedings of the American Chemical Society, Division of Agricultural & Food Chemistry, subdivision of Natural Products Symposium "Biosynthesis and Metabolism of Secondary Natural Products" held in Atlanta, Georgia, April 1991. The objective of the conference was to bring together people from apparently diverse fields, ranging from biotechnology, metabolism, mechanistic organic chemistry, enzymology, fermentation, and biosynthesis, but who share a common interest in either the biosynthesis or the metabolism of natural products. It is our intention to help bridge the gap between the fields of mechanistic bio-organic chemistry and biotechnology. Our thanks go to Dr. Henry Yokoyama, co-organizer of the symposium, the authors who so kindly contributed chapters, the conference participants, and to those who assisted in the peer review process. We also thank the financial supporters of the symposium: ACS/AGFD, NIH General Medical Sciences, and the agricultural, pharmaceutical, biotechnology, and chromatography companies. A full list of the supporting corporations and institutions is given on the following page. Pharma-Tech and P.C., Inc. are manufacturers of instrumentation for high-speed countercurrent chromatography. We thank the Agricultural Research Service and the U. S. Department of Agriculture for granting me permission to co-organize the conference and for us to complete the book. Richard J. Petroski Susan P. Available in PDF, EPUB and Kindle. Book excerpt: Authored by leading experts in the enzymology of natural product biosynthesis, this completely revised and updated edition provides a description of the types of natural products, the biosynthetic pathways that enable the production of these molecules, and an update on the discovery of novel products in the post-genomic era. Although some 500 000 – 600 000 natural products have been isolated and characterized over the past two centuries, there may be a 10-fold greater inventory awaiting immediate exploration based on biosynthetic gene cluster predictions. The approach of this book is to codify the chemical logic that underlies each natural product structural class as they are assembled from building blocks of primary metabolism. This second edition integrates many new findings into the sets of principles of the first edition that parsed categories of natural product chemistries into the underlying enzymatic mechanisms and the catalytic machinery for building the varied and complex end product metabolites. New chapters include evaluation of a core set of thermodynamically activated but kinetically stable metabolites that power both primary and secondary metabolic pathways. Also, after decades of uncertainty about the existence of various pericyclase classes, a series of genome mining, heterologous expression, and enzymatic activity characterization have validated a plethora of pericyclases over the past decade. The several types of pericyclases are involved in biosynthetic complexity generation of almost every major category of natural products. This text will serve as a reference point for chemists of every subdiscipline, including synthetic organic chemists and medicinal chemists. It will also be valuable to bioinformatic and computational biologists, pharmacognocists and chemical ecologists, and bioengineers and synthetic biologists.

Available in PDF, EPUB and Kindle. Book excerpt: Asymmetric Synthesis of Natural Products Fully updated learning resource covering the concept of using natural product chemistry for strategies in asymmetric synthesis The third edition of Asymmetric Synthesis of Natural Products introduces students to the rapidly growing field of natural products in organic chemistry, discussing the practical, mainly pharmacological, importance of selected compounds and emphasizing the target-oriented approach of organic synthesis which is key in industrial strategies. To aid in reader comprehension, the text includes key references and an Index of Compounds. The textbook is based on two lecture courses (Asymmetric Synthesis & Asymmetric Synthesis of Natural Products), which the author has delivered more than 50 times over the past 20 years in Finland, the UK, Italy, and Greece. This third edition is fully updated from the earlier versions (published by Wiley in 1993 and 2012). The importance of natural products as truly renewable raw materials in sustainable chemistry and circular economy is illustrated through applications of e.g. organocatalysis, organometallic catalysis, and biocatalysis. The contents consist of traditional text supplemented with illustrations (such as chemical drawings and structural formulae). Three dimensional aspects are also discussed with the use of 3D renderings of structures for both reaction mechanisms (molecular modeling) and crystallographic data. Sample topics covered in the textbook include: The foundations of asymmetric synthesis, including the theory and applications of individual asymmetric reactions Sustainable development, the circular economy, and use of renewable raw materials that have become prominent in many fields of science and technology Various natural product classes, including carbohydrates, amino acids, peptides, proteins, nucleosides, nucleotides, nucleic acids, and polyketides The properties of these natural product classes, including their structures, biosynthesis, and interrelationships, as well as examples of asymmetric syntheses and the practical value of these compounds Asymmetric Synthesis of Natural Products is a comprehensive, authoritative, and up-to-date learning resource on the subject for advanced level undergraduate or early-stage graduate students. It is also useful for specialists already working in synthesis who wish to learn about asymmetric synthesis.

Book excerpt: Asymmetric Synthesis, Volume 4: The Chiral Carbon Pool and Chiral Sulfur, Nitrogen, Phosphorus, and Silicon Centers describes the practical methods of obtaining chiral fragments. Divided into five chapters, this book specifically examines initial chiral transmission and extension. The opening chapter describes the so-called chiral carbon pool, the readily available chiral carbon fragments used as building blocks in synthesis. This chapter also provides a list of 375 chiral building blocks, along with their commercial sources, approximate prices, and methods of synthesis. Schemes involving almost 1,300 structures and the use of chiral building blocks to prepare alkaloids, arachidonic acid metabolites, compounds with chemotherapeutic and pharmacological activity, insect pheromones, sugars, vitamins, terpenoids, and miscellaneous compounds are also included in this text. The remaining chapters provide a similar overview of the chemistry of compounds chiral at sulfur, phosphorus, nitrogen, and silicon. Synthetic chemists and researchers, especially those engaged in pharmaceutical research and specialty chemicals production, will find this book invaluable.
