This is because the cuticle is commonly regarded as an inert substance. It is hoped that this book will dispel this fallacy. The study of cuticle in its proper context now involves many of the wider aspects of biology which are currently in vogue (e. g. how a hormone like ecdyson induces a specific enzyme like dopa decarboxylase; the unsolved major problem of cell gradient and polarity; the involvement of cyclic AMP in hormonal mechanisms; the extra cellular control of cuticular enzymes, of the mechanical proper ties of cuticle structural proteins, and of the orientation of fibrous molecules; and the relation of chromosome puffing to the synthesis of specific proteins). Studies on cuticle demand a variety of techniques, and examples of the following are illustrated in this book (fluorescence, phase contrast, polariza tion and Nomarski interference microscopy; infrared absorp tion; transmission and scanning electron microscopy; autora diography analyzed by electron microscopy; negative staining in the electron microscope; optical diffraction, high angle X-ray diffraction, low angle X -ray diffraction and selected area electron diffraction). I am well aware that the biophysical parts of this book are less incomplete than other aspects. A developmental biologist or a biochemist would have further elaborated other parts ofthe subject matter. Only one previous author, RICHARDS (1951)hasdevoted a book to arthropod cuticle.

The size and general nature of insects present special problems to the student of entomology. Therefore, tech niques developed in the study of insects or in those fields concerned with the control of insect pests are often unique. Methods for measuring things are common to all sciences. Advances sometimes depend more on how something was done than on what was measured; indeed a given field often progresses from one technique to another as new methods are discovered, developed, and modified. Just as often, some of these techniques find their way into the classroom when the problems involved have been sufficiently ironed out to permit students to master the manipulations in a few laboratory periods. Although methods may be considered commonplace where they are used, in another context even the simplest procedures may save con siderable time. In the meantime, palaeontology was revealing an amazing number of extinct forms that on the one side have contributed to a radical revisitation of arthropod phylogeny, but on the other have provided evidence of a previously unexpected disparity of arthropod and arthropod-like forms that often challenge a clear-cut delimitation of the phylum.

Here the results of a decade of electronmicroscopic studies on insect cells were summarized in an organized and integrated fashion for the first time, and the ultrastructural characteristics of different specialized cells and tissues were abundantly illustrated in the 117 plates this monograph contained. In the intervening period great progress has been made in the field of Insect Ultrastructure. Organelles not even mentioned in Smith's book, such as synaptonemal complexes, clathrin baskets, fusomes, and retinular junc tions, have been identified and functions proposed for them. There have also been many technical advances that have profoundly influenced the direction of subsequent research. A spectacular example would be the development by Miller and Beatty of the chromosomal spreading technique which allowed for the first time ultrastructural studies on segments of chromosomes containing genes in various stages of replication and transcription. Then there is the freeze-fracture procedure first described by Moor and his colleagues. This technique permitted an analysis of intercellular junctions that was impossible with the conventional sectioning methods. The results greatly clarified our understanding of the channels for ion movement and the permeability barriers between cells and also the membrane changes that occur during the embryonic differentiation and metamorphosis of various types of insect cells.

It tackles a major unsolved problem in developmental biology - how does chemistry create architecture outside cells? Fibrous composites occur in all skeletal systems including plant cell walls, insect cuticles, moth eggshells, bone and cornea. They function like man-made fibreglass, with fibres set in a matrix. The fibrous molecules are long, extracellular and water-insoluble and to be effective they must be orientated strategically. The underlying hypothesis of this book is that the fibres are orientated by self-assembly just outside the cells during a mobile liquid crystalline phase prior to stabilization. The commonest orientations of the fibres are plywood laminates (orthogonal and helicoidal), and as parallel fibres. These may be imitated in vitro by liquid crystalline chemicals. The book takes an interdisciplinary approach and will be relevant to biologists, biochemists, biophysicists, material scientists and to liquid crystals chemists.
