Book excerpt: Knowledge of the mechanism of action of drugs at cellular, subcellular, or molecular levels is of vital importance not only in giving the basis of inter pretation of the systemic action of drugs but also in improving existing drugs; in designing new forms of drugs; and in giving the basis of therapeutic applications. Classical pharmacology, concerning the action of drugs at integrated levels, does not necessarily give sufficient information as to the mechanism of action of drugs. A variety of sophisticated concepts utilizing the methods of physics, chemistry, biophysics, biochemistry, and physiology must be synthesized to understand the mechanism of action. Only since the last decade, however, have these techniques been fully applied to pharma cological investigations. It is of utmost importance to realize that a new dimension of pharmacological research has indeed emerged as a result of such a multidisciplinary approach; this approach is encompassed in general and cellular pharmacology. Such recent studies of drug actions have led to a number of important findings. Certain chemicals and drugs were found to possess highly specific actions on cellular functions, so that they are widely being used as powerful tools for the study of a variety of physiological and pharmacological prob lems. Our knowledge of the cellular mechanisms of drug action has provided the basis for interpreting the systemic effects of the drugs and insight into the molecular mechanism involved.

Available in PDF, EPUB and Kindle. Book excerpt: Most drugs, toxins, hormones, and the like bring about their biologic actions by reacting with specific receptors somewhere in the body. Scientists working in all areas of biologic science have shown increasing interest in the analysis of drug-receptor interactions in the broadest sense. Studies of drugs (binding) to receptors in situ and to isolated and partly purified receptors are becoming common practice. The action of a drug in the body is, however, a kinetic event not only with respect to transport of drug molecules to the environment of the receptors, but also with respect to the drug-receptor interaction itself. Kinetics of Drug Action is an integrative approach to drug transport through the body, membrane transport toward the receptors, and the kinetics of drug receptor interaction. This volume is aimed at providing a critical and penetrating study of the problems relevant to the kinetics or drug action from drug dosage to the final response. It is felt that the critical surveys presented in this volume will contribute significantly to receptor study research in various biologic fields and to a better understanding of drug action. I would like to express my gratitude to our secretary Miss MARGOT JANSSEN for the extensive typing of manuscripts and to our laboratory assistant Miss COBY HURKMANS for her dedicated assistance in the correcting some of the manuscripts and preparating the index.

Book excerpt: Surface activity is present in living systems; for example in body fluid or cell soup and molecules of surface-active nature are crucial to living matter and its organization. Surface Activity in Drug Action proposes "a liquid membrane hypothesis of drug action" for surface-active drugs. Chapters 1-7 contains an account of the hypothesis and chapter 8 contains a general account of the application of surface activity in therapeutics. The methodology and presentation of the information makes Surface Activity in Drug Action valuable reading for students and researchers interested in surface activity. * Is clearly written * Includes contributions from prominent names in the field, such as Bhise and Subrahmanyam* Contains a general account of the application of surface activity in therapeutics

Book excerpt: This report from the Committee on Military Nutrition Research reviews the history of caffeine usage, the metabolism of caffeine, and its physiological effects. The effects of caffeine on physical performance, cognitive function and alertness, and alleviation of sleep deprivation impairments are discussed in light of recent scientific literature. The impact of caffeine consumption on various aspects of health, including cardiovascular disease, reproduction, bone mineral density, and fluid homeostasis are reviewed. The behavioral effects of caffeine are also discussed, including the effect of caffeine on reaction to stress, withdrawal effects, and detrimental effects of high intakes. The amounts of caffeine found to enhance vigilance and reaction time consistently are reviewed and recommendations are made with respect to amounts of caffeine appropriate for maintaining alertness of military personnel during field operations. Recommendations are also provided on the need for appropriate labeling of caffeine-containing supplements, and education of military personnel on the use of these supplements. A brief review of some alternatives to caffeine is also provided.

Book excerpt: An understanding of the mechanism of action of anticancer agents depends on investigations into the cell cycle effects of these agents. The intent of this volume is to review our current knowledge about these basic cell cycle effects and suggest what directions future investigations should take. Chapters 1 and 2 are general, basic chapters dealing with the biology of the cell cycle per se and quantitative cytokinetics (techniques) respectively. The remaining nine chapters each cover a specific anticancer compound as examples of various classes of anticancer agents. In bringing this information together for the first time this volume represents a major step in the understanding of the mechanisms that are involved in the treatment of cell cycle disorders.
