A huge limitation in RNA silencing therapy is the lack of a suitable delivery mechanism. While hydrogels for RNA delivery this purpose rely on supramolecular chemistries to bind RNA molecules to the gel matrix, none covalent interactions. These supramolecular interactions are often poorly characterized, and, due to a less-than-optimal stability, can have limitations in therapeutic application. In this work, we propose a means of tunable, slow, and sustained local siRNA delivery.In our work, thiol-functionalized siRNA was reacted via Michael addition with acrylate-functionalized dextran polymers with ester groups that allow for degradation by hydrolysis. Following this coupling, radical chemistry was employed to crosslink the dextran molecules into a hydrogel network. As the gel matrix decayed, we determined that siRNA molecules were released. By changing the concentration of the dextran polymer matrix, release profiles of siRNA are achieved.
