Book excerpt: This thesis addresses fundamental scientific questions such as: How are complex natural products synthesized in vivo? Can we replicate these conditions in a laboratory environment? What is the biological function of such secondary metabolites? What are the biological origins of chirality? These issues are explored in an accessible manner using a multidisciplinary approach spanning chemistry, biology and physics to investigate an interesting family of complex natural products isolated from marine molluscs – the tridachiahydropyrones. The work has achieved: Elegant biomimetic syntheses of a number of the tridachiahydropyrone compounds in vitro using organic synthesis techniques The characterization of the interactions between these compounds and a range of model membrane systems using a series of fluorescence spectroscopic studies The investigation of the antioxidant and photoprotective properties of the compounds by means of biophysical assay techniques The synthesis of tridachiahydropyrone utilizing the model membrane systems as biomimetic reaction media.

Book excerpt: Marine natural products (MNPs) are, undoubtedly, highly valuable starting points for drug-development programs. Generally, MNPs show a well-defined spatial orientation that have evolved for thousands of years to interact with their biological target and the special and unique conditions present in seas, lakes, oceans and rivers allow the biosynthesis of highly potent compunds. Nowadays, the processes to discover a new drug from MNPs are bio-assay guided, giving more importance to the biological results rather than to the molecular structure. After a molecular entity has shown to be biologically relevant the next step is to determine the structural and stereochemical information of such molecule. It is at this step where the project described in this doctoral thesis started. Phormidolides B, C and D (PM B-D) is a family of complex molecules that were isolated in 2010 by the pharmaceutical company Pharmamar S.A. during an expedition to collect samples in the Pemba Island (Madagascar). The extracts rendered a mixture of PM B-D that, once purified, were tested on an IC50 assay against three tumour cell lines. The results showed activity in the low micromolar range with a still unknown mechanism of action. After extensive HRMS, 1D-NMR and 2D-NMR techniques a possible structure was proposed. Following previous UB doctoral theses (Dr. Adriana Lorente and Dr. Janire Lamariano) this doctoral thesis describes, as a compendium of publications, several synthetic studies with the final objective of achieving a robust total synthesis to confirm the structural information of PM B-D by chemical synthesis. After a general introduction (Chapter 1), Chapter 2 (Org. Lett. 2015, 17, 6246.) describes an optimization for the synthesis of the macrocyclic core present in PM B-D. Chapter 3 (Chem. Eur. J. 2016, 21, 7033.) deals, for the first time, with the synthesis and introduction of the most challenging part of the molecule: the bromo-methoxy-diene (BMD) moiety present at the end of the polyhydroxylated chain. After this finding, Chapter 4 (Org. Lett., 2016, 18, 4485.) illustrates the enantioselective synthesis of the polyol chain present in Oscillariolide and Phormidolides, including the previously mentioned BMD moiety. Chapter 5 reports the preparation of the fatty acids present in PM C and D. Finally, after the previous synthetic optimizations, Chapter 6 describes the first synthetic approach for the construction of this large family of natural products using a highly convergent approach. Finally, as an annex a review article entitled "Role of the Nozaki-Hiayama-Kishi-Takai coupling in the synthesis of natural products" (Chem. Rev., 2017, 117, 8420.) is included to deeply explain the uses of this reaction used, as well, for the synthesis of phormidolide B-D. A section with the general conclusions extracted from this thesis is also included.

Book excerpt: Underwater exploration is a fascinating activity because of the richness of sea life, the beautiful colors, and the contrast with terrestrial life. It can be done by diving or snorkeling -today there is fine equipment for diving and taking underwater pictures and, apart from being a source of pleasure, it forms the basis of modern ecological and behavioral marine studies. Diving is an emotional activity. The silence of the submarine world, only interrupted by breathing, has more than once evoked the monsters of the fabulous tales of my youth. Maybe because, although much time has elapsed since then, I still have a youth's ability to evaluate everything for the fascination it conveys. However, no more than that can be gained from underwater watehing. I am not saying that this is destined to be a futile exercise. Picasso watched sea urchins much in the same way while revealing their quintessence in his paintings at the Castle of Antibes. What I mean is that today marine life can also be viewed from the perspective of the natural products involved. This knowledge, subsequent to Picasso's time, adds much to the overall picture.

Book excerpt: This book highlights the latest international research on different aspects of medicinal plants and fungi. Studies over the last decade have demonstrated that bioactive compounds isolated from medicinal fungi have promising antitumor, cardiovascular, immunomodulatory, anti-allergic, anti-diabetic, and hepatoprotective properties. In the light of these studies, the book includes chapters (mostly review articles) by eminent researchers from twelve countries across the globe working in different disciplines of medicinal plants and fungi. It discusses topics such as the prevention of major neurodegenerative and neurotoxic mechanisms by Centella asiatica; the medicinal properties and therapeutic applications of several mushrooms species found in different parts of the world; and fungal endophytes as a source of bioactive metabolites including anticancer and cardioprotective agents. There are also chapters on strategies for identifying bioactive secondary metabolites of fungal origin; the use of genomic information to explore the biotechnological potential of medicinal mushrooms; and solid state fermentation of agro-industrial and forestry residues for the production of medicinal mushrooms. It is a valuable resource for the researchers, professionals and students working in the area of medicinal plants and fungi.

Nicolaou and E. Sorensen present the most remarkable and ingenious total syntheses from outstanding synthetic organic chemists. To make the complex strategies more accessible, especially to the novice, each total synthesis is analyzed retrosynthetically. The authors then carefully explain each synthetic step and give hints on alternative methods and potential pitfalls. Numerous references to useful reviews and the original literature make this book an indispensable source of further information. Special emphasis is placed on the skillful use of graphics and schemes: Retrosynthetic analyses, reaction sequences, and stereochemically crucial steps are presented in boxed sections within the text. For easy reference, key intermediates are also shown in the margins. Graduate students and researchers alike will find this book a gold mine of useful information essential for their daily work. Every synthetic organic chemist will want to have a copy on his or her desk.

Book excerpt: The Shikimate Pathway gives a bird's eye view of the shikimate pathway and its implications for the life of a range of organisms. Topics covered in this book include the chemistry of intermediates in the shikimate pathway; biosynthesis of aromatic amino acids in this pathway; its metabolites; and its role in higher plants. This book is comprised of six chapters and begins by introducing the reader to shikimic acid, a natural product derived from the plant Illicium religiosum, along with the mechanistic and stereochemical aspects of the reactions of the shikimate pathway. The biosynthesis of aromatic amino acids from chorismate is also described, and then the discussion turns to the chemical properties and the detailed stereochemistry of intermediates and enzymes in the shikimate pathway. The next chapter examines the biosynthesis of isoprenoid quinones involved in electron transport and the folic acid group of co-enzymes in the shikimate pathway. The metabolism of the aromatic amino acids in microorganisms and higher organisms is considered, along with the biosynthesis and physiological functions of phenylpropanoid compounds and their derivatives in the shikimate pathway in higher plants. This book will be of general value to practitioners in the many and varied areas of biochemical research associated with metabolism.

This book provides a state-of-the-art account of a few selected hot spots by scientists at the edge in this extremely active field. It puts special emphasis on two main streams of research. One is the role of post-translational modifications of proteins, mostly histones, on chromatin structure and accessibility. The other one deals with parental genomic imprinting, a process which allows to express a few selected genes from only one of the parental allele while extinguishing the other.
