Book excerpt: Iron oxide nanoparticles are highly researched for their use in biomedical applications such as drug delivery, diagnosis, and therapy. The inherent biodegradable and biocompatible nanoparticle properties make them highly advantageous in nanomedicine. The magnetic properties of iron oxide nanoparticles make them promising candidates for magnetic fluid hyperthermia applications. Designing an efficient iron oxide nanoparticle for hyperthermia requires synthetic, surface functionalization, stability, and biological investigations. This research focused on the following three areas: optimizing synthesis conditions for maximum radiofrequency induced magnetic hyperthermia, designing a simple and modifiable surface functionalization method for specific or broad biological stability, and in vitro and in vivo testing of surface functionalized iron oxide nanoparticles in delivering effective hyperthermia or radiotherapy. The benzyl alcohol modified seed growth method of synthesizing iron oxide nanoparticles using iron acetylacetonate as an iron precursor was investigated to identify significant nanoparticle properties that effect radiofrequency induced magnetic hyperthermia. Investigation of this synthesis under atmospheric conditions revealed a combination of thermal decomposition and oxidation-reduction mechanisms that can produce nanoparticles with larger crystallite sizes and decreased size distributions. Nanoparticles were easily surface functionalized with (3-Glycidyloxypropyl)trimethoxysilane (GLYMO) without the need for organic-aqueous phase transfer methods. The epoxy ring on GLYMO facilitated post-modifications via a base catalyzed epoxy ring opening to obtain nanoparticles with different terminal groups. Glycine, serine, [gamma]-aminobutryic acid (ABA), (S)-( - )-4-amino-2-hydroxybutyric acid (SAHBA), ethylenediamine, and tetraethylenepentamine were successful in modifying GLYMO coated-iron oxide nanoparticles to provide colloidal and varying biological stability while also allowing for further conjugation of chemotherapeutics or radiotherapeutics. The colloidal stability of cationic and anionic nanoparticles in several biologically relevant media was studied to address claims of increased cellular uptake for cationic nanoparticles. The surface functionalized iron oxide nanoparticles were investigated to determine effects on cellular uptake and viability. In vitro tests were used to confirm the ability of iron oxide nanoparticles to provide effective hyperthermia treatment. S-2-(4-Aminobenzyl)-1,4,7,10-tetraazacyclododecane tetraacetic acid (DOTA) was coupled to SAHBA and carboxymethylated polyvinyl alcohol surface functionalized iron oxide nanoparticles and radiolabeled with 177Lu. The capability of radiolabeled iron oxide nanoparticles for delivering radiation therapy to a U87MG murine orthotopic xenograft model of glioblastoma was initially investigated.

Book excerpt: This thesis presents experimental studies of iron oxide nanoparticle synthesis, functionalization, and intracellular hyperthermal effects on murine macrophages as a model in vitro system. Colloidal suspensions of magnetic nanoparticles (MNPs) are of particular interest in Magnetic Fluid Hyperthermia (MFH). Iron oxide nanoparticles (IONPs) have garnered great interest as economical, biocompatible hyperthermia agents due to their superparamagnetic activity. Here we seek to optimize the synthetic reproducibility and in vitro utilization of IONPs for application in MFH. We compared aqueous synthetic protocols and various protective coating techniques using various analytical techniques and in vitro assays to assess the biocompatibility and feasibility of the various preparations of nanoparticles. Using a co-precipitation of iron salts methodology, iron oxide nanoparticles (IONPs) with an average diameter of 6-8nm were synthesized and stabilized with carboxylates. By performing calorimetry measurements in an oscillating magnetic field (OMF) with a frequency of 500 kHz and field strength of 0.008Tesla the superparamagnetic behavior of these particles was confirmed. To further investigate these IONPs in a biological application, citric acid-stabilized particles, in conjunction with heat generated by these IONPs when exposed to an OMF, were assessed to determine their effects on cell viability in a RAW 267.4 murine macrophage model system. Our results show that 91.5-97% of cells that have ingested IONPs die follow exposure to an OMF. Importantly, neither the IONPs (at applicable concentrations) nor the OMF show cytotoxic effects. These particular particles have promising preliminary results as hyperthermic agents in both the current literature and simple, proof-of-concept experiments in our laboratory setting. We present experimental results for the synthesis, characterization, and utilization of iron oxide nanoparticles in MFH. Our results show that while IONPs have potential in MFH, efforts to advance IONPs from laboratory to clinical contexts, where reliable generative techniques and consistent performance properties are necessary, will require an understanding of the influence of the diverse intrinsic structural and magnetic characteristics as well as surface chemistry of nanoparticles, as well as the mechanisms of particle uptake and cell death due to intracellular hyperthermia.

Iron oxide nanoparticles have been widely explored in localized therapy, targeted delivery, and magnetic resonance imaging. However, inaccessible MRI instruments for most research labs and lack of targeting ligands limits the further exploration of iron oxide nanoparticles in routine tumor diagnosis and efficient therapy. Therefore, the preparation of dual-imaging or targeted nanoparticles is highly desirable. In our studies, fluorescent gold nanoclusters or anti & ndashdisialoganglioside & ndashGD2 monoclonal antibodies are integrated onto iron oxide nanoparticle surfaces. The gold nanoclusters provide additional fluorescent imaging capability, which can be easily accessed in common research labs. The conjugation of cancer cell & ndashtargeting antibodies allows for specific localization of nanoparticles. Indeed, these are the two central themes of this dissertation. To prepare multifunctional nanoparticles, high & ndashquality iron oxide nanoparticles were first synthesized in organic solvents following a modified & ldquoheat & ndashup & rdquo method. During synthesis, a modification was made by introducing a co & ndashsurfactant (trioctylphosphine oxide & ndashTOPO). TOPO facilitated the subsequent ligand exchange process because it weakly bound to nanoparticle surfaces and prevented the formation of densely & ndashpacked surfactant coatings. As a result, hydrophilic molecules (such as polyacrylic acid, polyethylenimine, glutathione and dopamine) were capable of replacing the original ligands, yielding water & ndashsoluble iron oxide nanoparticles. In addition to water solubility, dopamine coatings offered nanoparticles additional conjugation capability upon surface oxidization. These surface & ndashoxidized nanoparticles can directly conjugate with amine and/or thiol group & ndashcontained molecules through Michael addition and/or Schiff base formation. Using this conjugation strategy, dual & ndashimaging nanoparticles were prepared by integrating dopamine & ndashcoated nanoparticles with protein (such as bovine serum albumin, trypsin and lysozyme) & ndash encapsulated fluorescent gold nanoclusters. All integrated nanoparticles maintained their functionalities and structural integrity in biological environments. Furthermore, effects of protein characteristics on the photo & ndashchemical properties of gold nanoclusters and integrated nanoparticles were systematically examined. Similarly, cancer cell & ndashtargeting molecules (e.g. anti & ndashGD2 monoclonal antibodies) were conjugated onto dopamine & ndashcoated nanoparticles. After conjugation, antibodies retained their high targeting specificity, suggested by our cellular targeting studies. More promisingly, conjugated nanoparticles were capable of transporting into cytosols, which opens up new possibilities in cancer & ndashcuring drug loading and targeted therapy. Besides these already & ndashpublished works, we have studied the biological responses of human monocytes to different surface & ndashcharged iron oxide nanoparticles (manuscript in preparation). For dual & ndashimaging nanoparticles, their stability and bio & ndashimaging potential will be evaluated in phorbol myristate acetate (PMA) & ndashtreated monocytes using differential interference contrast (DIC) and confocal microscopy.

Book excerpt: Uniformly sized superparamagnetic iron oxide nanoparticles (SPIONs) with inorganic diameters of 3-35 nm were synthesized. New surface ligand coatings were designed and synthesized, and the resulting hydrophilic SPIONs in biological buffers were found to be compact, stable, highly magnetic, and biocompatible. Furthermore, the hydrophilic SPIONs were stable in vitro in serums and cells as well as in vivo in mice. Functionalized SPIONs demonstrated the ability of specific labeling. Finally, the hydrophilic SPIONs have potential as a non-toxic alternative to Gadolinium based contrast agents for T1-weighted magnetic resonance imaging (MRI) and they have shown potential in multicolor MRI as well as magnetic particle imaging.

Book excerpt: Recent development of a variety of superparamagnetic and ferromagnetic iron/iron oxide (Fe/Fe3O4) nanoparticles with different surface chemistry have been widely studied for numerous biological applications such as drug delivery, as diagnostics, hyperthermia and magnetic resonance imaging. The wide applications of Fe/Fe3O4 nanoparticles are possible since they exhibit favorable properties as high magnetization ability, are smaller than 100 nm in size, they can be coated with several ligands which allow drug delivery at a specific site and are biocompatible. By using Fe/Fe3O4 nanoparticles as drug delivery agents treatment costs and side effects can be reduced, however treatment efficacy can be increased. We have demonstrated that Fe/Fe3O4 nanoparticles can be utilized in different methods depending on their properties, to be used as therapeutic agents for cancer treatment. In one method we have taken advantage of the Fe/Fe3O4 nanoparticles magnetic ability to produce hyperthermia (heat) in cancer cells when subjected to an alternative magnetic field. Here we use the cell based delivery system since the size of the nanoparticles are small they can be taken up by monocyte/ macrophage like cells for systemic transportation to the inflamed cancer cite. The hyperthermia study was conducted in mice with pancreatic cancer. This study demonstrated that the life expectancy of the mice increased by 31%. In the next method we took the advantage of the surface chemistry of the Fe/Fe3O4 nanoparticles and changed it with dopamine-peptide and dopamine-thiosemicarbazone ligands. The advantage of the peptide is to deliver the nanoparticle to its target site and the thiosemicarbazone analogue is used as an iron chelator that would initiate apoptosis in cancer cells. This nanoplatform was tested in 4T1 breast cancer cell line and normal fibroblast cell line and demonstrated that it was effective towards the cancer cell line than the normal cell line at a ratio of 5:1 of thiosemicarbazone analogue : dopamine on the nanoparticle. However further studies are needed to be done to clarify the effectiveness of this nanosystem.
