Book excerpt: Nanoparticles have garnered much attention in pharmaceutical and biomedical fields because their small size and high surface area facilitate drug absorption, improve access to cells and organs, and enhance optical imaging. However, delivery of nanoparticles to the body is not always feasible or effective. Here, nanoparticle assemblies (flocs or clusters) for pulmonary drug delivery and biomedical imaging in cells are shown to facilitate delivery, interactions with cells, and manipulation of optical properties of inorganic/organic nanocomposites. The formation of aggregates by physical techniques and their mechanisms are described in detail. For pulmonary delivery, particles with aerodynamic diameters between 1-5 [mu]m deposit efficiently in the deep lungs. However, crystalline, non-porous, poorly water soluble drugs of this size require long dissolution times, limiting absorption by the body. Therefore, drug dissolution must be "decoupled" from deposition to improve absorption. To address this challenge, drug nanoparticles were dispersed within 4-[mu]m water droplets when administered via nebulization or as micron-sized flocs using a pressurized metered dose inhaler (pMDI). Upon deposition in aqueous media, the aerosolized nanoparticle assemblies dissociated into constituent nanoparticles, raising the available surface area for dissolution and increasing dissolution rates, relative to solid particles. Poorly water soluble drug nanoparticles were prepared using a controlled precipitation (CP) or thin film freezing (TFF) process, in which stable nanoparticles (30-300 nm in diameter) with high potencies (>90 wt% drug) were produced by rapidly nucleating drug solutions in the presence of strongly adsorbing polymers or by freezing, respectively. Amorphous, nanoparticles prepared by CP produced stable aqueous dispersions with high fine particle fractions (FPF) of 77% and total emitted doses (TED) of 1.5 mg/min upon nebulization. CP and TFF also produced anisotropic particles (aspect ratios >5), which formed stable suspensions in a hydrofluoroalkane propellant. Inefficient packing of anisotropic particles formed loose, open flocs that stacked upon each other to prevent settling. Upon pMDI actuation, atomized propellant droplets shear apart and template portions of the floc to yield porous particles with high FPFs (49-64%) and TEDs (2.4 mg/actuation). The controlled assembly of gold nanoparticles into clusters is also of great interest for biomedical imaging and therapy because clusters exhibit improved near infrared absorbance (where blood and tissue are most transparent), relative to single spherical particles, and can biodegrade into clearable particles. Gold nanoparticles (5 nm) were assembled into clusters between 30 to 100 nm in diameter with high gold loadings, resulting in strong NIR absorbance. The assembly was kinetically controlled with weakly adsorbing polymers by manipulating electrostatic, van der Waals, steric, and depletion forces. Furthermore, clusters assembled with a biodegradable polymer deaggregated back into primary particles in physiological media and within cells. This kinetic assembly platform is applicable to a wide variety of fields that require high metal loadings and small particle sizes.

Although they can be synthesised by the reduction of bulk materials down to the nanoscale, it is often more efficient and effective to build them from 'the bottom up' in solution, using atomic or molecular building blocks to assemble the desired nanostructure in a precisely controlled chemical environment. In this thesis, the assembly of organic and inorganic nanoparticles by such a method is discussed. -- Conjugated polymers were used in an attempt to create nanoparticles which exhibit the useful optical properties of quantum dots, but do not contain any toxic materials. These polymers are organic semiconductors which exhibit excellent optical characteristics such as bright fluorescence and high quantum yields. Importantly, they are generally biocompatible. Two methods were employed to create aqueous nanoparticle dispersions of conjugated polymers: miniemulsion and micellar encapsulation. These methods, and different refinements of each method, produced nanoparticles ranging from ca. 1000 nm to below 10 nm. The nanoparticles were capped with poly(ethylene glycol), an excellent surfactant for biological applications, and then functionalised with carboxylic acid, thiol and amine groups. Preliminary studies of these nanoparticles in cellular imaging showed that they are taken up by various cell types, with no evidence of toxicity. Superparamagnetic iron oxide nanoparticles were then incorporated in the solid conjugated polymer nanoparticles in order to create magnetic-fluorescent composites. -- Inorganic nanoparticles were synthesised by a novel metal-organic route. Here, mercury acetate was used as a mercury precursor for the formation of mercury selenide quantum dots in trioctylphosphine oxide. The initial synthesis created a dispersion of spherical nanoparticles.

Nanomedicine utilizes nanoparticles such as liposomes, polymeric nanoparticles, carbon nanotubes, nanowires, quantum dots and inorganic nanoparticles to diagnose and treat various diseases. This dissertation research has focused on the development of inorganic nanoparticles for diagnostic and therapeutic applications. Targeted nanoparticulate magnetic resonance imaging (MRI) contrast agents discussed in chapter 3 showed high MRI signal and will be invaluable for future tissue specific imaging and investigation of molecular and cellular events. Specifically, this study highlights that simple one-step method for preparing extremely stable and biocompatible NPs of the gadolinium ferrocyanide coordination polymer. These NPs exhibit no cytotoxicity. Furthermore, we have demonstrated that such NPs possess extremely high T1-weighted relaxivity, suggesting the potential of this coordination-polymer structural platform in the development of new-generation T1-weighted cellular MR probes for biological receptors or markers within the cell to study molecular events as well as for in vivo MR imaging in biomedical research and clinical applications. In chapter 4 synthesis and characterization of nanoparticulate contrast agent for X-ray computed tomography is reported. These nanoparticles offer a much higher contrast efficacy compared to clinical iodinated agents at 120 kVp. Together with long circulation time and low toxicity, these nanoparticles can act as a high-performance CT contrast agent for in vivo applications. The remaining chapters of this dissertation concern the synthesis of therapeutic nanoparticles. The first part of the chapter 5 describes the synthesis and characterization of Au@ZnMoS4 nanoparticles as copper depleting agent for the treatment of Wilson's disease. Their utility as copper depleting agent has been clearly demonstrated in vitro. In the second part of Chapter 5, the synthesis and characterization of therapeutic nanoparticles for angiogenesis inhibition is reported. In this dissertation project, anti-angiogenic function and mechanism of ZnMoS4 NPs in primary HUVECs was investigated in detail.

Described herein is work on biological and synthetic nanostructures as potential building blocks for nanostructured materials by design.In the first part of this thesis we present our investigation into the functional properties and chemical control of two bio-organic materials: tobacco mosaic virus coat protein and the reversibly photoswitchable protein Dronpa. Attachment of a hexahistidine tag to tobacco mosaic virus coat protein by genetic engineering resulted in unprecedented assembly behavior for the coat protein, forming nanostructured rafts, nanofibers, and hexagonally packed disks in addition to structures normally observed for the wild-type coat protein. The type of assembly formed was controlled by the addition of ethanol or nickel(II)-nitrilotriacetic acid, enabling the modified protein to serve as a reliable template for the future preparation of hybrid nanostructured materials. Scanning tunnelling microscopy and spectroscopy obtained of Dronpa monolayers on gold allowed us to define the relationships between Dronpa fluorescence switching, orientation, and conductance. Modification of Dronpa with a hexahistidine tag resulted in better self-assembled monolayers of Dronpa, and also demonstrated the influence of molecular orientation on protein conductivity. Recent applications of magnetic iron oxide nanoparticles for separation, catalysis, and magnetic imaging have stimulated interest in the synthesis and modification of ultrasmall water-soluble iron oxide nanoparticles (the topic of Chapters 4 and 5) as components for the preparation of functional materials. In response, an efficient ligand exchange method for the synthesis of aqueous stable magnetic iron oxide nanoparticles was developed to yield monodisperse nanoparticles functionalized with the small, highly charged ligands dopamine and Tiron. Motivated by the need to better understand the nature of ligand binding to iron oxide, model benzoic acid derivatives and catechol-based ligands were reacted with oleic acid stabilized iron oxide nanoparticles. Fourier transform infrared spectroscopy was used to examine the ligand exchange, purification, and the resulting complexes. The correlations observed between features in the obtained spectra and solubility of the ligand-stabilized nanoparticles enabled the assignment of binding modes for the series of model ligands--which will assist in the rational design of custom ligands for iron oxide. Advances in monodispersity, chemical control, reaction monitoring, and knowledge gained regarding the structure-functional relationships of the bio-organic and inorganic materials examined in this work will assist in the design of future hybrid inorganic-organic nanostructured materials. " --
