Available in PDF, EPUB and Kindle. Book excerpt: Biomedical applications of gold complexes based on N-heterocyclic carbenes (NHCs) are beginning to unfold. Some cationic gold(I) NHCs complexes show antimitochondrial activities, a very promising action mode in the fight against cancer; due to their positive charge these complexes target preferentially tumor cells, leading to cell death. In this work of thesis, three groups of new NHC-based gold and heterobimetallic complexes involving aliphatic or aromatic amino-functionalized NHCs with interesting potential in biomedical research have been synthesized and characterized. The first group is formed by three luminescent heterobimetallic gold(I)-ruthenium(II) complexes containing heteroditopic bipyridine-NHC ligands. These compounds have been biologically investigated by in vitro tests for their antitumoral, antileishmanial and antimalarial activities. Finally, imaging studies in cancer cells have been performed exploiting the luminescent properties of the most active compound. The second group of molecules is concerned by cationic gold(I) complexes containing two 1-[2-(diethylamino)ethyl]imidazolydene ligands. First the complexes have been tested for their antiproliferative activity in prostate cancer cell line PC-3. Lipophilicity (Log P) has been determined for these complexes. The most active complex has been tested for the cytotoxic activities in five human cancer cell lines and primary endothelial cells demonstrating a potent action and selectivity for cancer cells. In addition, antileishmanial and antimalarial activities of these compounds have been investigated showing interesting results. The third group is formed by a hetero-dinuclear gold(I)-silver(I) and a trinuclear gold(I)-copper(II) complexes containing phenanthroline-NHC ligands. The compounds are formed by two different organometallic units potentially able to act as multi-targeting anticancer drug.

The author demonstrates significant advances made in anti-cancer gold(III), gold(I) and platinum(II) complexes. Detailed chemical synthesis, in vitro and/or in vivo anti-cancer activities are clearly presented including: (i) a class of Au(III) complexes containing a highly fluorescent N^N^N ligand and NHC ligand that simultaneously act as fluorescent thiol “switch-on” probes and anti-cancer agents; (ii) a dinuclear gold(I) complex with a mixed diphosphine and bis(NHC) ligand displaying favorable stability and showing significant inhibition of tumor growth in two independent mice models with no observable side effects; and (iii) a panel of stable luminescent cyclometalated platinum(II) complexes exhibiting high specificity to localize to the endoplasmic reticulum (ER) domain, inducing ER stress and cell apoptosis. These works highlight the clinical potential that gold and platinum complexes offer for cancer treatment.

New silver(I) and gold(I) halide mono-NHC complexes are synthesized, structurally characterized, and compared to other existing silver(I) and gold(I) complexes. The gold(I) cation firmly bound to the carbene, undergoes oxidative addition with halogens to afford gold(III) NHC complexes, or dehalogenation in presence of inorganic silver salts to afford cationic gold(I) mono-NHC complexes in coordinating solvent. The coordinating solvent can be replaced by a neutral or anionic group to yield gold(I) NHC complexes with unusual ligands such as olefins, pyridines, sugars. In contrast, the silver(I) cation is weakly bound to the carbene, and silver(I) NHCs decompose in presence of halogens or inorganic silver salts.

Abstract: Abstract of thesis entitled "Anti-Cancer N-Heterocyclic Carbene Complexes of Gold(III), Gold(I) and Platinum(II): Thiol "Switch-on" Fluorescent Probes, Thioredoxin Reductase Inhibitors and Endoplasmic Reticulum Targeting Agents" Submitted by ZOU TAOTAO for the degree of Doctor of Philosophy at The University of Hong Kong in Feb 2015 Despite the success of cisplatin in combating various cancers, the problems of resistance and side effects have limited its wide application in cancer treatment. Gold complexes have recently emerged as promising candidates for cancer treatment. Unlike cisplatin, gold complexes do not favor DNA binding, but instead preferentially bind SH/SeH-containing proteins/enzymes in cancer cells, giving hope that these compounds may overcome cisplatin's resistance problems. Since the side effects of cisplatin are, at least partially, caused by instability-associated, off-target binding interactions, the incorporation of N-heterocyclic carbene (NHC) ligands may be a useful strategy to improve the stability of metal-based chemotherapeutics due to the strong  donating ability of NHC to form strong M-C(NHC) bonds. Meanwhile, the ease of modification and steric effects of NHC ligands provide favorable lipophilicity, reactivity and binding specificity of metal complexes. Indeed, some Pt(II)-NHC complexes have been reported to specifically target proteins/organelles rather than DNA. 3+ + The reduction of 4-coordinated Au to 2-coordinated Au by thiols has been 3+ demonstrated. Meanwhile, owing to the low energy d 2 2 orbital of Au, most gold(III) x -y complexes are non-emissive in solutions. In Chapter 3, by taking advantage of these unique properties, the synthesis of a class of non-emissive Au(III) complexes containing highly fluorescent N DEGREESN DEGREESN ligands and NHC ligands is described. In these compounds, the NHC ligand prevented further reduction of Au(III) to Au(0) and increased the III + solubility of the Au(III) complexes. The [Au (N DEGREESN DEGREESN)(NHC)] complex was readily reduced by GSH to form [Au (NHC)(GS)], accompanied by the release of the fluorescent H N DEGREESN DEGREESN ligand, therefore acting as a fluorescent "switch-on" probe for thiols. In addition, the anti-cancer activity of Au(I) is owed to its ability to inhibit thiol-enzymes; thus, the Au(III) complexes could also serve as precursors/prodrugs, activated by metal reduction, to deliver anti-cancer active Au(I) species. The major problem of most Au(I) anti-cancer agents is the lack of in vivo anti-tumor activity due to their high thiol-reactivity which leads to many off-target binding interactions with blood thiols. In Chapter 4, the synthesis of a dinuclear gold(I) complex bearing a mixed diphosphine and bis(NHC) ligand is described. This dinuclear Au(I) complex displayed a favorable stability that conveys potent thioredoxin reductase inhibition without being attacked by blood thiols. This complex significantly inhibited tumor growth in two independent mouse models with no observable side effects. The dinuclear Au(I) complex also exhibited anti-angiogenesis activity in a tumor model and inhibited sphere formation of cancer stem cells in vitro. A number of synthetic approaches towards both the ligands and the prepared metal complexes have been developed, with a concerted effort on achieving the desired Au(I) or Ag(I) complexes with minimal reaction steps and synthetic style. The thesis body is divided into two main sections. The first section addresses the preparation of suitable ligand precursors of potential Au(I) and Ag(I) complexes in the form of halo- and phosphino-functionalised imidazolium salts. Several series of haloalkylimidazolium salts were prepared that encompass a range of halogens (Cl, Br, I), alkyl substituents (Me, i-Pr, t-Bu, n-Bu), differing alkyl linker length (n = 0-3), and a variety of organic spacers employed to bridge multi-imidazolium moieties. Novel bidentate and multidentate phosphinoalkylimidazolium salts were synthesised from the various haloalkylimidazolium salts, via the substitution of a halide with nucleophilic diphenylphosphide. A new approach towards rare methylene bridged phosphinomethylimidazolium salts was achieved from the reactions of halomethylimidazolium salts with diphenylphosphine. The second section investigates the preparation of Au(I) and Ag(I) complexes from the halo- and phosphino-functionalised imidazolium salts. A series of dicationic 10, 12, and 14-membered metallacyclic Ag(I) complexes were prepared from the bidentate phosphinoalkylimidazolium salts. The dinuclear Ag(I) metallacycles combine two phosphino-functionalised NHC ligands that are bridged by two coordinated Ag(I) ions in an exclusively head-to-head arrangement. A dinuclear Ag(I) metallacycle was investigated for transmetallation potential to a Au(I) complex and found to selectively transmetallate at the Ag(I) coordinated to the NHC ligands to form a bimetallic metallacycle. Unexpected phosphine oxidation of a 10-membered dinuclear Ag(I) metallacycle resulted in complex disproportionation to an isolable and rare silver(I) trimer. Metal-NHC complexes from haloalkylimidazolium salts have not been reported previously, a novel approach to the synthesis of a series of Au(I) complexes from haloalkylimidazolium salts and a respective gold source was developed and is reported herein. Different synthetic approaches towards Au(I) complexes with the phosphinoalkylimidazolium salts explored a variety of ways to generate the NHC from an imidazolium in the presence of the phosphine. A one-pot, high yielding synthesis of a dinuclear Au(I) complex from PPh3 was also devised, with controlled assembly of the complex resulting in a similar head-to-head ligand arrangement to the dinuclear Ag(I) metallacycles. As an aside, a family of mononuclear [Au(R2NHC)2]+ complexes (R = Me, i-Pr, t- Bu, n-Bu, Cy) prepared previously in our research group, was expanded because of the promising antimitochondrial activity shown by [Au(i-Pr2NHC)2]+. Two new [Au(R2NHC)2]+ complexes with simple alkyl chain functionality were prepared with fine-tuned lipophilicity in close proximity to that of [Au(i-Pr2NHC)2]+.

Abstract: A library of luminescent cyclometalated gold(III) complexes containing various bis-cyclometalating R-C DEGREESN DEGREESC ligands derived from 2,6-diphenylpyridine and ancillary alkynyl ligands, [Au(R-C DEGREESN DEGREESC)(C≡CR')], has been synthesized. Some of the complexes have been structurally determined by X-ray crystallography. These complexes were found to exhibit intense emission in dichloromethane solution at 298 K, originating from metal-perturbed triplet intraligand (IL) excited states of the R-C DEGREESN DEGREESC ligand, with substantial charge transfer character from the aryl moiety to the pyridyl ring. In the presence of electron-rich alkynyl ligands, the emission origin could be switched to an alkynyl-to-cyclometalating ligand ligand-to-ligand charge transfer (LLCT) excited state. This class of complexes was also demonstrated to show rich electroluminescence properties as emitters in organic light-emitting devices. In addition, the supramolecular assembly of this class of complexes has also been revealed in gelation studies. N-Heterocyclic carbenes have been incorporated into the gold(III) metal center to prepare a series of luminescent mononuclear and dinuclear gold(III) complexes, [{Au(C DEGREESN DEGREESC)}n(NHC)](PF6)n and [{Au(tBuC DEGREESN DEGREESCtBu)}n(NHC)]- (PF6)n (n = 1, 2). The X-ray crystal structures of most of the complexes have been determined. The emissions of these complexes were assigned to originate from the metal-perturbed intraligand excited state of the bis-cyclometalating ligand. One of the C2-bridged dinuclear complexes was found to exhibit two distinct reduction couples, tentatively correlated to the presence of significant intramolecular π-π interaction in the complex. Two novel series of luminescent mononuclear alkynylgold(III) complexes, [Au(C DEGREESN)(C≡CR)2] (HC DEGREESN = 2-phenylpyridine (Hppy) and derivatives) and [Au(C DEGREESN DEGREESN)(C≡CR)]PF6 (HC DEGREESN DEGREESN = 6-phenyl-2,2'-bipyridine and derivatives), have been synthesized. Some of the X-ray crystal structures have been determined. The former class of complexes with bidentate C DEGREESN ligands has been observed to show tunable emission spanning across the visible spectrum from 462 to 697 nm. With the exception of [Au(ppy)(C≡C-C6H4-NH2-p)2] which showed a low-energy band originated from a 3LLCT [π(C≡C-C6H4-NH2)→π*(C DEGREESN)] excited state, all complexes exhibited vibronic-structured emission bands originated from the intraligand transition of the cyclometalating C DEGREESN ligand in dichloromethane solution at 298 K. On the other hand, most of the complexes with the tridentate C DEGREESN DEGREESN-type ligand have been observed to exhibit vibronic-structured emission bands attributed to the intraligand transition of the C DEGREESN DEGREESN ligand in low-temperature butyronitrile glass.
