Book excerpt: New water soluble, attainable to ATRP polymerization Cl-terminated poly(glycidol) macroinitiators were prepared by modification of (Omega)-hydroxyl group of poly(glycidol acetal) using 2-chloropropionyl chloride fallowed by selective acidic deprotection of acetal groups. The obtained macroinitiators of different molar masses were successfully employed in ATRP of NIPAM and 4VP to give well-defined stimuli sensitive block copolymers of targeted molar ratio of blocks. The results obtained from light scattering methods showed formation of stable aggregates upon stimuli (pH or temperature) by all the obtained polymers. Additionally, photocrosslinkable block copolymers of glycidol and NIPAM having incorporated moieties of chromophore (2-(dimethyl maleinimido)-N-ethyl-acryl amide) were prepared using macroinitiator technique and used to synthesis of temperature sensitive microgels. Conjunction points have been successfully formed by UV irradiation of polymer water solution above cloud point. The influence of such parameters like block ratio, block length, amount of chromophore, concentration, irradiation time, temperature and heating rate on the properties of obtained microgels was investigated. The obtained core-shell structures were stable under critical conditions and showed continuous volume phase separation process upon increase of temperature, fully reversible and reproducible (no hysteresis effect). Thus, the proposed method not only gave the opportunity to control size or swelling degree of microgels, but also diminished gradient in crosslinking density (random chromophore distribution in polymer backbone), improved colloid stability (poly(glycidol) shell) and completely eliminated additives (surfactants, initiators, stabilizers).

Book excerpt: Water-soluble stimuli responsive block copolymers are a rapidly emerging class of materials with great potential in biomedical and technological applications. In this work a novel class of pentablock copolymers are synthesized via atom transfer radical polymerization techniques and their stimuli-responsive self-assembly properties are characterized. Aqueous solutions of these materials are observed to form micelles and hydrogels in response to changes in both temperature and pH. Cryogenic transmission electron microscopy (cryo-TEM) and small angle neutron and X-ray scattering (SANS and SAXS) techniques are used to investigate the nanoscale structures formed by these pentablock copolymers in solution. The gel structure and mechanical properties are investigated with SANS and rheological techniques. The multi-responsive properties of these materials are utilized to formulate a stimuli responsive drug delivery formulation that exhibits thermoreversible gelation and pH dependent release rate of model drugs.

The objective of this book is to fill this gap by presenting a snapshot of the latest advances in the synthesis, characterization and applications of microgels. This book consists of three sections. The second section is devoted to exploiting microgel properties and engineering techniques. This diversity promises to deliver the next generation of advanced functional materials for a wide range of specific applications. In particular, the past decade has seen a growing interest in the development of nanoscale materials for sophisticated technologies. Aqueous colloidal microgels have emerged as a promising class of soft materials for multiple biotechnology applications. The amalgamation of physical, chemical and mechanical properties of microgels with optical properties of nanostructures in hybrid composite particles further enhances the capabilities of these materials. This work covers the general areas of responsive polymer microgels and their composites, and encompasses methods of fabricating microgel-based drug delivery systems for controlled and targeted therapeutic applications.The first part of this thesis is devoted to acquainting the reader with the fundamental aspects of the synthesis, functionalization and characteristic properties of stimulus-responsive microgels constructed from poly(N-isopropylacrylamide) (poly(NIPAm)) and other functional comonomers. In particular, the role of electrostatics on the swelling-deswelling transitions of polyampholyte microgels upon exposure to a range of environmental stimuli including pH, temperature, and salt concentration are discussed. The templated synthesis of bimetallic gold and silver nanoparticles in zwitterionic microgels is also described.The latter part of this thesis focuses on the rational development of microgel-based drug delivery systems for controlled and targeted drug release. Specifically, the development of a biofunctionalized, pH-responsive drug delivery system (DDS) is illustrated, and shown to effectively suppress cancer cells when loaded with an anticancer agent. In another chapter, the design of tailored hybrid particles that combine the thermal response of microgels with the light-sensitive properties of gold nanorods to create a DDS for photothermally-induced drug release is discussed. The photothermally-triggered volume transitions of hybrid microgels under physiological conditions are reported, and their suitability for the said application evaluated. In another component of this work, it is explicitly shown that electrostatic interactions were not needed to deposit gold nanorods on poly(NIPAm)-derived particles, thereby eliminating the need for incorporation of charged functional groups in the microgels that are otherwise responsible for large, undesirable shifts and broadening of the phase transition.
