Book excerpt: A challenging task, in the area of magnetic resonance imaging is to develop contrast enhancers with built-in antioxidant properties. Oxidative stress is considered to be involved in the onset and progression of several serious conditions such as Alzheimer’s and Parkinson’s disease, and the possibility to use cerium-contained nanoparticles to modulate such inflammatory response has gained a lot of interest lately. The rare earth element gadolinium is, due to its seven unpaired f-electrons and high symmetry of the electronic state, a powerful element for contrast enhancement in magnetic resonance imaging. Chelates based on gadolinium are the most commonly used contrast agents worldwide. When introducing external contrast agents there is always a risk that it may trigger inflammatory responses, why there is an urgent need for new, tailor-made contrast agents. Small sized cerium oxide nanoparticles have electronic structures that allows coexistence of oxidation states 3+ and 4+ of cerium, which correlates to applicable redox reactions in biomedicine. Such cerium oxide nanoparticles have recently shown to exhibit antioxidant properties both in vitro and in vivo, via the mechanisms involving enzyme mimicking activity. This PhD project is a comprehensive investigation of cerium oxide nanoparticles as scaffold materials for gadolinium integration. Gadolinium is well adopted into the crystal structure of cerium oxide, enabling the combination of diagnostic and therapeutic properties into a single nanoparticle. The main focus of this thesis project is to design cerium oxide nanoparticles with gadolinium integration. A stepwise approach was employed as follows: 1) synthesis with controlled integration of gadolinium, 2) material characterization by means of composition crystal structure, size, and size distribution and 3) surface modification for stabilization. The obtained nanoparticles exhibit remarkable antioxidant capability in vitro and in vivo. They deliver strongly enhanced contrast per gadolinium in magnetic resonance imaging, compared to commercially available contrast agents. A soft shell of dextran is introduced to encapsulate the cerium oxide nanoparticles with integrated gadolinium, which protects and stabilizes the hard core and to increases their biocompatibility. The dextran-coating is clearly shown to reduce formation of a protein corona and it improves the dispersibility of the nanoparticles in cell media. Functionalization strategies are currently being studied to endow these nanoparticles with specific tags for targeting purposes. This will enable guidance of the nanoparticles to a specific tissue, for high local magnetic resonance contrast complemented with properties for on-site reduced inflammation. In conclusion, our cerium oxide nanoparticles with integrated gadolinium, exhibit combined therapeutic and diagnostic, i.e. theragnostic capabilities. This type of nanomaterial is highly promising for applications in the field of biomedical imaging.

Book excerpt: Integrating multiple functionalities into individual nanoscale materials is of tremendous importance in biomedicine, since this would expand the capabilities of nanoscale materials to perform multiple tasks. Here, fluorescence optical imaging and magnetic resonance imaging (MRI) are combined within the same nanoparticle based material. NaYF4: Tm3+, Yb3+, showed NIR to NIR upconversion, giving deep tissue penetration, which is an advantage in medical imaging. Their lanthanide ions (Tm3+/Yb3+) derived optical properties in the near infrared region of the spectrum, where tissue is maximally transparent. Multimodal properties may be imparted to NaYF4: Tm3+, Yb3+ via the functionalization of gadonium-tetraazacyclododecanetetraacetic acid (Gd-DOTA), a commercially available MRI contrast agent. Gd-DOTA was covalently bonded to the surface of the silica coated UCNPs using the click reaction. This imaging probe may be used for both MRI and fluorescent imaging. Another approach to achieve bimodalities is by doping Gd3+ ions into the systems of UCNPs, which provides both magnetic and optical properties. Ultra-small NaGdF4, Tm3+, Yb3+ of 5 nm was chosen since it is an excellent T1 contrast agent. The surface chemistry of nanoparticles has direct impact on its physiochemical properties. Therefore, it is aimed to design nanoparticle based blood pool contrast agent for disease diagnostics, which involved the functionalization the polyethylene glycol (PEG) polymer on the ultra-small UCNPs. Cisplatin and follow-up drugs carboplatin (ParaplatinTM) and oxaliplatin (EloxatinTM) have been the gold standard for metallodrugs in clinical settings as antineoplastic agents. While effective, these drugs (either alone or in combination therapy) have faced a number of clinical challenges resulting from their limited spectrum of activity, high toxicity leading to significant side effects, resistance, poor water solubility, low bioavailability and short circulating time. In the past 10 years, various unconventional non-platinum metal-based agents have emerged as a potential alternative for cancer treatment. These compounds are highly effective and selective in cancers resistant to cisplatin and other chemotherapeutic agents. Research in this area has recently exploded with a relevant number of patents and clinical trials, in addition to reports in scientific journals. Furthermore, in parallel to the synthesis of coordination and organometallic compounds comprising many different metals and unconventional platinum-based derivatives, researchers are focused on optimizing mechanistic and pharmacological features of promising drug candidates. This Special Issue aims to highlight the latest advances in anticancer metallodrugs with a focus on unconventional anticancer agents, as well as novel activation, targeting and delivery strategies aimed at improving their pharmacological profile.

Book excerpt: This thesis contains two distinct part: the first two chapters focus on research efforts towards the use of Dextran Sulfate-Coated Iron Oxide (SDIO) Nanoparticles as an MRI contrast agent for the diagnosis of brain inflammation, while the third chapter is a personal reflection on experiences from assisting with the development and initial implementation of an online, UC-wide Introduction to Research course. In Chapter 1, the synthesis and characterization of SDIO nanoparticles is described in detail. The synthetic work in this chapter showed great reproducibility with previous syntheses in our lab. New characterization efforts helped illuminate the physical and magnetic degradation process of SDIO nanoparticles in water and acidic solutions, intended to help guide future work into how the particles break down in vivo. Chapter 2 covers a series of cell studies that are intended as support for the potential future use of SDIO nanoparticles as an MRI contrast agent for diagnosing brain inflammation through receptor-specific uptake in activated microglia. While prior work in our lab has already shown promising results in using SDIO for this purpose in mice, several underlying questions and concerns needed to be addressed before continuing further with animal models. Firstly, possible off-target uptake by astrocytes was examined, as this could potentially cause background signal and false positives. Secondly, additional studies into the biocompatibility of SDIO nanoparticles with cortical cells were undertaken, to further assess whether the particles are a safe diagnostic tool. Chapter 3 details my personal involvement with the development and implementation of an online Introduction to Research course, under the guidance of Dr. Angelique Louie. My experiences with this course have proven incredibly valuable for personal development as I transition into my future career as a high school science teacher.
