Abstract: Abstract of thesis entitled "SYNTHESES AND PHOTOPHYSICS OF LUMINESCENT MONO- AND DINUCLEAR RHENIUM COMPLEXES CONTAINING FUNCTIONAL LIGANDS" Submitted by Wong Man Chung for the degree of Doctor of Philosophy at The University of Hong Kong ABSTRACT With the growing interest in the exploration of inorganic/ organometallic sensing materials, a series of mononuclear rhenium(I) and rhenium(V) and dinuclear rhenium(I) (DIC-18-C-6)Cl], complexes with crown ether pendants of formula [Re(CO) [{Re(CO) Cl}(DDIC)], [Re(CO)(DIC-S)Cl], [Re(CO)(DIC-3S)Cl], [Re(CO) (DIC- 3 2 3 3 3 Se)Cl], [Re(CO) (DPPZC)Cl] and [Re (PPh ) (C≡C-benzo-15-crown-5) ] have been 3 3 2 2 synthesized. The photophysical, electrochemical and cation-binding properties of these systems have been studied. The structure of [Re(CO) (DIC-Se)Cl] has been established by X-ray crystallography. The electronic absorption energies of the rhenium(I) complexes were found to be sensitive to the presence of metal cations. The stability constants of [Re(CO)(DIC-18-C-6)Cl] and [{Re(CO) Cl}(DDIC)] for alkali and 3 3 2 alkaline-earth metal cations have been determined by UV-visible spectrophotometry. The complexes [Re(CO)(DIC-S)Cl], [Re(CO)(DIC-3S)Cl] and [Re(CO) (DIC-Se)Cl] 3 3 3 showed a downfield shift of methylene protons adjacent to the sulphur or selenium atom on the crown ether moiety upon addition of silver ion in deuterated acetonitrile. Their stability constants for silver ion have also been determined by UV-visible and H NMR spectroscopies. Direct evidences for the formation of the inclusion species between the crown ether-containing rhenium(V) complex [Re (PPh ) (C≡C-benzo-15-crown-5) ] and 3 2 2 lithium or sodium ions were obtained from electrospray ionization-mass spectrometry. A series of mononuclear and dinuclear rhenium(I) complexes with thiolate and selenolate ligands, [Re(CO) (N-N)(S-C H -X-p)] and [{Re(CO) (N-N)} (μ-S-C H -X- 3 6 4 3 2 6 4] t p)], have been synthesized (N-N = bpy, Me -bpy, Bu -bpy, phen; X = CH, OCH, 2 2 3 3 C(CH ), Cl, F). Unsymmetrical dinuclear rhenium(I) thiolate complexes have been 3 3 prepared by utilizing the mononuclear complexes as precursors. Their electrochemical properties have also been studied. The photophysical behaviour of the dinuclear complexes has been studied. Attempts to investigate the photophysical behaviour of the mononuclear counterparts were unsuccessful owing to their low photo-stability. The crystal structures of the complexes [Re(CO) (phen)(SC H -OCH -p)], [Re(CO) ( Bu - 3 6 4 3 3 2 bpy)(SePh)], [{Re(CO) (bpy)} (μ-SC H -CH -p)]OTf, [Re(CO) (phen)(μ-SC H -OMe- 3 2 6 4 3 3 6 4 p)Re(CO) (bpy)]OTf and [Re(CO) (Me -bpy)(μ-SC H -Me-p)Re(CO) (NO -phen)]OTf 3 3 2 6 4 3 2 have also been determined. Besides, a series of luminescent rhenium(I) acetylide complexes with diimine ligands, [Re(CO) (N-N)(C≡C-R)] (N-N = bpy, Me -bpy, phen, NO -phen; R = Ph, C H - 3 2 2 6 4 Cl-p, C H -C H -p, C H -C≡CH-p, C H -N=N-C H -OC H, EEME), have been 6 4 8 17 6 4 6 4 6 4 12 25 synthesized and their photophysical and electrochemical properties studied. Book excerpt: Alzheimer’s disease (AD) is characterized by two main protein aggregate hallmarks in the brain: extracellular deposition of the amyloid-? (A?) in senile plaques and intracellular neurofibrillary tangles (NFTs) consisting of hyperphosphorylated tau protein. The past decade has seen great progress in the development of imaging probes for the non-invasive detection of A? and tau aggregates. Here positron emission tomography (PET), single-photon emission computed tomography (SPECT) and magnetic resonance imaging (MRI), are highly promising technologies for clinical diagnostics. However, as a research tool, optical imaging is superior because it is real-time, sensitive, inexpensive, not radioactive and that it in particular affords high-resolution studies both in vitro and in vivo. Fluorescent probes are especially useful for designing novel binding scaffolds for structure investigations of protein aggregates. This thesis describes design, synthesis and evaluation of a series of fluorescent probes for detection of amyloid fibrils, especially A? or tau aggregates in vitro. Firstly, trans-stilbenoid vinylbenzene-1,2-diol with benzene, naphthalene, anthracene, and pyrene are investigated with respect to their photophysical properties free in solution and when bound to amyloid fibrils, including time-resolved fluorescence measurements. It is noted that the extended conjugated systems retained the amyloid targeting properties of the probes and both the anthracene and pyrene moieties extensively enhanced the fluorescence intensity and prolonged lifetimes. Secondly, the synthesis of two molecules, Py1SA and Py2SA, based on pyrene linked to salicylic acid via a trans-stilbene C = C bond is presented. The compounds show strikingly different emission spectra when bound to preformed A?1-42 fibrils as well as to fibrils from four other distinct proteins. Additionally, excited state intramolecular proton transfer (ESIPT) coupled-charge transfer (ICT) is observed for the anionic form of the probes in polar solvents. This is likely the reason for the spectral differences of the probes when bound to amyloid fibrils. Moreover, the synthesis of a further development of the Congo red analogue X-34 [2,5-bis(4’-hydroxy-3’-carboxy-styryl) benzene] by rational design and synthesis is described. Full photophysical characterization was performed, including recording absorbance and fluorescence spectra, Stokes shift, quantum yield and fluorescence lifetimes. All ligands displayed high affinity towards recombinant amyloid fibrils of A?1-42 and tau as well as selectivity towards the corresponding disease-associated protein aggregates in human post mortem AD tissue. Lastly, the synthesis of a set of 2,1,3-benzothiadiazole (BTD)-based ligands with different conjugated spacers and variable patterns of OH substitutions of bis-styryl-BTD prototypes were developed. A? binding affinities (A?1-42 and A?1-40 fibrils) and the specificity towards A? plaques of all ligands were determined. These findings extend the structure to activity relationships of BTD-based ligands for A? fibril binding. Throughout the studies in this dissertation, new interesting properties of small molecule fluorescence probes have been discovered and analyzed. This knowledge should facilitate the development of noninvasive probes for early detection of Alzheimer's disease and to distinguish different A? fibril polymorphs.

Each chapter addresses a different topic in the emerging field of photocatalysis, which has become an indispensable tool for organic synthesis. Photocatalysis employs environmentally harmless and abundant visible light in the presence of a photosensitizer, and as such offers an attractive alternative to harmful UV light in photo-mediated reactions. This book introduces the novel concept of merging gold catalysis with visible light photocatalysis in a dual catalytic fashion, which demonstrates their compatibility with each other for first time and has inspired the development of various reactions. Moreover, a novel trifluoromethylation method, which combines radical addition chemistry with a polar rearrangement to synthesize valuable fluorinated compounds, is presented, since compounds featuring fluorinated functionality are the subject of increasing attention in pharmaceutical, agrochemical and material research. It also develops an external photocatalyst-free photochemical method for the synthesis of valuable indolizine heterocycles, where the product mediates its own formation. Lastly, it describes the synthesis and characterization of two novel highly porous metal-organic frameworks (MOFs). The comprehensive text is rounded out with illustrations and color figures.
