Merr.] plants can capture atmospheric nitrogen (N) through symbiotic relationships with soil bacteria in a process called "biological N fixation" (BNF), reducing the reliance on synthetic fertilizers. To meet growth of the global population and increasing demand for quality protein and oil, the need for soybean will inexorably grow. One of the challenges to further improve soybean seed yield is the high demand of N in comparison to cereals and other oilseed crops. Further characterization of the environmental and management effects on BNF process is needed to enhance its capacity for supplying N to the crop. This dissertation is organized in four chapters outlining these topics through a series of experiments describing the association between plant nutrition, BNF, and seed composition with seed yield generation. Chapter 1 presents a synthesis analysis on a nutrient-based physiological framework on four key variables determining N efficiency in soybean (i.e., seed yield, N seed concentration, total N uptake, and nitrogen harvest index). Results showed that N partitioning to the seeds is the main parameter responsible in seed yield variation instead of N seed concentration. An experiment in Chapter 2 explored the hypothesis of the carbon (C) cost of BNF on biomass production, yield formation, and seed composition. Chapters 3 and 4 explored the historical component behind seed yield generation and seed composition. Results from Chapter 3 describe the shift in allocation of reproductive biomass using the allometric approach, whereas Chapter 4 provides a detailed description of the dynamics of oil and fatty acid accumulation under two N regimes.

They investigate the fate of nitrogen fixed in soybean nodules by tracer experiment with 15N2 gas. The results indicate that major part of fixed N in bacteroids (a symbiotic state of rhizobia) is excreted rapidly to cytosol of infected cells in the form of ammonia, then the ammonia is assimilated into amino acids via GS/GOGAT pathway. Then the fixed nitrogen is assimilated into ureides, allantoin and allantoic acid, and then transported to the shoots via xylem. A small portion of fixed N was assimilated in the bacteroids directly into glutamate and alanine. On the other hand, nitrate absorbed from the roots are mainly assimilated into asparagine. The characteristics of nitrate absorption and metabolism was studied. It is well known that nitrate is a potent inhibitor to nodulation and nitrogen fixation, although the inhibitory mechanism is not fully understood. The authors recently found that nitrate depresses individual nodule growth and nitrogen fixation activity rapidly and reversibly when nodules were in direct contact with nitrate. The indirect effects of nitrate on nodule growth and nitrogen fixation activity were different among treatment concentration and period of supply. The continuous supply of low levels of nitrate from the lower part of roots promoted the nodulation and nitrogen fixation of the upper part of the roots. Hypernodulation mutant lines of soybean were isolated which have profuse nodulation compared with parents. They also exhibit partial-nitrate tolerant to nodulation. The characteristics of hypernodulation mutant lines were studied in relation to nitrate inhibition. The results suggest that lower nitrate absorption and assimilation activity in hypernodulation mutants may be one reason to milder inhibition by nitrate on hypernodulation mutant lines.

Book excerpt: Nitrogen (N) is present in proteins, enzymes, cell structures, purines and pyrimidines in DNA and RNA molecules, photosynthetic pigments, and several other types of molecules in all living organisms. Nonetheless, even though N makes up more than 78% of the atmosphere, it is reported to be the most frequent deficient nutrient in plants. Nitrate (NO3- ) and ammonium (NH4+) are the N forms absorbed by plants from soil, but legume crops can establish symbiotic relationships with rhizobia bacteria, and fix N2 from the atmosphere. In soybean, increasing yield and protein content are raising the crop's N requirement; therefore, enhanced N2 fixation is seen as a reliable path to avoid the use of N fertilizers. In this study, the objective was to perform a comprehensive screening in greenhouse and field conditions of soybean genotypes for traits related to N2 fixation. The purpose was to identify among the soybean genotypes different N2 fixation profiles at early and late stages, as well as to investigate their capacity to accumulate above-ground N and supply carry-over N to following crops. The results showed different profiles among the soybean genotypes for early and late N2 fixation capacity, both in greenhouse and field evaluations. Different traits were correlated to either early or late N2 fixation activity. Soybean and winter-rye shoot dry mass were evaluated in the field to assess above-ground N accumulation and carry-over N, respectively. Soybean genotypes were identified with specific capacities to accumulate N in above-ground biomass or supply N to winter-rye. The patterns of N2 fixation identified in this study, as well as the different abilities to accumulate N above-ground or supply N to following crops, could assist in the selection of superior lines with improved N2 fixation capacity.

Grain legumes such as soybean as requires host specific Brayrhizobium japonicum to enhance growth, nitrogen fixation, and grain yield. However, limited information exists on how commercial Bradyrhizobium inoculants affect symbiotic plant performance and yield of soybean, and as well as soil health in Ghana's cropping systems. A field study (2-yr) was conducted at CSIR-Savanna Agricultural Research Institute's experimental field at Nyankpala, Ghana to determine the impacts of Bradyrhizobium inoculants on; (1) growth, nodulation, nitrogen fixation, grain yield of soybean, and (2) soil biological and chemical properties. We also evaluated the commercial inoculants effects on the subsequent maize and soybean crops. The experiment was laid out as a split-plot design where the main plot consisted of tropical soybean (Glycine max crosses (TGX)) varieties; Jenguma (TGX1448-2E), Afayak (TGX1834-5E), and Songda (TGX 1445-3E). The subplot consisted of three commercial Bradyrhizobium japonicum inoculants with different strains, Biofix (USDA 110), NoduMax (USDA 110) and Legumefix (USDA 532c) plus an uninoculated control. Assessment was made on nodulation pattern, shoot biomass, nitrogen fixation, grain yield, and residual N balance. Bulk and rhizosphere soils were sampled and analyzed for soil pH, available soil N (NO3-N and NH4+-N) and P, and soil microbial community structure by phospholipids fatty acid (PLFA) analysis. Inoculants improved nodulation, shoot biomass, nitrogen fixation and grain yield of soybean. Greater responses were associated with NoduMax and Biofix. Inoculation increased grain yield by ~30 %. Commercial inoculants also increased microbial biomass, and available P and NH4+-N. Afayak outperformed the other soybean varieties for biomass dry matter, nodulation (nodule number) and grain yield. Afayak also stimulated greater microbial biomass and available P compared to Jenguma. Furthermore, enhance microbial biomass was found in the rhizosphere compared to the bulk soil due to soil enrichment with root exudate and commercial inoculants. In assessing, the previous year commercial inoculants effect on the subsequent soybean and maize crops, three (3) independent mineral N fertilizer rates (0, 50 and 100 kg N ha-1) were added to the soybean-maize rotation phase. Biofix yielded superior maize shoot dry matter and grain yield. Maize grain yield from previous commercial inoculants was equivalent to grain yield from 50 kg N ha-1mineral N fertilizer ). Thus inoculating soybean with commercial inoculants reduced mineral N nutrition for the subsequent maize crop by 50%. In the soybean-soybean phase, the previous Biofix and the uninoculated control produced significant soybean grain yield than the previous NoduMax. In conclusion, TGX soybean varieties exhibited superior performance when inoculated with commercial inoculants especially Biofix and NoduMax. However yearly inoculation of soybean is needed to sustain enhanced grain yield and soil quality in Northern Ghana.
