Book excerpt: Proteins are dynamic structures that are in constant fluctuations, and their ability to undergo structural changes is critical to their function. However, their fastest dynamics in thermal equilibrium have remained largely unexplored. In this work, studies that examine the dynamics of aqueous proteins using two-dimensional infrared echo spectroscopy (2D IR) are presented. In particular, investigations of fast fluctuations in proteins and peptides within the context of structural changes upon denaturation are discussed. 2D IR is a nonlinear optical spectroscopic technique that can measure ultrafast dynamics of complex molecules in the picoseconds regime, timescales ~6-10 orders of magnitude faster than nuclear magnetic resonance. In addition, the relatively low energy mid-IR laser pulses used in this study probe the relevant nuclear degrees of freedom without significantly perturbing the protein structure or dynamics. Brief descriptions of the experimental setup and methods, as well as analysis and interpretation, are given.

Book excerpt: This monograph is neither a historical outline of the development of the concepts of protein biosynthesis and the structure and functions of the ribosomes, nor an exhaustive survey of the literature on these questions. The monograph is based upon an analysis of the modern trends in this field. The purpose of the monograph was to formulate more or less generalized representations of the structure and" function of the ribosome, as we envision it at the present day. It may be that this attempt is premature for a number of reasons, and the concepts outlined here will very soon be revised. Nonetheless, despite this risk, we believe it to be advisable to undertake this attempt for the following reasons: firstly, the undertaken analysis could aid in the comprehension of the substantial mass of extremely scattered experimental data on the ribosomes presently available; secondly, in any event, even if most of the concepts outlined rapidly become obselete, they can still serve as a stimulus for a whole series of experiments; and thirdly, we hope that some of the concepts outlined will still remain essentially correct and relatively stable. In view of the aforementioned, we should make the following reservations. First of all, we made no attempt to cite all the literature on the problems discussed, but considered it sufficient to illustrate the various premises with one or several sample references.

Mathematical biology is a fast growing, well recognised, albeit not clearly defined, subject and is, to my mind, the most exciting modern application of mathematics. The increasing use of mathematics in biology is inevitable as biol ogy becomes more quantitative. The complexity of the biological sciences makes interdisciplinary involvement essential. For the mathematician, biology opens up new and exciting branches while for the biologist mathematical modelling offers another research tool commmensurate with a new powerful laboratory technique but only if used appropriately and its limitations recognised. However, the use of esoteric mathematics arrogantly applied to biological problems by mathemati cians who know little about the real biology, together with unsubstantiated claims as to how important such theories are, does little to promote the interdisciplinary involvement which is so essential. Mathematical biology research, to be useful and interesting, must be relevant biologically.
