Book excerpt: This dissertation described preparation, characterization and intracellular localization of peptide-modified gold nanoparticles as multifunctional nuclear targeting vectors. The vectors have the potential advantages for intracellular diagnostics and therapeutic delivery. Gold nanoparticles were used in this study as carriers to combine multiple-functional cell-translocating peptides and antisense oligonucleotides. Two proteins, bovine serum albumin (BSA) and streptavidin (SA), have been systematically studied for crosslinking peptides or oligonucleotides with gold nanoparticles. Protocols for modifying gold nanoparticles with BSA-peptide conjugates are described within. The resulting constructs were characterized using a number of techniques including UV-visible absorbance spectroscopy and fluorescence spectroscopy in order to quantify peptide-BSA binding isotherms, exchange rates, critical flocculation concentrations, and the composition of mixed peptide-BSA monolayers on gold nanoparticles. The abilities of BSA-peptide/gold complexes to target the nucleus of several cell lines (HeLa, 3T3-NIH and HepG2) were explored. Four peptide/nanoparticle complexes were investigated, particles modified with the adenovirus nuclear localization signal (NLS), the adenovirus receptor-mediated endocytosis (RME) peptide, the adenovirus fiber containing the RME and NLS and the NLS for SV 40. The translocation of the gold nanoparticles was identified by video-enhanced color differential interference contrast (VEC-DIC) microscopy. The strong affinity between SA and biotin was employed to conjugate SA with biotinylated peptide and oligonucleotide. Systematical studies have been done to characterize the complexes, including SA adsorption on nanoparticles, biotinylated DNA adsorption on SA-coated nanoparticles, stabilities in solution, exchange rate in cell growth media and the composition of mixed SA-biotin peptide and SA-biotin DNA monolayers on gold nanoparticles. Book excerpt: Plasmon rulers consisting of optically coupled metal nanoparticles linked by biological macromolecules have provided a tool for bio-imaging and enzymology that is non-bleaching, non-blinking, and biocompatible. A pair of plasmonic nanoparticles couple when they are in spatial proximity, resulting in spectral shifts and increased scattering intensity. Previ- ous research has developed synthetic techniques for precisely assembling gold nanoparticles using DNA. Two self-assembled plasmonic nanoparticles have been shown to be an enzyme- responsive material in one-dimension. In order to visualize the subtle conformational changes ubiquitous in biological systems, three-dimensional, anisotropic nanoparticle assemblies are required. This dissertation reports two examples of next-generation plasmon rulers. DNA-assembled dimers of gold nanorods can act as a three-dimensional plasmon ruler only if the relative orientation of the particles can be controlled. By tuning the conditions of electrophoretic separation and the DNA structure, a sample of gold nanorod dimers that are attached side-by-side can be isolated from those attached end-to-end. TEM characterization of these oriented dimers shows a statistically significant deviation from randomly oriented nanorod pairs. Because of the high yield of oriented nanorod pairs, this colloidal sample can serve as an ensemble biosensor. A three-dimensional plasmon ruler can also be synthesized by increasing the complexity of the underlying DNA structure. Four DNA strands, each attached to a gold nanoparticle, hybridize such that each strand folds into one face of a chiral pyramid. The sequence of the four strands was designed to maximize the yield of one enantiomer based on tertiary structure of the double helices. This structure is a circular dichroism-based nanoparticle ruler for subtle biological events, such as the extension of DNA by a DNA-binding protein. Additionally, the DNA-assembled gold nanoparticle system was characterized by state- of-the-art electron microscopy. A method called individual particle electron tomography can be used to image single macromolecules. By tuning the parameters of the negative stain, the transmission electron microscope (TEM), and the buffer conditions, this method has been extended for the reconstruction of a 3D density map of an individual DNA double helix labeled by two nanoparticles. Previous work on cryo-EM imaging of DNA nanostructures without gold nanoparticles has relied on averaging many images together to create a com- posite density map. This work represents an electron microscopy constructed density map of a DNA double helix. Liquid-phase TEM can probe and visualize dynamic events with structural or functional details at the nanoscale in a liquid medium. The grapheme liquid cell was adopted to seal an aqueous sample solution containing DNA-assembled gold nanoparticle dimers against the high vacuum in TEM. Quantitative analysis of collected real time nanocrystal trajec- tories reveals the fidelity of the DNA in the presence of the electron beam as well as the reconstruction of the 3D configuration and motion of the nanostructure.

Their utility often stems from their unique optical and electronic properties, but their broad applicability is due to their robust surface chemistry that allows a wide variety of ligands to be bound to them to tune stability, solubility, and reactivity. However, there has been little work focusing on linking these two aspects of nanoparticles, despite their importance. The lack of work, in part, stems from the significant challenges arising from the inherent dispersity within nanoparticle populations and complicated characterization techniques in which sample measurement can be difficult or provide limited information. The focus of this thesis is to refine the understanding of the structure-property relationship in gold nanosystems by improving the characterization of gold surfaces and by controlling gold nanoparticle properties through surface chemistry. This thesis reports the characterization of the gold-thiol surface, where thermal treatment of gold nanoparticles (AuNPs) volatilized gold and sulfur, which was collected and subsequently quantified by inductively coupled plasma mass spectrometry and atomic emission spectroscopy. It was found that the mass loss in these samples ranged from 0-30\% gold by mass, rather than purely from the thiol ligands, and that the level of gold desorption is related to the Au--S bond strength.This study calls into question the usefulness of thermogravimetric analysis as a characterization technique of gold nanosytems. Additionally, the influence of surface chemistry on the electronic structure of gold nanosystems has been probed using magnetic spectroscopies. First, electron spin resonance has been used to examine the perturbation to the superatomic structure of Au$_{25}$(SR)$_#x18;$ nanoclusters (NCs) by a series of alkanethiolate ligands in hexane and tetrahydrofuran solvents. It has been found that the nanocluster electronic is sensitive to surface chemistry, with the $g_x$ component of the electronic \textit{g}-factor shifting linearly towards the free electron \textit{g}-value, $g_e$, and the perturbation is large, given the minute changes in ligand identity. The changes are dissimilar to those seen in gold nanoparticles with similar surface chemistry, indicating that use of AuNCs as model systems for larger, metallic AuNPs should be reevaluated. Finally, Evans Method nuclear magnetic resonance spectroscopy has been used to probe the density of states (DOS) of gold nanoparticles protected by aromatic thiolate ligands. Aromatic ligands are a common tool for tuning the electronic structure of inorganic complexes, and this thesis reports their capability to alter the electronic structure of gold nanoparticles. The DOS of these nanoparticles increases with increasing electron withdrawing character of the aromatic ligands, and it has been found that trends in controlling the gold nanoparticle's DOS found with aromatic thiols cannot necessarily be compared to those found with aliphatic thiols. This work examines not only how surface chemistry influences the properties of nanosystems, but also the usefulness of model systems and naive assumptions about characterization techniques.

The first part aroused from a long-lasting idea to manipulate adsorption of nanoparticles at liquid-liquid with an external electric field to form 'smart' mirrors and/or filters. Therefore, Chapters 3 to 5 are dedicated to explore fundamental aspects of charged nanoparticles self-assembly and to investigate optical properties (extinction and reflectance) in a through manner. Novel tetrathiafulvalene (TTF)-assisted method leads to self-assembly of nanoparticles into cm-scale nanofilms or, so-called, metal liquid-like droplets (MeLLDs) with remarkable optical properties. The second part (Chapters 6 to 8) clarifies the role of nanoparticles in interfacial electron transfer reactions. They demonstrate how nanoparticles are charged and discharged upon equilibration of Fermi levels with redox couples in solution and how it can be used to perform HER and ORR. Finally, Chapter 9 gives a perspective outlook, including applications of suggested methods in fast, one-step preparation of colloidosomes, SERS substrates as well as pioneer studies on so-called Marangony-type shutters drive by the electric field.

Despite the seemingly unlimited potential of nanotechnology, there is growing concern about the potential health and environmental impacts of the production and use of nanomaterials produced using the conventional methods. Recently, plants have been successfully used for the synthesis of nanoparticles in a nontoxic and environmentally benign manner. However, the potential applications of these biosynthesised nanoparticles are yet to be fully explored. The main objective of the present study was to develop and optimise a biological method for the synthesis of gold and silver nanoparticles, utilising tomato fruit extract as a reducing and stabilising agent. The nanoparticles obtained were characterised and compared to those produced using a chemical method. Scanning electron microscopy equipped with energy dispersive X-ray analysis confirmed the identity of the biosynthesised nanoparticles. The effects of pH, extract concentration, and temperature on the size, size distribution, and shape of nanoparticles were investigated. High temperatures and high pH were found to increase the production rate of nanoparticles, favouring the formation of monodisperse and smaller-sized nanoparticles. The biosynthesised gold and silver nanoparticles exhibited the characteristic plasmon absorption peaks, which centred at approximately 530 nm and 410 nm, respectively. In addition, the nanoparticles were observed to have a net negative charge and were crystallised in a face-centred cubic structure. Fourier-transform infrared spectroscopy analysis indicated that the nanoparticles were capped with biomolecules derived from the tomato extract. Biosynthesised gold nanoparticles were shown to bind efficiently to a synthetic metal-binding polypeptide, implying their potential in therapeutic delivery applications. In a follow-up experiment, the ESAT-6 antigen from Mycobacterium tuberculosis was fused with the metal-binding polypeptide and successfully expressed in Nicotiana benthamiana leaves, with recombinant protein yields ranging from 0.01% to 0.23% of total soluble protein. The recombinant proteins were purified and their identities were confirmed by western blot showing the expected molecular weights. The purified proteins were conjugated to three different samples of biosynthesised gold nanoparticles covering different size ranges. The resultant AuNP-GBP:ESAT-6 conjugates were then used to immunise C57BL/6 mice. The results showed that GBP:ESAT-6 formulated with AuNPs with an average size of 10.3 nm induced elevated antigen-specific immune responses. In addition, no toxic effects were observed in mice treated with the biosynthesised AuNPs. Taken together, this study provides an additional evidence with respect to the use of plant-derived materials as an eco-friendly, nontoxic and viable approach for the synthesis of nanoparticles.
