This book was released on 2015 with total page 220 pages. Available in PDF, EPUB and Kindle. Book excerpt: Cowpea (Vigna unguiculata) is an edible annual leguminous crop cultivated by many subsistence and rural communities in developing countries in subtropical areas of the world. Cowpea is a very versatile, well adapted and nutritious grain legume. However, cowpea seed is prone to fungal infestation under suboptimal storage conditions. Some of these fungi, including Aspergillus and Fusarium spp., produce their associated mycotoxins under these conditions. Fumonisins are produced primarily by Fusarium verticillioides and F. proliferatum and are known to be toxic to vertebrates and plants. Fumonisin B1 is phytotoxic to cowpea seeds and is speculated to inhibit ceramide synthase in the sphingolipid pathway in plants. This study was done to determine the phytotoxic effects of FB1 on cowpea seedlings and to provide insight on the mode of action of the toxin at a molecular level. Surface-disinfected seeds were imbibed for 10 h in sterile distilled water amended with FB1 to yield final concentrations of 2, 20 and 40 mg/L. Slow imbibed seeds (placed in moist paper towels) and seeds placed in sterile distilled water for 10 h served as the positive and negative controls, respectively. Additionally cowpea seeds were inoculated with the conidia of three different FB1-producing Fusarium verticillioides strains. Percentage emergence was determined after seven dpi whereas, seedling mass, length and seedling vigour index where determined after seven and 21 dpi. Total chlorophyll content was measured after 14 and 21 dpi. Stunted growth was observed in FB1 treated seedlings. Emergence was reduced by all three FB1 concentrations as well as in seeds treated with F. verticillioides strain MRC 8265. Seedlings imbibed in 40 mg/L FB1 had reduced seedling length. Seedling mass was reduced by all three FB1 concentrations as well as all three Fusarium strains. Total chlorophyll content was higher for seeds imbibed in all three FB1 concentrations when compared to both controls. This is in contrast to other studies which reported that FB1 causes chlorosis in plants. In order to evaluate the effect of FB1 on ceramide synthase gene expression seeds were imbibed in a 20 mg/L FB1 solution and the control seeds were imbibed in sterile distilled water. RNA was extracted from untreated and treated samples after 0, 3 and 12 dpi with the latter being divided into shoot and roots samples. cDNA was synthesised from the extracted RNA samples. Amplification of the ceramide synthase gene was done using primers that were designed to the conserved regions of Glycine max and Phaseolus vulgaris ceramide synthase gene homologues. Sequence analysis revealed that the designed primers did amplify the correct gene from cowpea seedlings. The expression levels of the ceramide synthase gene from cowpea were thus tested using semi-quantitative PCR amplification. No significant differences in ceramide synthase gene expression were observed between the control and toxin treated samples. Two FB1 unrelated differences were observed. A possible developmental difference was observed as ceramide synthase gene expression decreased over time in both the control and treated samples. It thus seems that FB1 did not influence ceramide synthase gene expression in cowpea.

Available in PDF, EPUB and Kindle. Book excerpt: Indian agriculture has witnessed spectacular advances in agricultural production in the last few decades. This was possible through green revolution in mid 60s leading to the country’s remarkable achievement in food grains and edible oil production. Seed has always been regarded as the most vital, basic and critical input in agriculture. It is interesting to note that today seed demand of only 10% farmers is met. Efforts are needed to provide good quality certify seeds to farmers. In recent years the awareness for seed health has increased among the growers, traders, and consumers. In post-GATT era and with emergence of WTO, seed health has assumed the global concern. Infection of seed borne pathogens results into seed rots, seedling decay, pre and post emergence mortality, distortion, discoloration, reduced seed size and shrivelledness of seeds. Their study and identification is primarily required to achieve satisfactory control. The book will be of great help to students, researchers, and people engaged in seed production activities. In this book, information about different seed borne diseases is presented in 20 chapters. There is an urgent need to find out effective, alternative methods of diseases control, which are less harmful to human beings and environment. There are chapters dealing with botanical pesticides and quarantine regulations. A chapter describes molecular aspects involved in seed borne diseases. The contents are divided into two parts (I) Seed borne diseases and (II) Wilt and foliar diseases and their control methods. Ecofriendly measures to control seed borne diseases are dealt in detail. The book provides comprehensive and integrated information on management of seed health written by experts in the field. It will be especially useful for students and young people involved in seed testing, seed industry as well as in teaching. Besides Agriculture Universities the book will be useful for all others offering courses related to Phytopathology and Seed Technology.

Book excerpt: This authored book gives insight into the morphology, physiology, genetics, plant protection and biotechnology of three important arid legumes, viz., moth bean, cowpea and horse gram. There are seven chapters for each crop that provide in-depth information on cultivation, genetic improvement, plant protection measures, management of physiological and abiotic stresses along with related genetic markers and biotechnological advances pertaining to these legumes. The chapters present research findings and brief reviews concerning the advances made in the improvement of these legumes.Legumes are utilized as pulses or grains aiding as an important source of protein for both human and animal consumption and also provide raw materials to the food and feed industries. They are also valuable for soil building, improving soil quality and biological nitrogen fixation. Physiology and genetics provide an inimitable source of information on the distinct aspects of basic and applied legume research for general readers, students, academicians and researchers. Arid legumes are crops characterized by inherent features and capabilities to withstand adverse and harsh climatic conditions, significantly replenish the soil, as well as provide protein and micronutrients. Adaptability to several stresses including drought makes them key to agriculture in areas receiving scanty rainfall. This comprehensive book disseminates significant information on the genetic diversity, cultivation, manipulation through mutagenic techniques, molecular biology and other breeding techniques. The book, therefore, is of importance to teachers, researchers and policymakers who are interested to acquire knowledge about moth bean, cowpea and horse gram. It also serves as an additional reading material for MSc and PhD students of agriculture and environmental sciences. The book is also useful to national and international agricultural scientists and policymakers.

Book excerpt: This publication provides an approach by which conservationists can determine whether or not long-term seed storage is feasible for a particular species, i.e. whether or not that species shows orthodox seed storage behaviour, and provides advice on the implementation of the protocol, examples of ways in which the results from seed storage studies could be misinterpreted due to confounding factors, as well as several alternative approaches for estimating seed storage behaviour prior to carryng out actual investigations with the seeds. In particular, the latter section introduces the concept of a multicriteria approach for estimating seed storage behaviour.
