Book excerpt: The papers collected in this volume reproduce contributions by leading sch- arstoaninternationalschoolandworkshopwhichwasorganizedandheldwith thegoaloftakinga snapshotofadiscipline undertumultuous growth. Indeed, the area of protein folding, docking and alignment is developing in response to needs for a mix of heterogeneous expertise spanning biology, chemistry, mathematics, computer science, and statistics, among others. Some of the problems encountered in this area are not only important for the scienti?c challenges they pose, but also for the opportunities they disclose intermsofmedicalandindustrialexploitation. Atypicalexampleiso?eredby protein-drug interaction (docking), a problem posing daunting computational problems at the crossroads of geometry, physics and chemistry, and, at the same time, a problem with unimaginable implications for the pharmacopoeia of the future. The schoolfocused on problems posed by the study of the mechanisms - hind protein folding, and explored di?erent ways of attacking these problems under objective evaluations of the methods. Together with a relatively small core of consolidated knowledge and tools, important re?ections were brought to this e?ort by studies in a multitude of directions and approaches. It is obviously impossible to predict which, if any, among these techniques will prove completely successful, but it is precisely the implicit dialectic among them that best conveys the current ?avor of the ?eld. Such unique diversity and richness inspired the format of the meeting, and also explains the slight departure of the present volume from the typical format in this series: the exposition of the current sediment is complemented here by a selection of quali?ed specialized contributions.

Book excerpt: It is increasingly evident that polyunsaturated fatty acids (PUFA), which in the past were often believed to be mere components of cellular membranes of neural tissue, are actually major determinants of the functional properties of neural cells and are intimately involved in brain disease processes. The two families of PUF A, which are derived from the essential polyenes, linoleic (18:26) and a-linolenic (18:33) acids, constitute a major proportion (-30%) of the total fatty acids esterified to brain phospholipids. Each family is distributed in membranes in a highly specific manner with respect to brain region, cell type, and position within each of the phosphoglycerides. Unlike their behavior in other organs, the PUF A components of brain turn over slowly and are relatively resistant to dietary modification. Their unique distribution and stability suggest that they play an important role in determining the functional properties of neural cells. However, until recently very little was known regarding the precise nature of their involvement.

Book excerpt: This assessment of the technical quality and relevance of the programs of the Measurement and Standards Laboratories of the National Institute of Standards and Technology is the work of the 165 members of the National Research Council's (NRC's) Board on Assessment of NIST Programs and its panels. These individuals were chosen by the NRC for their technical expertise, their practical experience in running research programs, and their knowledge of industry's needs in basic measurements and standards. This assessment addresses the following: The technical merit of the laboratory programs relative to the state of the art worldwide; The effectiveness with which the laboratory programs are carried out and the results disseminated to their customers; The relevance of the laboratory programs to the needs of their customers; and The ability of the laboratories' facilities, equipment, and human resources to enable the laboratories to fulfill their mission and meet their customers' needs.

Book excerpt: Membrane proteins, representing nearly 40% of all proteins, are key components of cells involved in many cellular processes, yet only a small number of their structures have been determined. Membrane Protein Structure Determination: Methods and Protocols presents many detailed techniques for membrane protein structure determination used today by bringing together contributions from top experts in the field. Divided into five convenient sections, the book covers various strategies to purify membrane proteins, approaches to get three dimensional crystals and solve the structure by x-ray diffraction, possibilities to gain structural information for a membrane protein using electron microscopy observations, recent advances in nuclear magnetic resonance (NMR), and molecular modelling strategies that can be used either to get membrane protein structures or to move from atomic structure to a dynamic understanding of a molecular functioning mechanism. Written in the highly successful Methods in Molecular BiologyTM series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Comprehensive and easy to use, Membrane Protein Structure Determination: Methods and Protocols serves as an ideal reference for scientists seeking to further our knowledge of these vital and versatile proteins as well as our overall understanding of the complicated world of cell biology.

Available in PDF, EPUB and Kindle. Book excerpt: Development of new imaging technologies in recent years has transformed neuroscience in profound ways. Following on the heels of the revolution based on the Green Fluorescent Protein, refined genetically-encoded fluorescent reporters and genetic targeting strategies now enable optical recording of synaptic transmission in defined neuronal populations at speeds approaching the enviable temporal resolution of electrophysiology. Super-resolution light microscopy permits observation of synapses and their molecular machinery at sub-diffraction resolution. At the ultrastructural level, automated forms of electron microscopy, improvements in specimen fixation methods, and recent efforts to correlate data from light and electron micrographs now make the reconstruction of functional neural circuits a reality. Finally, the use of optogenetic actuators, such as channelrhodopsins, allows precise temporal and spatial manipulation of neuronal activity and is revealing profound insights into the organization of neural circuits and their roles in behavior. This research topic highlights recent advances in both light and electron microscopy, with a specific focus on approaches that combine innovations from several different fields to obtain novel information about synapse structure and function. We are confident that this collection of articles - three original research papers, six reviews, one methods paper and one perspective article - will enable neuroscientists to achieve the next generation of experiments aimed at cracking the neural code.
