Book excerpt: Folded peptides - and peptide motifs within proteins - are abundant in living organisms, where they are essential for the biological activities of the peptides and proteins. During the past decades, much research has been dedicated to understanding the rules that govern peptide folding. Simultaneously, a range of strategies have been established for the conformational stabilization of bioactive peptides, as well as for the de novo design of peptides with defined secondary structures. These methods are either based on the chemical modification of the peptide backbone, such as cyclization and stapled peptides, or on the use of a range of non-proteinogenic amino acids that, in a defined sequential arrangement, induce secondary structures peptides. Such building blocks include D- and other non-proteinogenic amino acids, as well as beta- and gamma-amino acids. This Research Topic comprises a collection of papers by an international group of 77 scientists with a background in synthetic, analytical, computational and medicinal chemistry, as well as in biochemistry and pharmacology. Their research is presented here in a total of 11 papers (8 original research reports and 3 reviews), covering diverse aspects of folded synthetic peptides. These studies include the preparation and characterization of new peptide monomers with interesting folding properties, the synthesis and conformational analysis of non-natural peptides, as well as the use of folded peptidomimetics as molecular switches. Additionally, a range of biomedical applications, such as antimicrobial, anti-inflammatory, antiangiogenic and immune-stimulating activities, are also reported. We hope this eBook will be a source of inspiration and knowledge for scientist in various disciplines related to folded peptides and their many applications, as well as for those who want to learn more about this fascinating field of research.

Available in PDF, EPUB and Kindle. Book excerpt: A peptidomimetic is a small protein-like chain designed to mimic a peptide with adjusted molecular properties such as enhanced stability or biological activity. It is a very powerful approach for the generation of small-molecule-based drugs as enzyme inhibitors or receptor ligands. Peptidomimetics in Organic and Medicinal Chemistry outlines the concepts and synthetic strategies underlying the building of bioactive compounds of a peptidomimetic nature. Topics covered include the chemistry of unnatural amino acids, peptide- and scaffold-based peptidomimetics, amino acid-side chain isosteres, backbone isosteres, dipeptide isosteres, beta-turn peptidomimetics, proline-mimetics as turn inducers, cyclic scaffolds, amino acid surrogates, and scaffolds for combinatorial chemistry of peptidomimetics. Case studies in the hit-to-lead process, such as the development of integrin ligands and thrombin inhibitors, illustrate the successful application of peptidomimetics in drug discovery.

Book excerpt: Peptidomimetics are compounds which mimic the biological activity of peptides while offering the advantages of increased bioavailability, biostability, bioefficiency, and bioselectivity against the natural biological target of the parent peptide. Examples of peptidomimetics have been isolated as natural products, synthesized as libraries from novel subunits, and designed on the basis of X-ray crystallographic studies and through an intricate knowledge of the biological mode of action of natural peptides. They offer challenging synthetic targets and are increasingly important medicinal agents and biological probes. As a consequence, peptidomimetics embrace much of what is modern medicinal and organic chemistry. This volume highlights some recent and exciting developments in the area.

Book excerpt: Natural Products: Discourse, Diversity and Design provides an informative and accessible overview of discoveries in the area of natural products in the genomic era, bringing together advances across the kingdoms. As genomics data makes it increasingly clear that the genomes of microbes and plants contain far more genes for natural product synthesis than had been predicted from the numbers of previously identified metabolites, the potential of these organisms to synthesize diverse natural products is likely to be far greater than previously envisaged. Natural Products addresses not only the philosophical questions of the natural role of these metabolites, but also the evolution of single and multiple pathways, and how these pathways and products may be harnessed to aid discovery of new bioactives and modes of action. Edited by recognized leaders in the fields of plant and microbial biology, bioorganic chemistry and natural products chemistry, and with contributions from researchers at top labs around the world, Natural Products is unprecedented in its combination of disciplines and the breadth of its coverage. Natural Produces: Discourse, Diversity and Design will appeal to advanced students and experienced researchers, from academia to industry, in diverse areas including ecology, industrial biotechnology, drug discovery, medicinal chemistry, agronomy, crop improvement, and natural product chemistry.
