Available in PDF, EPUB and Kindle. Book excerpt: Neurotransmission is a multicomponent process. Transmitters, released by neuronal activity, act on pre- and postsynaptic receptors, and many books detail advances in the receptor field. In addition, after their release from nerve endings, transmitters are removed from the neuronal vicinity by uptake into neuronal or glial cells by specific tra- porter proteins that have been studied intensely over the last 30 years; this information is scattered throughout numerous publishing vehicles. Therefore, the primary aim of this second edition of N- rotransmitter Transporters: Structure, Function, and Regulation is to offer a comprehensive picture of the characterization of neurotransmitter transporters and their biological roles. The transporter field has moved forward in stages. In the first phase, progress came from the use of substrate or blocker ligands selectively targeting transporters, the application of model systems allowing the study of transmitter tra- port shielded from storage, and the development of mathematical models for describing transport phenomena. In the second phase, roughly covering the last decade, advances in DNA techniques allowed the cloning of numerous genes coding for different transporter proteins. In the current, third stage, a wealth of information is being accumulated in studies relating transporter structure with function, experiments addressing regulation by posttranslational transfor- tion, investigations into transport modulation by trafficking processes and genomic influences, characterization of channel properties of tra- porters by electrophysiological approaches, and the creation of transgenic animals under- or overexpressing a given transporter protein.

Book excerpt: This book presents an authoritative review of the most significant findings about all the epigenetic targets (writers, readers, and erasers) and their implication in physiology and pathology. The book also covers the design, synthesis and biological validation of epigenetic chemical modulators, which can be useful as novel chemotherapeutic agents. Particular attention is given to the chemical mechanisms of action of these molecules and to the drug discovery prose which allows their identification. This book will appeal to students who want to know the extensive progresses made by epigenetics (targets and modulators) in the last years from the beginning, and to specialized scientists who need an instrument to quickly search and check historical and/or updated notices about epigenetics.

Available in PDF, EPUB and Kindle. Book excerpt: This volume provides a collection of contemporary perspectives on using activity-based protein profiling (ABPP) for biological discoveries in protein science, microbiology, and immunology. A common theme throughout is the special utility of ABPP to interrogate protein function and small-molecule interactions on a global scale in native biological systems. Each chapter showcases distinct advantages of ABPP applied to diverse protein classes and biological systems. As such, the book offers readers valuable insights into the basic principles of ABPP technology and how to apply this approach to biological questions ranging from the study of post-translational modifications to targeting bacterial effectors in host-pathogen interactions.

Available in PDF, EPUB and Kindle. Book excerpt: Stimulant drugs are widely used in the treatment of ADHD in children and adults. Hundreds of studies over the past 60 years have demonstrated their effectiveness in improving attention span, increasing impulse control, and reducing hyperactivity and restlessness. Despite widespread interest in these compounds, however, their mechanisms of action in the central nervous system have remained poorly understood. Recent advances in the basic and clinical neurosciences now afford the possibility of elucidating these mechanisms. The current volume is the first to bring this expanding knowledge to bear on the central question of why and how stimulants exert their therapeutic effects. The result is a careful, comprehensive, and insightful integration of material by well-known scientists that significantly advances our understanding of stimulant effects and charts a course for future research. Part I presents a comprehensive description of the clinical features of ADHD and the clinical response to stimulants. Part II details the cortical and subcortical neuroanatomy and functional neurophysiology of dopamine and norepinephrine systems with respect to the regulation of attention, arousal, activity, and impulse control and the effects of stimulants on these systems. Part III is devoted to clinical research, including recent studies of neuroimaging, genetics, pharmacodynamic and pharmacokinetic properties of stimulants, effects on cognitive functions, neurophysiological effects in humans with and without ADHD and in non-human primates, and comparison of stimulants and non-stimulants in the treatment of ADHD. Part IV is a masterful synthesis that presents alternative models of stimulant drug action and generates key hypotheses for continued research. The volume will be of keen interest to researchers and clinicians in psychiatry, psychology, and neurology, neuroscientists studying stimulants, and those pursuing development of new drugs to treat ADHD.

Available in PDF, EPUB and Kindle. Book excerpt: Plants produce a huge array of natural products (secondary metabolites). These compounds have important ecological functions, providing protection against attack by herbivores and microbes and serving as attractants for pollinators and seed-dispersing agents. They may also contribute to competition and invasiveness by suppressing the growth of neighboring plant species (a phenomenon known as allelopathy). Humans exploit natural products as sources of drugs, flavoring agents, fragrances and for a wide range of other applications. Rapid progress has been made in recent years in understanding natural product synthesis, regulation and function and the evolution of metabolic diversity. It is timely to bring this information together with contemporary advances in chemistry, plant biology, ecology, agronomy and human health to provide a comprehensive guide to plant-derived natural products. Plant-derived natural products: synthesis, function and application provides an informative and accessible overview of the different facets of the field, ranging from an introduction to the different classes of natural products through developments in natural product chemistry and biology to ecological interactions and the significance of plant-derived natural products for humans. In the final section of the book a series of chapters on new trends covers metabolic engineering, genome-wide approaches, the metabolic consequences of genetic modification, developments in traditional medicines and nutraceuticals, natural products as leads for drug discovery and novel non-food crops.

Available in PDF, EPUB and Kindle. Book excerpt: The availability of various in vitro and in vivo techniques has considerably advanced the research on drug transport and metabolism across the blood-brain barrier (BBB). These specialized and sophisticated experimental strategies are of fundamental importance if one is to gain a greater understanding of enhanced and selective drug delivery to the brain. The reader will find in this book methods for in vitro endothelial/astrocyte cell culture models, and for in vivo intracerebral microdialysis to study drug tranport across the BBB. This book, however, is not merely a laboratory manual consisting of recipes for BBB research; it permits the presentation of the different methods in fine detail, revealing tricks and short cuts that frequently do not appear in the literature. The researcher is well aware that differences (subtle or otherwise) in experimental steps used in different laboratories may influence the outcome of any particular procedure. The book also illustrates the accessibility and the application of the different methods in different species. Background information of the protocol is given in every chapter, which also contains a literature list that the reader may wish to refer to for further information. This volume will be invaluable to basic researchers as well as to those involved in the search for agents suitable for pharmaceutic intervention in the central nervous system.
