Available in PDF, EPUB and Kindle. Book excerpt: While the genomic revolution has quickly led to the deposit of more than 30,000 structures in the protein data bank (PDB), less than one percent of those contributions represent membrane proteins despite the fact that membrane proteins constitute some 20 percent of all proteins. This discrepancy becomes significantly troublesome when it is coupled
Book Synopsis Structural Genomics, Part B by : Andrzej Joachimiak
Download or read book Structural Genomics, Part B written by Andrzej Joachimiak and published by Academic Press. This book was released on 2009-11-26 with total page 117 pages. Available in PDF, EPUB and Kindle. Book excerpt: Structural genomics is the systematic determination of 3-D structures of proteins representative of the range of protein structure and function found in nature. The goal is to build a body of structural information that will predict the structure and potential function for almost any protein from knowledge of its coding sequence. This is essential information for understanding the functioning of the human proteome, the ensemble of tens of thousands of proteins specified by the human genome. While most structural biologists pursue structures of individual proteins or protein groups, specialists in structural genomics pursue structures of proteins on a genome wide scale. This implies large-scale cloning, expression and purification. One main advantage of this approach is economy of scale. Examines the three dimensional structure of all proteins of a given organism, by experimental methods such as X-ray crystallography and NMR spectroscopy Looks at structural genomics as a foundation of drug discovery as discovering new medicines is becoming more challenging and the pharmaceutical industry is looking to new technologies to help in this mission

Book Synopsis Structural Proteomics and Its Impact on the Life Sciences by : Joel Sussman
Download or read book Structural Proteomics and Its Impact on the Life Sciences written by Joel Sussman and published by World Scientific. This book was released on 2008 with total page 597 pages. Available in PDF, EPUB and Kindle. Book excerpt: The role played by structural proteomics in the first decade of the 21st century is equivalent to that played by the Human Genome Project in the last decade of the 20th century. The development of high-throughput technologies that permit the solution of hundreds of 3D structures of individual proteins, proteinOCoprotein complexes and proteinOCodrug complexes, just by one laboratory in a single year, will provide a knowledge base which will change the face of structural biology. This will have an immediate influence on medicinal chemistry and molecular pharmacology, as well as an increasing impact on such disciplines as neurobiology, developmental biology, immunology and molecular medicine. This book presents a state-of-the-art overview of the structural proteomics field, ranging from policy issues related to funding and goals, through the high-throughput procedures for protein production, to the solution of the structures of proteins and higher-order entities, via a multidisciplinary approach involving molecular biology, X-ray crystallography, NMR and electron microscopy, as well as bioinformatics analysis. This is the first book to provide such a comprehensive coverage of a rapidly evolving field.

Available in PDF, EPUB and Kindle. Book excerpt: Structural Genomics and Drug Discovery: Methods and Protocols focuses on high throughput structure determination methods and how they can be applied to lay the groundwork for structure aided drug discovery. The methods and protocols that are described can be applied in any laboratory interested in using detailed structural information to advance the initial stages of drug discovery. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and key tips on troubleshooting and avoiding known pitfalls. Authoritative and practical, Structural Genomics and Drug Discovery: Methods and Protocols seeks to aid scientists in the further study into structural genomics approach as an efficient initial step toward drug discovery and the methods described will be useful to anyone interested in moving in this direction.
Book Synopsis Crystallization of Membrane Proteins by : Hartmut Michel
Download or read book Crystallization of Membrane Proteins written by Hartmut Michel and published by CRC Press. This book was released on 2018-01-18 with total page 343 pages. Available in PDF, EPUB and Kindle. Book excerpt: The precise knowledge of the structure of biological macromolecules forms the basis of understanding their function and their mechanism of action. It also lays the foundation for rational protein and drug design. The only method to obtain this knowledge is still crystallography. At present, the structures of about 400 proteins are known at or nearly at atomic proteins. However, only two of them are membrane proteins or complexes of the membrane proteins. The reasons for the difference is not the crystals of membrane proteins resists forming special problems when being analysed. The reason is that the membrane proteins resist into forming into well-ordered crystals. The intention of this book is to help to produce well-ordered crystals proteins and to provide guidelines, it is aimed at both biochemists and protein crystallographer‘s.

Book Synopsis Sequence — Evolution — Function by : Eugene V. Koonin
Download or read book Sequence — Evolution — Function written by Eugene V. Koonin and published by Springer Science & Business Media. This book was released on 2013-06-29 with total page 482 pages. Available in PDF, EPUB and Kindle. Book excerpt: Sequence - Evolution - Function is an introduction to the computational approaches that play a critical role in the emerging new branch of biology known as functional genomics. The book provides the reader with an understanding of the principles and approaches of functional genomics and of the potential and limitations of computational and experimental approaches to genome analysis. Sequence - Evolution - Function should help bridge the "digital divide" between biologists and computer scientists, allowing biologists to better grasp the peculiarities of the emerging field of Genome Biology and to learn how to benefit from the enormous amount of sequence data available in the public databases. The book is non-technical with respect to the computer methods for genome analysis and discusses these methods from the user's viewpoint, without addressing mathematical and algorithmic details. Prior practical familiarity with the basic methods for sequence analysis is a major advantage, but a reader without such experience will be able to use the book as an introduction to these methods. This book is perfect for introductory level courses in computational methods for comparative and functional genomics.

Book Synopsis Membrane Protein Protocols by : Barry S. Selinsky
Download or read book Membrane Protein Protocols written by Barry S. Selinsky and published by Springer Science & Business Media. This book was released on 2008-02-03 with total page 330 pages. Available in PDF, EPUB and Kindle. Book excerpt: Knowledge of the three-dimensional structure of a protein is absolutely required for the complete understanding of its function. The spatial orientation of amino acids in the active site of an enzyme demonstrates how substrate specificity is defined, and assists the medicinal chemist in the design of s- cific, tight-binding inhibitors. The shape and contour of a protein surface hints at its interaction with other proteins and with its environment. Structural ana- sis of multiprotein complexes helps to define the role and interaction of each individual component, and can predict the consequences of protein mutation or conditions that promote dissociation and rearrangement of the complex. Determining the three-dimensional structure of a protein requires milligram quantities of pure material. Such quantities are required to refine crystallization conditions for X-ray analysis, or to overcome the sensitivity limitations of NMR spectroscopy. Historically, structural determination of proteins was limited to those expressed naturally in large amounts, or derived from a tissue or cell source inexpensive enough to warrant the use of large quantities of cells. H- ever, with the advent of the techniques of modern gene expression, many p- teins that are constitutively expressed in minute amounts can become accessible to large-scale purification and structural analysis.
