Book excerpt: Since its inception the research area of platelet pharmacology has always been a vigorous one and it is a characteristic that new approaches to the understanding of platelet function are rapidly and thoroughly investigated. The intensity of this activity is attri butable, probably, to an appreciation by research workers in the field that a satisfactory therapeutic control of platelet function has yet to be realized. Also that if and when this problem of con trolling platelet function is achieved the benefits to clinical medicine will be immense since platelets are known to be involved in a multiplicity of events coupled within the haemostatic mechanisms and inflammatory responses. Aberrations in the behaviour of plate lets is part of the aetiology of atherosclerosis, myocardial and cerebral infarction and thrombosis. At this point in time, research in platelet function is in a particularly rapid state of flux. The recent findings of research workers active in the field and also workers investigating mechan isms of stimulus response coupling in other cells, have provided interesting insights into the generality of mechanisms involved in the function of responsive cells. One may itemize these developments as the area of cell receptor/ligand interaction, induction of cell ular response by protein phosphorylation and calcium flux. The mech anism of these latter events occurs through the activity of phospho lipase generating transient intermediates. These intermediates may act as ionophores or enzyme activators or may, in the case of eico sanoids, reinforce and make irreversible the cellular response.

Book excerpt: This volume contains a comprehensive collection of methods for the isolation of platelet membranes, subcellular organelles, the cytoskeleton, and for the assay and purification of platelet receptors. Key Features * Platelets are cellular elements with the highest density of receptors per membrane surface area involved in binding of adhesive molecules, clotting, enzymes, and vasoactive amines * Platelets are essential for the arrest of bleeding and for formation of intravascular thrombi contributing to heart attacks, strokes, and disseminated intravascular coagulation, and are involved in immune complex disease * Platelets are useful for studying the processes of cell adhesion, neurotransmitter uptake, stimulus-response coupling, including signal transduction and secretion

Book Synopsis Biochemistry of Platelets by : David Phillips
Download or read book Biochemistry of Platelets written by David Phillips and published by Elsevier. This book was released on 2012-12-02 with total page 477 pages. Available in PDF, EPUB and Kindle. Book excerpt: Biochemistry of Platelets is a comprehensive review of the biochemistry of platelets, with emphasis on the molecular basis for the various biological processes in which they participate. Topics range from stimulus-response coupling mechanisms to platelet contractile proteins, platelet membrane glycoproteins, and storage organelles in platelets and megakaryocytes. The expression and function of adhesive proteins on the platelet surface is also discussed. Comprised of 12 chapters, this book begins with a description of the morphological and metabolic responses to agonists, as well as the involvement of certain processes in the coupling of agonist-receptor interactions to platelet responses. The following chapter deals with platelet arachidonate metabolism and platelet-activating factor, focusing on the release of arachidonate from platelet lipid stores; pathways of platelet arachidonate metabolism and effects of arachidonate metabolites; and inhibition of platelet arachidonate metabolism by aspirin. The structure, function, and modification in disease of platelet membrane glycoproteins are then discussed, along with prothrombin activation on platelets and platelet regulation of thrombus formation. Secreted platelet proteins as markers for pathological disorders are also considered. This monograph is intended as a reference for investigators involved in platelet research as well as a source of information for those working in other areas of biological investigation.

Book Synopsis Immunopharmacology of Platelets by : M. Joseph
Download or read book Immunopharmacology of Platelets written by M. Joseph and published by Academic Press. This book was released on 1995-07-05 with total page 267 pages. Available in PDF, EPUB and Kindle. Book excerpt: Platelets have long been the subject of much investigation in the areas of vascular physiology, biochemistry, immunology and physiopathology. Their ability to react to environmental stimuli, to modify their shape and metabolism despite being anuclear, has fascinated scientists. This volume looks specifically at the involvement of blood platelets outside their 'classical' field of application, in the physiopathological mechanics of allergy and inflammation. The coverage includes the role of platelets in bacterial, parasitic and viral diseases and in tumour cell interactions.
