Book excerpt: Asymmetric Synthesis, Volume 4: The Chiral Carbon Pool and Chiral Sulfur, Nitrogen, Phosphorus, and Silicon Centers describes the practical methods of obtaining chiral fragments. Divided into five chapters, this book specifically examines initial chiral transmission and extension. The opening chapter describes the so-called chiral carbon pool, the readily available chiral carbon fragments used as building blocks in synthesis. This chapter also provides a list of 375 chiral building blocks, along with their commercial sources, approximate prices, and methods of synthesis. Schemes involving almost 1,300 structures and the use of chiral building blocks to prepare alkaloids, arachidonic acid metabolites, compounds with chemotherapeutic and pharmacological activity, insect pheromones, sugars, vitamins, terpenoids, and miscellaneous compounds are also included in this text. The remaining chapters provide a similar overview of the chemistry of compounds chiral at sulfur, phosphorus, nitrogen, and silicon. Synthetic chemists and researchers, especially those engaged in pharmaceutical research and specialty chemicals production, will find this book invaluable.

Available in PDF, EPUB and Kindle. Book excerpt: A thorough understanding of stereochemistry is essential for the comprehension of almost all aspects of modern organic chemistry. It is also of great significance in many biochemical and medicinal disciplines, since the stereoisomers of a compound can have dramatically different biological properties. This text explains how the different properties of stereoisomers of a compound arise, and what processes can be used to prepare and analyze stereoisomerically pure compounds. It also presents prominent coverage of the stereochemistry of inorganic and organometallic compounds, which is likely to increase in importance, as these compounds are used as symmetric catalysts in asymmetric synthesis. Modern stereochemical terminology is used throughout, although reference is also made to older terms which are still widely used. A set of problems at the end of each chapter aims to further the reader's understanding of how the content can be applied. The book is designed mainly as a textbook for undergraduate students and as a reference source for more advanced levels, but is also intended for academic and professional organic chemists.

Available in PDF, EPUB and Kindle. Book excerpt: Molecular shape, form, and symmetry play a central role in organic chemistry, and this text presents a brief introduction to the conceptual basis of stereochemistry. Its focus lies in the fundamentals of structural stereochemistry, rather than the dynamic aspects that are more relevant to reaction mechanisms. The three-part treatment deals with structure and symmetry, stereoisomerism, and the separation and configuration of stereoisomers. The first section reviews molecular architecture, relating empirical bonding geometries to the hybridization of the central carbon atom. Students receive a nonrigorous treatment of symmetry elements and point groups, with particular focus on the presence or absence of reflection symmetry. The second section classifies stereoisomers according to symmetry properties and to the nature of their barriers; it also discusses the dependence of optical activity on structure and concludes with an examination of topological isomerism. The third and final section explores the conceptual basis of asymmetric syntheses and kinetic resolutions. Each of the major sections features a series of exercises that reinforce and extend the preceding material, and answers are provided. Preface to the Dover edition. Answers to Exercises. Bibliography. Index.

Available in PDF, EPUB and Kindle. Book excerpt: The world is chiral. Most of the molecules in it are chiral, and asymmetric synthesis is an important means by which enantiopure chiral molecules may be obtained for study and sale. Using examples from the literature of asymmetric synthesis (more than 1300 references), the aim of this book is to present a detailed analysis of the factors that govern stereoselectivity in organic reactions. It is important to note that the references were each individually checked by the authors to verify relevance to the topics under discussion. The study of stereoselectivity has evolved from issues of diastereoselectivity, through auxiliary-based methods for the synthesis of enantiomerically pure compounds (diastereoselectivity followed by separation and auxiliary cleavage), to asymmetric catalysis. In the latter instance, enantiomers (not diastereomers) are the products, and highly selective reactions and modern purification techniques allow preparation - in a single step - of chiral substances in 99% ee for many reaction types. After an explanation of the basic physical-organic principles of stereoselectivity, the authors provide a detailed, annotated glossary of stereochemical terms. A chapter on "Analytical Methods" provides a critical overview of the most common methods for analysis of stereoisomers. The authors then follow the 'tried-and-true' format of grouping the material by reaction type. Thus, there are four chapters on carbon-carbon bond forming reactions (enolate alkylations, organometal additions to carbonyls, aldol and Michael reactions, and cycloadditions and rearrangements), one chapter on reductions and hydroborations (carbon-hydrogen bond forming reactions), and one on oxidations (carbon-oxygen and carbon-nitrogen bond forming reactions). Leading references are provided to natural product synthesis that have been accomplished using a given reaction as a key step. In addition to tables of examples that show high selectivity, a transition state analysis is presented to explain - to the current level of understanding - the stereoselectivity of each reaction. In one case (Cram's rule) the evolution of the current theory is detailed from its first tentative (1952) postulate to the current Felkin-Anh-Heathcock formalism. For other reactions, only the currently accepted rationale is presented. Examination of these rationales also exposes the weaknesses of current theories, in that they cannot always explain the experimental observations. These shortcomings provide a challenge for future mechanistic investigations.

Available in PDF, EPUB and Kindle. Book excerpt: Asymmetric synthesis remains a challenge to practicing scientists as the need for enantiomerically pure or enriched compounds continues to increase. Over the last decade, a large amount of literature has been published in this field. Principles and Applications of Asymmetric Synthesis consolidates and evaluates the most useful methodologies into a one-volume resource for the convenience of practicing scientists and students. Authored by internationally renowned scientists in the field, this reliable reference covers more than 450 reactions and includes important stoichiometric as well as catalytic asymmetric reactions. The first chapter reviews the basic principles, common nomenclature, and analytical methods, and the remainder of the book is organized according to reaction type. The text examines such topics as: Carbon-carbon bond formations involving carbonyls, enamines, imines, and enolates Asymmetric C-O bond formations including epoxidation, dihydroxylation, and aminohydroxylation Asymmetric synthesis using the Diels-Alder reaction and other cyclizations Applications to the total synthesis of natural products Use of enzymes in asymmetric synthesis Practicing chemists in the pharmaceutical, fine chemical, and agricultural professions as well as graduate students will find that Principles and Applications of Asymmetric Synthesis affords comprehensive and current coverage.

Book excerpt: This seminal series, first edited by Ernest Eliel, responsible for some of the major advances in stereochemistry and the winner of the ACS Priestley Medal in 1996, provides coverage of the major developments of the field of stereochemistry. The scope of this series is broadly defined to encompass all fields of chemical and biological sciences that are founded on molecular and supramolecular interactions. Insofar as chemical, physical, and biological properties are determined by molecular shape and structure, the importance of stereochemistry is fundamental to and consequential for all natural sciences. Topics in Stereochemistry serves as a multidisciplinary series that enriches all of chemistry. Aimed at advanced students, university professors and teachers as well as researchers in pharmaceutical, agricultural, biotechnological, polymer, materials, and fine chemical industries, Topics in Stereochemistry publishes definitive and scholarly reviews in stereochemistry and has long been recognized as the gold standard reference work in this field. Covering the effect of chirality on all aspects of molecular interaction from the fundamental physical chemical properties of molecules and their molecular physics to the application of chirality in new areas such as its applications in materials science, Topics in Stereochemistry explores a wide variety of properties, both physical and chemical of isomers with a view to their applications in a number of disciplines from biochemistry to materials science.
