Book excerpt: This 1999 edition of The Neural Crest contains comprehensive information about the neural crest, a structure unique to the vertebrate embryo, which has only a transient existence in early embryonic life. The ontogeny of the neural crest embodies the most important issues in developmental biology, as the neural crest is considered to have played a crucial role in evolution of the vertebrate phylum. Data that analyse neural crest ontogeny in murine and zebrafish embryos have been included in this revision. This revised edition also takes advantage of recent advances in our understanding of markers of neural crest cell subpopulations, and a full chapter is now devoted to cell lineage analysis. The major research breakthrough since the first edition has been the introduction of molecular biology to neural crest research, enabling an elucidation of many molecular mechanisms of neural crest development. This book is essential reading for students and researchers in developmental biology, cell biology, and neuroscience.

Book excerpt: The craniofacial complex encases the brain along with other organs for vital functions of the body, such as taste, mastication, vision, smell, breathing, hearing and balance. The fusion, patterning, outgrowth and shaping tissues with heterogeneous developmental origins are necessary for craniofacial development. Mesenchymal stem cells (MSCs) have the potential to be used in the repair of craniofacial tissues, because these cells are adept at changing into cartilage, tendon, bone, muscle and fat. Craniofacial bone reconstruction is significant when it comes to craniofacial repairs, because it provides support to nearby soft tissues. Furthermore, it provides anchorage for dental structures which guide facial appearance, as well as structural stability. The regeneration of craniofacial tissue and organs is possible using bio-ink along with various stem cells and biomaterials in 3D bio printers. This book unravels the recent studies on craniofacial tissues and organs. It aims to shed light on the processes of repair and regeneration of these tissues and organs. This book will help new researchers by foregrounding their knowledge in this area of study.

Book excerpt: Mineralized Tissues in Oral and Craniofacial Science is a major comprehensive update on knowledge in the field of mineralized tissues in the oral and craniofacial region. Drs. McCauley and Somerman assembled an international team of researchers and clinicians, offering a global perspective on the current knowledge in this field. Basic and clinical correlates reinforce the significance of research to clinical diagnoses and therapies, written in a manner that lends easily to their use for case study teaching venues. Section 1 features the many aspects of bone in the craniofacial region, including embryology, cell biology, and stem cell biology. Section 2 focuses on teeth-tooth development, dentin, enamel, cementum, and tooth regeneration. Section 3 discusses the interaction between bones and teeth, including those associated with inflammatory processes, periodontal ligaments, biomechanics, and other impact factors-such as nutrition, metabolic bone diseases and therapeutic modalities. The novel approach of linking the basic principles of the cell and molecular biology of hard tissues to clinical correlates will appeal to readers at all levels of their research careers, both students and faculty; faculty interested in a comprehensive text for reference; and clinicians interested in the biologic aspects of bones and teeth.

Conventional treatment strategy includes the use of autogenous or allogeneic bone, biomaterials, and osteogenic growth factors. However, there has been no effective therapy for most cases so far. Stem cell-based gene therapy is the latest research method with possible applications in humans. The present study aims to (1) characterize rabbit mesenchymal stem cells (MSCs) relating to growth pattern, surface antigens, and the potential for multi-differentiation; (2) determine the transduction efficiency and duration of recombinant adeno-associated virus2 carrying enhanced green fluorescent protein (rAAV2EGFP) reporter gene in rabbit MSCs and study the effects of rAAV2EGFP transduction on stem cells' phenotype and capacity of multi-differentiation; (3) evaluate the differentiation characteristics of rabbit MSCs following recombinant adeno-associated virus 2 carrying bone morphogenetic protein 2 gene (rAAV2BMP2) transduction; (4) investigate whether MSCs transduced by rAAV2BMP2 could successfully induce bone regeneration in rabbit critical-size cranial defects. MSCs were isolated from bone marrows of rabbit tibias and cultured. Cell counting and colony-forming assays demonstrated that growth rates of MSCs dropped substantially with increasing passages. Flow cytometry on MSCs at passage 1 showed that cells expressed high level of CD49a and low level of CD44 as well as stage-specific embryonic antigen 4 (SSEA4). Multi-differentiation and reverse transcriptase-polymerase chain reaction (RTPCR) tests demonstrated that rabbit MSCs were capable to differentiate into osteocytes, chondrocytes and adipocytes. Immunofluorescence microscopy showed that rabbit MSCs produced a series of hematopoietic growth factors, including stem cell factor (SCF), vascular endothelial growth factor-A (VEGFA) and granulocyte macrophage colony-stimulating factor (GMCSF). Subsequently, rabbit MSCs were transduced with rAAV2EGFP in vitro. By comparing the transduction efficiency with different doses of rAAV2EGFP particles, multiplicity of infection (MOI) of 1 x 10 4 was identified as an optimal parameter for the transduction of rAAV2 in rabbit MSCs. Fluorescent microscopy demonstrated long-term expression of EGFP in rabbit MSCs after transduction both in vitro and in vivo. In addition, cell proliferation assay, adipogenic induction test and flow cytometry showed that rAAV2EGFP transduced MSCs exhibited a similar pattern with non-transduced cells on the cell growth, capacity of adipogenic differentiation and expression of surface antigens, indicating that rabbit MSCs maintain their stem cell properties after rAAV2EGFP transduction. Book excerpt: Dental caries, periodontitis, tooth loss, and bone resorption are considered prevalent health problems that have direct affect on the quality of life. While, advances in stem cell biology and biotechnology have sparked hope for devastating maladies, such as diabetes, cardiovascular diseases, etc., it also provides a strategy of regenerative therapy for dental tissues. From the prospective of tissue engineering, it is of utmost importance to understand and emulate the complex cell interactions that make up a tissue or organ. Unlike other tissues in the body, dental tissues are unique in their development, function, and even in their maintenance throughout life. The harmonized stimulations of biology and mechanical regulators to promote cellular activities have matured our understanding of the value of regenerative therapy of dental tissue versus the reparative treatment. In this book, we review the current knowledge available to regenerate alveolar bone, periodontal structure, and pulp/dentin complex. The book provides researchers with detailed information about development and functional characteristics of the dental unit with detailed protocols covering a comprehensive range of various approaches to engineer dental tissues: to use isolated cells or cell substitutes as cellular replacement, to use acellular biomaterials capable of inducing tissue regeneration, and/or to use a combination of cells, biomaterial and growth factors. We are well aware, with the concept changes in the field toward in-vitro biomimetics of in-vivo tissue development. The theoretical frame work integrating these concepts of developmental biology and developmental engineering is yet to be emphasized and implemented. Until this happens, we consider this book of regenerative dentistry as a call for scientists to achieve, researchers to innovate, practitioners to apply, and students to learn the art and science of regenerative therapy in dentistry.
