Public awareness of the dangers of heart disease has never been more pervasive. Now, though, ten years into a new millennium, scientists are gearing up for the next great challenges in tackling this pervasive condition. Cell therapy is going to be a key weapon in the fight against heart disease. It has the potential to address many cardiovascular conditions. From heart failure to atrioventricular nodal dysfunction, the young but promising field of cell therapy is set to play a significant role in developing the cures that the upcoming decades of hard work will yield. Regenerating the Heart: Stem Cells and the Cardiovascular System organizes the field into a digestible body of knowledge. Its four sections cover mechanical regeneration, electrical regeneration, cardiac tissues and in vivo stem cell therapies. An array of talented researchers share the fruits of their labors, with chapters covering such crucial issues as the cardiogenic potential of varying stem cell types, the ways in which they might be used to tackle arrhythmias, their possible application to biological replacements for cardiac tissues such as valves, and the varying approaches used in the in vivo evaluation of stem cell therapies, including methods of delivering stem cells to the myocardium. This comprehensive survey of an area of research with such exciting potential is an invaluable resource both for veteran stem cell researchers who need to monitor fresh developments, and for newly minted investigators seeking inspirational examples.

Only one-third of these heart attack victims recover completely. The remainder suffer the consequences of myocardial infarction and its ill fated remodeling process, resulting in chronic congestive heart failure. This malady alone is the leading cause of hospital admissions in the United States. New breakthroughs in stem cell therapy and tissue engineering have promised to reverse this dismal outcome by cardiovascular repair. World authorities, including scientists and regulatory authorities, have joined in a collaborative effort to present for the reader the first collective review of stem cell therapy for the treatment of cardiovascular disease. These contributions in basic science, pre-clinical and clinical experience guided by the regulatory pathways, assure a rapid course of translational research and clinical trials. The contents of this publication will become a prerequisite for those preparing to meet the challenges of this exciting and potentially rewarding field of stem cell research.

Book excerpt: Over the past 5 years there has been great excitement and controversy in the scientific, financial, and lay literature for the potential of stem cell-based strategies for the prev- tion and treatment of chronic heart failure (CHF). Not that long ago we believed we were born with a set number of cardiac myocytes and that once damaged there was no hope to replace them. The interest in the field stems from the magnitude of cardiovascular disease in the world. Our ability to treat and help patients survive acute myocardial infarction (MI) has resulted in a near epidemic of CHF. There are more than 5 million Americans who currently carry the diagnosis of CHF. With more than 1 million MIs a year in the United States, there are approx 500,000 new cases of CHF diagnosed each year. The goal of Stem Cells and Myocardial Regeneration is to present, in a coherent manner, the current state of knowledge of stem cell-based therapies for cardiac dysfunction, including current findings in both the laboratory and the clinic trials. The first section of this Stem Cells and Myocardial Regeneration focuses on the magnitude of the problem and the successes and failures of what we consider optimal medical therapy. It is on this background that stem cell-based therapy needs to build.

Available in PDF, EPUB and Kindle. Book excerpt: There is hardly an area of research developing so quickly and raising so many promises as stem cell research. Adult, embryonic and recently available induced pluripotent stem cells not only foster our understanding of differentiation of endo-, ecto- and mesodermal lineages to all organs of the body, but foremost nourish the hope that cells grown in culture can be used for regeneration of diseased organs such as the heart damaged by myocardial infarction. This book focuses on perspectives of stem cells for regenerative therapy of cardiovascular diseases. Based on the EC consortium INELPY, it reviews the field and disseminates major outcomes of this project. Thus it introduces the reader to this fascinating area of research and incorporates very recent findings interesting to the expert, spanning the field from bench to bedside. The compilation of contributions is unique as there is yet no similar comprehensive overview combining stem cell research with preclinical and clinical evaluation as well as engineering of tissue patches for transplantation. As such it will be an invaluable source of information for all researchers in the stem cell and tissue regeneration field including bioengineers as well as for all clinicians interested in regenerative therapies, especially for ischemic cardiomyopathies.

Available in PDF, EPUB and Kindle. Book excerpt: Over the past decade, significant efforts have been made to develop stem cell-based therapies for difficult to treat diseases. Multipotent mesenchymal stromal cells, also referred to as mesenchymal stem cells (MSCs), appear to hold great promise in regards to a regenerative cell-based therapy for the treatment of these diseases. Currently, more than 200 clinical trials are underway worldwide exploring the use of MSCs for the treatment of a wide range of disorders including bone, cartilage and tendon damage, myocardial infarction, graft-versus-host disease, Crohn’s disease, diabetes, multiple sclerosis, critical limb ischemia and many others. MSCs were first identified by Friendenstein and colleagues as an adherent stromal cell population within the bone marrow with the ability to form clonogenic colonies in vitro. In regards to the basic biology associated with MSCs, there has been tremendous progress towards understanding this cell population’s phenotype and function from a range of tissue sources. Despite enormous progress and an overall increased understanding of MSCs at the molecular and cellular level, several critical questions remain to be answered in regards to the use of these cells in therapeutic applications. Clinically, both autologous and allogenic approaches for the transplantation of MSCs are being explored. Several of the processing steps needed for the clinical application of MSCs, including isolation from various tissues, scalable in vitro expansion, cell banking, dose preparation, quality control parameters, delivery methods and numerous others are being extensively studied. Despite a significant number of ongoing clinical trials, none of the current therapeutic approaches have, at this point, become a standard of care treatment. Although exceptionally promising, the clinical translation of MSC-based therapies is still a work in progress. The extensive number of ongoing clinical trials is expected to provide a clearer path forward for the realization and implementation of MSCs in regenerative medicine. Towards this end, reviews of current clinical trial results and discussions of relevant topics association with the clinical application of MSCs are compiled in this book from some of the leading researchers in this exciting and rapidly advancing field. Although not absolutely all-inclusive, we hope the chapters within this book can promote and enable a better understanding of the translation of MSCs from bench-to-bedside and inspire researchers to further explore this promising and quickly evolving field.

Available in PDF, EPUB and Kindle. Book excerpt: Vascular diseases are the leading cause of death worldwide. Distinguished clinical and surgical approaches have attempted to overcome its morbidity and mortality; still 17.9 million people die every year due to vascular affections. Stem cell therapy has emerged as a promising therapeutic strategy. Stem cells synthesize and secrete cytokines that promote cell recruitment, immunomodulation, extracellular matrix remodeling, angiogenesis, and neuroregeneration, all of which promote regeneration. Besides that, stem cells are also capable of differentiating in various cell types, being employed in tissue engineering. Preclinical and clinical investigations have reported efficacy of stem cell therapy for various vascular diseases. Even though results are encouraging, the studies demonstrate variation in stem cell type and origin, route and protocol for administration, and concomitant use of other treatment strategies, impairing easy interpretation of results and clinical application. The purpose of this book is to compile and comprise the current state of the evidence regarding stem cell therapy for each vascular disease, elucidating possible clinical applications. More than an objective guide for readers on the use of this novel treatment strategy, this publication will advocate for stem cell therapy use and development and will be of significant interest to physicians in a wide range of disciplines as well as researchers.

Book excerpt: The second edition of Stem Cells: Scientific Facts and Fiction provides the non-stem cell expert with an understandable review of the history, current state of affairs, and facts and fiction of the promises of stem cells. Building on success of its award-winning preceding edition, the second edition features new chapters on embryonic and iPS cells and stem cells in veterinary science and medicine. It contains major revisions on cancer stem cells to include new culture models, additional interviews with leaders in progenitor cells, engineered eye tissue, and xeno organs from stem cells, as well as new information on "organs on chips" and adult progenitor cells. In the past decades our understanding of stem cell biology has increased tremendously. Many types of stem cells have been discovered in tissues that everyone presumed were unable to regenerate in adults, the heart and the brain in particular. There is vast interest in stem cells from biologists and clinicians who see the potential for regenerative medicine and future treatments for chronic diseases like Parkinson's, diabetes, and spinal cord lesions, based on the use of stem cells; and from entrepreneurs in biotechnology who expect new commercial applications ranging from drug discovery to transplantation therapies. Explains in straightforward, non-specialist language the basic biology of stem cells and their applications in modern medicine and future therapy Includes extensive coverage of adult and embryonic stem cells both historically and in contemporary practice Richly illustrated to assist in understanding how research is done and the current hurdles to clinical practice
