Available in PDF, EPUB and Kindle. Book excerpt: This volume focuses on the cell biology and physiology of skeletal muscle regeneration. This Book is a collection of classic and cutting edge protocols optimized for mice, but in most cases adaptable to rat or other mammalian models, that will allow an investigator to develop and implement a research study on skeletal muscle regeneration. Chapters address the three major areas of study: provoking regeneration by inducing damage to muscle, analyzing the progenitor cells of skeletal muscle, and quantifying overall muscle function. Subjects discussed include: inducing skeletal muscle injury by eccentric contraction; volumetric muscle loss; single myofiber isolation and culture; satellite cell transplantation; muscle clearing for whole mount immunostaining; luciferase tracking of muscle stem cells; mitochondrial and mitophagy flux analysis; in vivo assessment of muscle contractility; force measurements on single isolated myofibers; and analysis of aerobic respiration in intact skeletal muscle tissue by microplate respirometry. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to each respective topic, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Cutting edge and practical, Skeletal Muscle Regeneration in the Mouse: Methods and Protocols is an essential laboratory reference for research in skeletal muscle growth, damage, repair, degeneration, and regenerative therapy in the mouse model system.

Book excerpt: The book is a collection of original research and review articles addressing the intriguing field of the cellular and molecular players involved in muscle homeostasis and regeneration. One of the most ambitious aspirations of modern medical science is the possibility of regenerating any damaged part of the body, including skeletal muscle. This desire has prompted clinicians and researchers to search for innovative technologies aimed at replacing organs and tissues that are compromised. In this context, the papers, collected in this book, addressing a specific aspects of muscle homeostasis and regeneration under physiopathologic conditions, will help us to better understand the underlying mechanisms of muscle healing and will help to design more appropriate therapeutic approaches to improve muscle regeneration and to counteract muscle diseases.

Available in PDF, EPUB and Kindle. Book excerpt: It was a long and troublesome history but an inspiring and excltmg one too-from the time of Zielonko, a Russian pathologist who in 1874 first transplanted muscles, to the present day microneurovascular muscle graft. Today we know that for functional recovery of muscle tissue vascularisation and neurotisation are of equal importance. With the establishment of microsurgery of small vessels and nerves, its application for myopias tic operations has gained increasing interest. It was my intention to bring together leading authorities from all over the world in the field of muscle transplantation. Open questions and unsolved problems had to be discussed between research workers in the basic medical sciences and our clinical colleagues; in this way progress can best be accomplished. This publication is the result of the "Vienna Muscle Symposium" (June 13-14, 1980). It gives an up-to-date viewpoint and should act as a stimulus to all who are interested in this particular field. I wish to express my sincere thanks to those who have contributed to this publication, to the two co-editors J. Holle and B. M. Carlson for their effective cooperation, and to Springer-Verlag Wien for their prompt and most efficient work.

Available in PDF, EPUB and Kindle. Book excerpt: The world is faced with an epidemic of metabolic diseases such as obesity and type 2 diabetes. This is due to changes in dietary habits and the decrease in physical activity. Exercise is usually part of the prescription, the first line of defense, to prevent or treat metabolic disorders. However, we are still learning how and why exercise provides metabolic benefits in human health. This open access volume focuses on the cellular and molecular pathways that link exercise, muscle biology, hormones and metabolism. This will include novel “myokines” that might act as new therapeutic agents in the future.

This book was released on 2016 with total page 466 pages. Available in PDF, EPUB and Kindle. Book excerpt: AIM: Sarocpenic obesity is a national concern within the United States because this metabolic syndrome is tied with reduced mobility and quality of life. Both obesity and aging are associated with insulin-resistance, chronic low-grade inflammation and muscle weakness. Skeletal muscle regeneration is a process that involves the coordinated effort of myogenic regulatory factors (MRFs), inflammatory signaling, and extracellular matrix (ECM) remodeling for optimal regeneration. It has been demonstrated that obesity and aging have a reduction in muscle regeneration. It has not been examined if sarcopenic obesity will further reduce muscle mass and the regenerative process. The purpose of this study was to determine how sarcopenic obesity alters inflammatory signaling and extracellular remodeling in mouse skeletal muscle. METHODS: One hundred two male C57BL6/J mice (4 weeks old) were randomly assigned to either a high fat diet (HFD, 60% fat) or normal chow. Both young (3 months old) and aged mice (22-24 months old) were injected with either PBS or bupivacaine. Muscles were excised 3 or 21 days after injection. RESULTS: Mean cross-sectional area was reduced by 26% in aged HFD mice. The aged lean mice had 9% greater mean cross-sectional area 21 days following muscle damage but no changes were observed in aged HFD. Aged mice had nearly 2-fold collagen III content than young mice. P-STAT3/STAT3 was reduced by 82% in the aged HFD mice in the TA. NF-mB was 3-5 fold greater in the aged HFD mice compared to the aged lean mice in the gastrocnemius and the TA. There was a main effect of injury to reduce collagen I and III content in aged injured, and a main effect of diet with a decrease of collagen I, collagen III, and MMP-9 gene expression in aged HFD mice 3 days following muscle damage. CONCLUSION: Sarcopenic obese mice have reduced cross-sectional area than aged lean mice along with increased collagen III content and basal alterations in MyoD, ECM proteins, and inflammation gene expression. During skeletal muscle regeneration sarocopenic obese mice had dysregulated inflammatory signaling and reduced ECM gene expression. These data suggest further impairment in the regenerative process in sarcopenic obese mice.

Book excerpt: The search for knowledge on cellular and molecular mechanisms involved in skeletal muscle mass homeostasis and regeneration is an exciting scientific area and extremely important to develop therapeutic strategies for neuromuscular disorders and conditions related to muscle wasting. The mechanisms involved in the regulation of skeletal muscle mass and regeneration consist of molecular signaling pathways modulating protein synthesis and degradation, bioenergetics alterations and preserved function of muscle stem cells. In the last years, different kinds of stem cells has been reported to be localized into skeletal muscle (satellite cells, mesoangioblasts, progenitor interstitial cells and others) or migrate from non-muscle sites, such as bone marrow, to muscle tissue in response to injury. In addition, myogenic progenitor cells are also activated in skeletal muscle wasting disorders. The goal of this research topic is to highlight the available knowledge regarding skeletal muscle and stem cell biology in the context of both physiological and pathological conditions. Our purpose herein is to facilitate better dissemination of research into skeletal muscle physiology field.
