Book excerpt: Robust auditory perception plays a pivotal function in processing behaviorally relevant sounds, particularly when there are auditory distractions from the environment. The neuronal coding enabling this ability, however, is still not well understood. In this study we recorded single-unit activity from the primary auditory cortex of alert common marmoset monkeys (Callithrix jacchus) while delivering conspecific vocalizations degraded by two different background noises: broadband white noise (WGN) and vocalization babble (Babble).Noise effects on single-unit neural representation of target vocalizations were quantified by measuring the response similarity elicited by natural vocalizations as a function of signal-to-noise ratio (SNR). Four consistent response classes (robust, balanced, insensitive, and brittle) were found under both noise conditions, with an average of about two-thirds of the neurons changing their response class when encountering different noises. These results indicate that the distortion induced by one particular masking background in single-unit responses is not necessarily predictable from that induced by another, which further suggests the low likelihood of a unique group of noise-invariant neurons across different background conditions in the primary auditory cortex. In addition, for a relatively large fraction of neurons, strong synchronized responses can be elicited by white noise alone, countering the conventional wisdom that white noise elicits relatively few temporally aligned spikes in higher auditory regions.The variable single-unit responses yet consistent population responses imply that the primate primary auditory cortex performs scene analysis predominately at the population level. Next, by pooling all single units together, pseudo-population analysis was implemented to gain more insight on how individual neurons work together to encode and discriminate vocalizations at various intensities and SNR levels. Population response variability with respect to time was found to synchronize well with the stimulus-driven firing rate of vocalizations at multiple intensities in a negative way. A much weaker trend was observed for vocalizations in noise. By applying dimensionality reduction techniques to the pooled single neuron responses, we were able to visualize the dynamics of neural ensemble responses to vocalizations in noise as trajectories in low-dimensional space. The resulting trajectories showed a clear separation between neural responses to vocalizations and WGN, while trajectories of neural responses to vocalization and Babble were much closer to each other together. Discrimination of neural populations evaluated by neural response classifiers revealed that a finer optimal temporal resolution and longer time scale of temporal dynamics were needed for vocalizations in noise than vocalizations at multiple different intensities. Last, among the whole population, a subpopulation of neurons yielded optimal discrimination performance.Together, for different background noises, the results in this dissertation provide evidence for heterogeneous responses on the individual neuron level, and for consistent response properties on the population level.

Book excerpt: There seems little doubt that from the earliest evolutionary beginnings, inhibition has been a fundamental feature of neuronal circuits - even the simplest life forms sense and interact with their environment, orienting or approaching positive stimuli while avoiding aversive stimuli. This requires internal signals that both drive and suppress behavior. Traditional descriptions of inhibition sometimes limit its role to the suppression of action potential generation. This view fails to capture the vast breadth of inhibitory function now known to exist in neural circuits. A modern perspective on inhibitory signaling comprises a multitude of mechanisms. For example, inhibition can act via a shunting mechanism to speed the membrane time constant and reduce synaptic integration time. It can act via G-protein coupled receptors to initiate second messenger cascades that influence synaptic strength. Inhibition contributes to rhythm generation and can even activate ion channels that mediate inward currents to drive action potential generation. Inhibition also appears to play a role in shaping the properties of neural circuitry over longer time scales. Experience-dependent synaptic plasticity in developing and mature neural circuits underlies behavioral memory and has been intensively studied over the past decade. At excitatory synapses, adjustments of synaptic efficacy are regulated predominantly by changes in the number and function of postsynaptic glutamate receptors. There is, however, increasing evidence for inhibitory modulation of target neuron excitability playing key roles in experience-dependent plasticity. One reason for our limited knowledge about plasticity at inhibitory synapses is that in most circuits, neurons receive convergent inputs from disparate sources. This problem can be overcome by investigating inhibitory circuits in a system with well-defined inhibitory nuclei and projections, each with a known computational function. Compared to other sensory systems, the auditory system has evolved a large number of subthalamic nuclei each devoted to processing distinct features of sound stimuli. This information once extracted is then re-assembled to form the percept the acoustic world around us. The well-understood function of many of these auditory nuclei has enhanced our understanding of inhibition's role in shaping their responses from easily distinguished inhibitory inputs. In particular, neurons devoted to processing the location of sound sources receive a complement of discrete inputs for which in vivo activity and function are well understood. Investigation of these areas has led to significant advances in understanding the development, physiology, and mechanistic underpinnings of inhibition that apply broadly to neuroscience. In this series of papers, we provide an authoritative resource for those interested in exploring the variety of inhibitory circuits and their function in auditory processing. We present original research and focused reviews touching on development, plasticity, anatomy, and evolution of inhibitory circuitry. We hope our readers will find these papers valuable and inspirational to their own research endeavors.

Book excerpt: To understand the neural representation of broadband, dynamic sounds in Primary Auditory Cortex (AI), we characterize responses using the Spectro- Temporal Response Field (STRF). The STRF describes and predicts the linear response of neurons to sounds with rich spectro-temporal envelopes. It is calculated here from the responses to elementary 'ripples, ' a family of sounds with drifting, sinusoidal, spectral envelopes - the complex spectrotemporal envelope of any broadband, dynamic sound can expressed as the linear sum of individual ripples. The collection of responses to all elementary ripples is the spectro-temporal transfer function. Previous experiments using ripples with downward drifting spectra suggested that the transfer function is separable, i.e., it is reducible into a product of purely temporal and purely spectral functions. Here we compare the responses to upward and downward drifting ripples, assuming separability within each direction, to determine if the total bi-directional transfer function is fully separable. In general, the combined transfer function for two directions is not symmetric, and hence units in AI are not, in general, fully separable. Consequently, many AI units have complex response properties such as sensitivity to direction of motion, though most inseparable units are not strongly directionally selective. We show that for most neurons the lack of full separability stems from differences between the upward and downward spectral cross-sections, not from the temporal cross-sections; this places strong constraints on the neural inputs of these AI units.
