Book excerpt: Each generation in a sexually reproducing organism such as a fly or a mouse passes through the bottleneck of meiosis, which is the specialized cell division that gives rise to haploid reproductive cells (sperm, eggs, spores, etc. ). The principal function of meiosis is to reduce the genome complement by half, which is accomplished through sequential execution of one round of DNA replication followed by two rounds of chromosome segregation. Within the extended prophase between DNA replication and the first meiotic division in most organisms, homologous maternal and paternal chromosomes pair with one another and undergo homologous recombination, which establishes physical connections that link the homologous chromosomes until the time they are separated at anaphase I. Recombination also serves to increase genetic diversity from one generation to the next by breaking up linkage groups. The unique chromosome dynamics of meiosis have fascinated scientists for well over a century, but in recent years there has been an explosion of new information about how meiotic chromosomes pair, recombine, and are segregated. Progress has been driven by advances in three main areas: (1) genetic identification of meiosis-defective mutants and cloning of the genes involved; (2) development of direct physical assays for DNA intermediates and products of recombination; and (3) increasingly sophisticated cy- logical methods that describe chromosome behaviors and the spatial and temporal patterns by which specific proteins associate with meiotic chromosomes.

This was a very nice opportunity for us to learn about the scientific and cultural back ground of our Chinese friends. Many of them came from the interior of China and had had very little opportunity to become acquainted with people from a western culture. Living with these students was a very pleasant and educational experience which gave us a good pic ture of the cultural life and educational system of China at that time--an intimate picture that one normally would not see without travelling in that country.

Book excerpt: An early view of eukaryotic chromosomes was that of static structures, which stored DNA not in use within a given cell type. It was thought that packaging of DNA into higher levels of chromatin structure would suffice to repress gene expression and that the challenge to the cell would be to rescue specific sequences from these structures. The exten sive packaging of inactive DNA was considered the primary difference between eukaryotic and prokaryotic genomes and except for that point both would be similarly regulated by cis-acting sequences and trans acting factors. Our view of eukaryotic chromosomes has evolved dra matically over the last decade. The picture of chromosomes that is emerging is that of dynamic breathing organelles actively regulating the flow of genetic information from the genome. Indeed chromatin is so fluid that even maintaining gene quiescence is an active process and is tightly regulated. Chromatin dynamics is a consequence of protein complexes that modify histones, remove histone modifications, mobi lize nucleosomes or stabilize nucleosomes. Awide variety of such com plexes have now been described. Some are abundant and may play glo bal roles in chromosome fluidity and function. Others are more rare and specialized for specific functions at discreet loci. Moreover, several complexes share biochemical activities and genetic studies suggest overlapping functions in vivo. Many components of these complexes were first revealed in genetic screens, while others were discovered by novel cell biological or biochemical approaches.

Book excerpt: The field of cell cycle regulation is based on the observation that the life cycle of a cell progresses through several distinct phases, G1, M, S, and G2, occurring in a well-defined temporal order. Details of the mechanisms involved are rapidly emerging and appear extraordinarily complex. Furthermore, not only is the order of the phases important, but in normal eukaryotic cells one phase will not begin unless the prior phase is completed successfully. Che- point control mechanisms are essentially surveillance systems that monitor the events in each phase, and assure that the cell does not progress prematurely to the next phase. If conditions are such that the cell is not ready to progress—for example, because of incomplete DNA replication in S or DNA damage that may interfere with chromosome segregation in M—a transient delay in cell cycle progression will occur. Once the inducing event is properly handled— for example, DNA replication is no longer blocked or damaged DNA is repaired—cell cycle progression continues. Checkpoint controls have recently been the focus of intense study by investigators interested in mechanisms that regulate the cell cycle. Furthermore, the relationship between checkpoint c- trol and carcinogenesis has additionally enhanced interest in these cell cycle regulatory pathways. It is clear that cancer cells often lack these checkpoints and exhibit genomic instability as a result. Moreover, several tumor suppressor genes participate in checkpoint control, and alterations in these genes are as- ciated with genomic instability as well as the development of cancer.
