Book excerpt: The performance of an organism or organismic subsystem is the result of the interaction between the performance capacity of the system and Its environment. Environmental conditions can stress an organism and thus affect it's performance. In this study, three whole organism performances were examined: critical swimming speed, fatigue time and response time to a sudden bright light. In addition, subsystem performances were examined by measuring changes in hematocrit and plasma levels of cortisol, glucose, lactic acid, osmolarity, sodium and potassium. Performance tests were made on juvenile chinook salmon stressed 0, 1, 2 or 3 times, with 1 or 3 h between stresses, and on fish allowed to recover 1, 3, 6, 12 and 24 h after each level of stress. A stress consisted of holding the fish in a dip net in the air for 30 sec. The physiological responses and the swimming tests were conducted on salt water adapted fish while the behavioral response was measured with fish in fresh water. Plasma levels of cortisol, lactic acid, osmolarity and sodium increased cumulatively following several acute handling stresses spaced I h apart, though each parameter returned to control levels in 6-12 h. Plasma glucose rose significantly by 1 h after the first stress and remained higher than control levels at all levels of stress and through 24 h after stress. Plasma potassium increased initially following one and two stresses, dropped below control levels within 1-6 h after the last stress, and then increased above control levels for the remainder of the 24 h. Following three stresses potassium was lower than controls initially and then was similar to the levels for one and two stresses throughout the rest of the 24 h recovery period. There was a decrease in hematocrit 3-6 h after each level of stress followed by a return to control levels within 12 h of the last stress. Critical swimming speed was measured by increasing the water velocity in a flow-through swim tube and noting the velocity at which each fish stopped swimming. Critical swimming speeds after handling were highly variable and no differences were found between stressed fish and unstressed fish at any level of stress or any recovery time. Fatigue time was measured as the time a fish can maintain position in a swim tube at a given constant water velocity (60 cm/sec). Following each fatigue test, fish were killed and blood samples were obtained. Unlike unstressed fish, which all fatigued within13 min, the times to fatigue of stressed fish varied with some fish fatiguing within a few minutes and some fish swimming the 60 min period. There was a depression in fatigue times immediately following one and three handling stresses spaced 1 h apart. Immediately after two stresses and with all groups given time to recover from stress, fatigue times were similar to or higher than for unstressed fish. Plasma levels of cortisol, glucose, osmolarity and sodium were higher in swimming fish than in non-swimming controls. Plasma concentrations of cortisol, glucose and lactic acid were all highly variable in fish following fatigue and no differences were found betweeen fish handled in a dip net and unhandled fish at any level of stress or any time after stress. Plasma osmolarity and sodium levels in fatigued fish immediately after one stress were higher than levels in unstressed fatigued fish. Plasma potassium was higher in fatigued fish than in unstressed fatigued controls at several time periods after one and three stresses. The behavior test consisted of exposing groups of salmon in fresh water to a sudden bright light and measuring the time it took each fish to reach cover. Unstressed fish reached cover within 15 sec. Stressed fish took longer to reach cover, with the greatest delay immediately after stress and a gradual decrease in response time with recovery from stress. Exposure to two and three consecutive stress with 3 h between stresses increased the response times and the recovery times indicating that the effects of stress were cumulative.

American Shad ( Alosa sapidissima) are an anadromous clupeid introduced to the West Coast in the late 1800's with annual returns to the Columbia River Basin that are greater than salmon. Shad conveyance of marine derived nutrients results in a net input of phosphorus (P) and nitrogen (N) to the lower Columbia River. The quantity of N and P is similar to current, but far less than historical inputs by salmon. However, the overall input of nutrients by shad is small in comparison to background river-loading. Juvenile shad are abundant, feed on Daphnia, and are present near the end of the juvenile salmon seasonal outmigration in July and August. An empirical feeding study of juvenile salmon and shad indicated that salmon fed at a high rate on Daphnia in July and fed on juvenile shad in August. No juvenile shad were collected in July, but in August, shad primarily consumed cyclopoid copepods. Laboratory trials yielded a Type-II functional response curve: y = 29.858 x*(4.271 + x)-1 indicating that at Daphnia densities typical of those in the lower Columbia during August, Daphnia reductions have little effect on salmon consumption. Book excerpt: In this thesis, we examined the effects of the exposures to anthropogenic pollutants on the fish, primarily juvenile chinook salmon, immune system using newly and recently developed immune assays. In addition, we developed a new assay for measuring immunocompetence of fish. In the first chapter, the Alamar Blue assay was developed to quantify the proliferation of chinook salmon (Oncorhynchus tshawytscha) leukocytes. Isolated splenic and pronephric leukocytes were stimulated with different concentration of mitogens (LPS, PWM, and ConA) for various incubation times. Optimum cell culture conditions (cell density, mitogen concentration, and incubation time) for the Alamar Blue assay were evaluated by comparison with flow cytometric analysis. The Alamar Blue dye was non-toxic for leukocytes, and the assay proved to be able to quantify the mitogenic responses using LPS, but PWM and ConA. In the second chapter, we determined the effects and mechanisms by which p, p'- DDE exposure might affect the immune system of chinook salmon (Oncorhynchus tshawytscha). Isolated salmon splenic and pronephric leucocytes were incubated with different concentrations of p, p'-DDE, and cell viability, induction of apoptosis, and mitogenic responses were measured by flow cytometry and Alamar Blue assay. p, p'- DDE significantly reduced cell viability and proliferation and increased apoptosis. The effect of p, p'-DDE on pronephric leukocytes was more severe than on splenic leukocytes, likely because pronephric leucocytes had a higher proportion of granulocytes, cells that appear more sensitive to p, p'-DDE. The effect of p, p'-DDE on leucocytes appeared to vary between developmental stages or season. The mitogenic response of leukocytes of chinook salmon exposed to p, p'-DDE in vivo exhibited a biphasic dose-response relationship. Only leukocytes isolated from salmon treated with 59 ppm p, p'-DDE had a significantly lower percentage of Ig+ blasting cells than controls. Our results support the theory that exposure to chemical contaminants could lead to an increase in disease susceptibility and mortality of fish due to immune suppression. In the third chapter, we evaluated the direct effects of in vitro exposures to tributyltin (TBT), widely used biocide, on the cell mediated immune system of chinook salmon (Oncorhynchus tshawytscha). Splenic and pronephric leukocytes isolated from juvenile chinook salmon were exposed for 6, 24, or 96 hr to a concentration range of 0.03 0.1 mg TBT 1−1 in cell cultures. Effects of TBT on cell viability, induction of apoptosis, and mitogenic responses were measured by flow cytometry. Splenic and pronephric leukocytes in the presence of TBT experienced a concentration-dependent decrease in the viability in cell cultures following the induction of apoptosis. In addition, pronephric lymphocytes exhibited a greater sensitivity to TBT exposure than pronephric granulocytes. The functional ability of splenic B-cells to undergo blastogenesis upon LPS stimulation was also significantly inhibited in the presence of 0.05, 0.07, or 0.10 mg 1−1 of TBT in the cell cultures. Flow cytometric assay with the fluorescent conjugated monoclonal antibody against salmon surface immunoglobulin was employed for the conclusive identification of B-cell in the chinook salmon leukocytes. Our findings suggest that adverse effects of TBT on the function or development of fish immune systems could lead to an increase in disease susceptibility and its subsequent ecological implications.

Book excerpt: The last major synthesis of our knowledge of fish migration and the underlying transport and guidance phenomena, both physical and biological, was "Fish Migration" published 16 years ago by F.R. Harden Jones (1968). That synthesis was based largely upon what could be gleaned by classical fishery-biology techni.ques, such as tagging and recapture studies, commercial fishing statistics, and netting and trapping studies. Despite the fact that Harden Jones also provided, with a good deal of thought and speculation, a theoretical basis for studying the various aspects of fish migration and migratory orientation, progress in this field has been, with a few excepti.ons, piecemeal and more disjointed than might have been expected. Thus we welcomed the approach from the NATO Marine Sciences Programme Panel and the encouragement from F.R. Harden Jones to develop a proprosal for, and ultimately to organize, a NATO Advanced Research Institute (ARI) on mechanisms of fish migration. Substantial progress had been made with descriptive, analytical and predictive approaches to fish migration since the appearance of "Fish ~ligration." Both because of the progress and the often conflicting results of research, we felt that the time was again right and the effort justified to synthesize and to critically assess our knowledge. Our ultimate aim was to identify the gains and shortcomings and to develop testable hypotheses for the next decade or two.
