Book excerpt: A gas-liquid chromatographic procedure was developed for the analysis of the essential amino acids for man using the N-trifluoroacetyl methyl ester derivative. The procedure requires 2-1/2 hours for preparation of the sample and gas chromatographic resolution of all the common protein amino acids. The method was applied to the analysis of standard mixtures of amino acids, a protein hydrolyzate of ribonuclease and the free amino acids in human blood plasma. (Author).

Book excerpt: Quantitation of Amino Acids and Amines by Chromatography: Methods and Protocols is intended to serve as a ready-to-use guide for the identification and quantification of amino acids and amines in various matrices, providing an overview on the theory and protocol of available methods. It presents chromatograms with exact elution programs enabling visual analysis and compares the advantages-disadvantages of various chromatographic techniques. In accordance with the chronological order of the development of chromatographic methods, different techniques are discussed: The possibilities of gas chromatography (GC), followed by those of the high performance liquid chromatography (HPLC) and the most recent techniques capillary electrophoresis (CE), capillary, electrochromatography (CEC).The characteristics of the given chromatographic procedure, relating to the topic in question, are classified according to the preliminary preparation/derivatization process(es), which means the simple methods, suitable for the analysis of the selected compound(s) in natural form, are followed by various derivatization proposals. Detailed protocols provide the reader with guidance in beginning tasks and on how to improve current methods. This book appeals to a wide audience and is recommended for those looking towards the wider reaches of identification and quantification of amino acids and amines.* Provides a systematic, and comprehensive summary of chromatographic techniques and derivatization processes* Compares advantages/disadvantages of various chromatographic techniques* Readers can undertake practical tasks using detailed protocols given in the book

Available in PDF, EPUB and Kindle. Book excerpt: Amino Acid Analysis (AAA) is an integral part of analytical biochemistry. In a relatively short time, the variety of AAA methods has evolved dramatically with more methods shifting to the use of mass spectrometry (MS) as a detection method. Another new aspect is miniaturization. However, most importantly, AAA in this day and age should be viewed in the context of Metabolomics as a part of Systems Biology. Amino Acid Analysis: Methods and Protocols presents a broad spectrum of all available methods allowing for readers to choose the method that most suits their particular laboratory set-up and analytical needs. In this volume, a reader can find chapters describing general as well as specific approaches to the sample preparation. A number of chapters describe specific applications of AAA in clinical chemistry as well as in food analysis, microbiology, marine biology, drug metabolism, even archeology. Separate chapters are devoted to the application of AAA for protein quantitation and chiral AAA. Written in the highly successful Methods in Molecular BiologyTM series format, chapters contain introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and notes on troubleshooting and avoiding known pitfalls. Authoritative and accessible, Amino Acid Analysis: Methods and Protocols provides crucial techniques that can be applied across multiple disciplines by anyone involved in biomedical research or life sciences.

There are currently very few volumes dedicated to the methodology - ployed by neurosclentlsts, and those that are available often seem either out of date or limited m scope. This series is about the methods most widely used by modern-day neurosclentists and 1s wrltten by their colleagues who are practicing experts. Volume 1 will be useful to all neurosclentists since It concerns those procedures used routinely across the widest range of subdisciplines. Collecting these general techniques together m a single volume strikes us not only as a service, but will no doubt prove of exceptIona utilitarian value as well. Volumes 2 and 3 - scribe all current procedures for the analyses of amines and their metabolltes and of ammo aads, respectively. These collections will clearly be of value to all neuroscientlsts working m or contemplating research m these fields. Similar reasons exist for Volume 4 on receptor binding techniques since experimental - tails are provided for all types of llgand-receptor bmdmg, including chapters on general prmciples, drug discovery and - velopment, and a most useful appendix on computer programs for Scatchard, nonlinear, and competitive displacement analyses. Volume 5 provides procedures for the assessment of enzymes - volved m biogenic amme synthesis and catabolism. Volumes in the NELJROMETHODS series will be useful to neurochemists, -pharmacologists, -physiologists, -anatomlsts, psychopharmacologists, psychlatnsts, neurologists, and chemists (organic, analytical, pharmaceutical, medicinal), in fact, everyone involved m the neurosclences, both basic and clinical.

Stricter control over additives in food and concern about the effects of contamination of food by industrial and agricultural chemicals are among the developments which are leading to an increasing emphasis on detailed and accurate analysis of food. However. analysis of food is required for many reasons, including detection of toxic components, monitoring legislation, detecting adulteration, formulation of controlled diets, controlling formulation during product development and detecting changes in food during storage and processing. Foods comprise a complex mixture of components and food analysis requires efficient methods of separation with high sensitivity or specificity of detection. Although many food components are involatile or thermally labile and therefore not suitable for analysis by gas chromatography, other components are volatile and this technique is the preferred analytical method. Developments in methods of derivati zation, injector design and column technology have also extended the applicability of gas chromatography to the analysis of relatively involatile compounds.

Book excerpt: During recent years there has been increasing interest in the value of a number of chemical and physical-chemical analytical methods for the detection and characterization of microorganisms. Furthermore, such methods are currently used in studies on microbial metabolic processes, on the role of microorganisms in the turnover of inorganic and organic compounds, and on the impact on environmental changes by microbial activity. Moreover, the introduction of some of these methods not only shortens the analytical time period compared to ·'traditional" techniques, but also improves the analytical quality. Mass spectrometry (MS) combined with chromatographic inlet systems, particularly gas chromatography (GC), belongs to those methods which during recent years have established their value for the above-mentioned purposes. The present volume starts with basic chapters on the principles for MS and common inlet systems, particulary Gc. It discusses applications of these techniques to a number of microbiological disciplines, e.g., ecologi cal and medical microbiology. Emphasis is laid on organic compound classes vii viii / PREFACE of special relevance to microbiology, e.g., volatiles, lipids, amino acids, peptides and carbohydrates. Some compound classes of a more general biochemical rather than specific microbiological importance, e.g., steroids and nucleotides, are dealt with briefly. The editors wish to thank all those who have contributed to this book. We hope it will stimulate further research in this futuristic field and will be of practical value.
