Book excerpt: Nitric oxide (NO) plays a key regulatory role in cardiovascular physiology due to its vasodilatory, anti-thrombotic and anti-atherogenic properties. In endothelial cells, the endothelial isoform of NO synthase (eNOS or NOS-3) catalyses the conversion of L-arginine to L-citrulline with the production of NO. NOS-3 activation is regulated by a number of factors, including Ca2+-calmodulin, phosphorylation of key residues, and interactions with associated proteins (including caveolin-l, and heat shock protein 90). The aims of this work were to clarify the roles of certain proteins, previously reported to associate with NOS-3, in NOS-3 regulation in human endothelial cells, as well as to identify novel protein associations with NOS-3 and to clarify the possible role of such associations . -- In human umbilical vein endothelial cells (HUVEC), I confirmed our previously reported finding that NOS-3 binds to the globular form of [beta]-actin (G-actin); of note, and not previously reported, we found that a wide variety of NOS-3 agonists (adenosine, salbutamol, histamine and thrombin) increased the degree ofNOS-3 / G-actin association, which was prevented by stabilization of [beta] -actin in its filamentous form (F-actin) using phalloidin. All agonists tested induced a marked increase in phosphorylation at Ser 1177 in NOS-3, as expected, but this was abolished by phalloidin pre-treatment. In studies of HEK293 cells transfected with either wild-type NOS-3 (wild type Ser 1177) or NOS-3 mutated at residue 1177 (1177 A or 1177D), I found that, whereas a marked association of [beta]-actin with NOS-3 was observed inlysates from cells transfected with wild-type NOS-3, the degree of this association was markedly attenuated III lysates isolated from cells transfected with either mutant form of NOS-3. -- I also demonstrated a novel interaction between NOS-3 and p-catenin in HUVEC.

Diverse and distinct auto-, para-, and endocrine stimuli arriving at the surface of endothelium, smooth muscle cells, cardiomyocytes and fibroblasts within the myocardium, engage cell type-specific receptors, which lead to transmission of signals across the cell plasma membrane and result in the production and activation of second messengers. The most common mechanism by which these second messengers function is via direct or indirect activation of specific protein kinases. The current challenge for scientists is to identify the specific substrates (e.g. metabolic enzymes, Ca2+-regulating proteins, transcription and mitotic factors) for the many protein kinases, to elucidate the biological significance of the cell type-specific expression heterogeneity of signalling proteins (e.g. membrane receptors, isoenzymes of protein kinase C, G-proteins) and to unravel the cross-talk interaction between the signalling systems (e.g. phospholipase C with adenylate cyclase and phospholipase C with phospholipase D). The multiplicity of receptor types, G-proteins, effector proteins, second messengers and protein kinases, their substrate proteins and the `cross-talk' interactions in the myocardium raises fundamental questions about the mechanisms that ensure the precision and timing of the myocardial responses to hormonal and pharmacological stimuli. This book provides an up-to-date source of information for all scientists and clinicians interested in the mechanisms by which external signals are transmitted to the interior and regulation of a variety of physiological, pathological and pharmacological responses.
