This book was released on 1998-05-15 with total page 578 pages. Available in PDF, EPUB and Kindle. Book excerpt: The third edition of this popular textbook builds on the excellent foundations laid down by the earlier editions. It provides a thorough introduction to the principles of rational drug design, adopting a 'from the bench to the market place' approach. As knowledge of biological systems has expanded and the number of techniques available for exploring and visualizing their components has increased, it has become possible to design drugs specifically for a given target. This unique insight has revolutionized the process of drug development for specific disease states, and in this textbook both novel and established approaches are incorporated. The introductory text explains the principles of drug design using real examples. These illustrate the discovery of 'lead' compounds and their manipulation to produce non-toxic drug candidates that will be successfully metabolized to interact with target receptors in a predicted fashion. In addition to fully updating the contents of the previous edition, the Editor has included important new sections on the pharmacological consequences of drug chirality, agonists and antagonists of neurotransmitters, and the process involved in proceeding from program sanction to clinical trials

Book excerpt: Since 1982, our ever-expanding group of investigators has been meeting in exotic parts of the world to discuss aspects of three enzyme systems. The 1996 meeting was no exception. Nearly 90 scientists from 15 countries met in the small city of Deadwood, South Dakota, for four days of stimulating talks and posters and incredible scenery. Once more this meeting reflected the changing trends in biochemical research. At the 1982 meeting most of the speakers discussed isolating new enzymes and trying to characterize them. At this meeting many speakers discussed interpretations of three-dimensional struc ture or regulatory elements of the genes controlling for the tissue-specific expression of the enzyme. Hopefully, readers will find the proceedings of the meeting to be of interest. Though they reflects the scientific information that was presented at the meeting, they do not indicate the level of personal interactions that went on during the meeting. Once again, the willingness of the participants to discuss unpublished data and to share thoughts about the future directions of their research helped make this, like our previous seven meetings, a special scientific experience for those who attended.

Book excerpt: The development, growth and survival of eukaryotic organisms require the proper regulation of tens of thousands of genes. Therefore, a correct temporal and spatial expression of genes is crucial. These expression patterns are controlled by a wide variety of mechanisms, but a major level of gene expression occurs at the transcription initiation process. In this part, the basic mechanisms of eukaryotic gene transcription will be discussed. Contents include: 1) General Introduction, 2) Characterization of the two coactivator interacting surfaces of the androgen receptor and their relative role in transcriptional control, 3) Screening for environmental (anti)androgenic activity, 4) Detection of endocrine disrupting activities in pesticides and polychlorinated biphenyls, and 5) General Discussion / Future Perspectives.

Since the discovery of the first microRNA (miRNA) called lin-4 on Caenorhabditis elegans in 1993, our understanding of gene expression has deepened. Further miRNA discoveries have led to the investigation of new ncRNA families and their function, biogenesis, and interactions with other types of molecules. Recent developments in bioinformatics, public databases, sequencing technologies, and biochemical techniques have increased our understanding of the biogenesis, function, and interactions of ncRNAs. This book provides an overview of current ncRNA research dealing with human diseases and cancer and plant miRNAs.

The amino-terminal domain (NTD) of the androgen receptor (AR) is indispensable for AR transactivation and contains a strong activation function 1. Its activity is affected by coregulators that influence a number of functional properties of AR. The NTD of the AR is 529 aa long and is a complex domain with several functions, namely p160 recruitment, interaction with the LBD, and it contains two transactivation functions Tau-1 And Tau-5. The aim of this study was to obtain a more detailed structure-function analysis of the NTD of the AR. Contents include: Introduction, N/C Interaction in AR-mediated Transactivation, Polyglutamine Stretch, SUMOylation of the AR, Activation Function of the hAR, Discussion, Summary and conclusions, Future Prospects."

Book excerpt: The steroid scaffold continues to be the structural basis of new drugs for a variety of targets and diseases. Indeed, steroids interact with enzymes and receptors in a strikingly specific manner. Chemistry and Biological Activity of Steroids aims to provide an updated overview of recent advances in the medicinal chemistry of steroids. Novel synthetic methods in the steroids field, including steroid biotransformations, new steroids able to tackle steroid receptors, and steroid enzymes with clinical relevance, are critically reviewed in this book. Furthermore, the diverse physiopathological roles of oxysterols and their therapeutic value are also discussed.

Available in PDF, EPUB and Kindle. Book excerpt: Synthesis of Essential Drugs describes methods of synthesis, activity and implementation of diversity of all drug types and classes. With over 2300 references, mainly patent, for the methods of synthesis for over 700 drugs, along with the most widespread synonyms for these drugs, this book fills the gap that exists in the literature of drug synthesis. It provides the kind of information that will be of interest to those who work, or plan to begin work, in the areas of biologically active compounds and the synthesis of medicinal drugs. This book presents the synthesis of various groups of drugs in an order similar to that traditionally presented in a pharmacology curriculum. This was done with a very specific goal in mind – to harmonize the chemical aspects with the pharmacology curriculum in a manner useful to chemists. Practically every chapter begins with an accepted brief definition and description of a particular group of drugs, proposes their classification, and briefly explains the present model of their action. This is followed by a detailed discussion of methods for their synthesis. Of the thousands of drugs existing on the pharmaceutical market, the book mainly covers generic drugs that are included in the WHO's Essential List of Drugs. For practically all of the 700+ drugs described in the book, references (around 2350) to the methods of their synthesis are given along with the most widespread synonyms. Synthesis of Essential Drugs is an excellent handbook for chemists, biochemists, medicinal chemists, pharmacists, pharmacologists, scientists, professionals, students, university libraries, researchers, medical doctors and students, and professionals working in medicinal chemistry. * Provides a brief description of methods of synthesis, activity and implementation of all drug types* Includes synonyms* Includes over 2300 references
