Book excerpt: Rearrangements with Migration to Electrophilic Heteroatomic Centers: Timeless Classics in Organic Synthesis covers the basic principles and synthetic applications of a critically important class of organic reactions that date back to the founding period of organic chemistry. These reactions are not only of historical and pedagogical interest, but also have great practical value to this day, being inevitable in any strategy concerning the introduction of nitrogen and oxygen functionality. The book is made up of three parts, dealing with the Beckmann reaction, the Hoffmann family of reactions and the Baeyer-Villiger reaction. The book strikes a balance between delineation and exhaustiveness, with up-to-date refences for readers requiring in-depth coverage. Relevant industrial applications, including pharmaceutical processes, are included. - Summarizes mechanistic background with an emphasis on synthetic applications - Includes up-to-date references that allow for locating original papers for experimental procedures - Provides applications in the synthesis of complex molecules and natural products to judge the scope and limitations of each reaction

Available in PDF, EPUB and Kindle. Book excerpt: Molecular rearrangements occupy center stage in the development of organic synthesis and, consequently, in the impressive achievements that have resulted in the total synthesis of many complex natural products in the last decades. The purpose of this chapter, in this volume of a book series dedicated to the Studies in Natural Product Chemistry, is to show a set of selected and organized molecular rearrangements based on a personal choice of expertise related to the synthesis of complex bioactive molecules. Our selection addresses [3,3]-sigmatropic rearrangements, which continue to provide excellent opportunities for reaching high chemical diversity, operating in efficient synthetic schemes. [3,3]-Sigmatropic rearrangements, such as the Cope and Claisen rearrangements, are among the most basic and useful transformations in synthetic organic chemistry. This is possibly due to the fact that these rearrangements are efficient methods for the synthesis of quaternary, sterically hindered chiral centers, and for the stereoselective formation of carbon–carbon and carbon–heteroatom bonds. In addition, [3,3]-sigmatropic rearrangements can be easily integrated in, and adapted to, cascade processes as a simple method to prepare complex molecules in atom-efficient reactions. Consequently, in the context of [3,3]-sigmatropic rearrangements, we focus on the transformations of propargylic esters, whose rearrangements, in different experimental conditions, have been largely exploited for the synthesis of interesting key and useful intermediates. The rearrangements can be promoted by protic acids, Lewis acids, and noble metals such as Pt, Au, or Cu salts, or complexes that have recently emerged as the most popular and potent electrophilic activators of alkynes toward a number of nucleophilic agents, under homogeneous conditions.

The synthesis of natural products, bioactive compounds, pharmaceuticals, and drugs is of fundamental interest in modern organic chemistry. New reagents and reaction methods towards these molecules are being constantly developed. By understanding the mechanisms involved and scope and limitations of each reaction applied, organic chemists can further improve existing reaction protocols and develop novel efficient synthetic routes towards frequently used drugs, such as Aspirin or Penicillin. Applied Organic Chemistry provides a summary of important (name) reactions and reagents applied in modern organic chemistry and drug synthesis. It covers rearrangement, condensation, olefination, metathesis, aromatic electrophilic substitutions, Pd-catalyzed C-C bond forming reactions, multi-component reactions, as well as oxidations and reductions. Each chapter is clearly structured, providing valuable information on reaction details, step-by-step mechanism, experimental procedures, applications, and (patent) references. By providing mechanistic information and representative experimental procedures, this book is an indispensable guide for researchers and professionals in organic chemistry, natural product synthesis, pharmaceutical, and medicinal chemistry, as well as post-graduates preparing themselves for a job in the pharmaceutical industry. Hot Topic: Reviews important classes of organic reactions (incl. name reactions) and reagents in medicinal chemistry. Useful: Provides information on reaction details, common reagents, and functional group transformations used to synthesize natural products, bioactive compounds, drugs, and pharmaceuticals, e.g. Aspirin, Penicillin. Unique: For every reaction the mechanism is explained step by step, and representative experimental procedures are given, unlike most books in this area. User-friendly: Chapters are clearly structured making it easy for the reader to compare different reactions. Applied Organic Chemistry is an indispensable guide for researchers and professionals in organic chemistry, natural product synthesis, pharmaceutical, and medicinal chemistry, as well as post-graduates preparing themselves for a job in the pharmaceutical industry.

Book excerpt: Synthetically useful organic reactions or reagents are often referred to by the name of the discoverer(s) or developer(s). Older name reactions are described in text books, but more recently developed synthetically useful reactions that may have been associated occasionally with a name are not always well known. For neither of the above are experimental procedures or references easy to find. In this monograph approximately 500 name reactions are included, of which over 200 represent newer name reactions and modern reagents. Each of these reactions are extremely useful for the contemporary organic chemistry researcher in industry or academic institutions. This book provides the information in an easily accessible form. In addition to seminal references and reviews, one or more examples for each name reaction are provided and a complete typical experimental procedure is included, to enable the student or researcher to immediately evaluate reaction conditions. Besides an alphabetical listing of reactions and reagents, cross references permit the organic practitioner to find those name reactions or reagents that enable specific transformations, such as, conversion of amines to nitriles, stereoselective reduction, fluoroalkylation, phenol alkynylation, asymmetric syntheses, allylic alkylation, nucleoside synthesis, cyclopentanation, hydrozirconation, to name a few. Emphasis has been placed on stereoselective and regioselective transformations as well as on enantioselective processes. The listing of reactions and reagents is supported by four indexes.

Book excerpt: Intended for students of intermediate organic chemistry, this text shows how to write a reasonable mechanism for an organic chemical transformation. The discussion is organized by types of mechanisms and the conditions under which the reaction is executed, rather than by the overall reaction as is the case in most textbooks. Each chapter discusses common mechanistic pathways and suggests practical tips for drawing them. Worked problems are included in the discussion of each mechanism, and "common error alerts" are scattered throughout the text to warn readers about pitfalls and misconceptions that bedevil students. Each chapter is capped by a large problem set.
