These proteins enhance the growth of cancer cells and protect tumors from treatments such as drugs or surgery. However, new drugs have recently been developed particularly those targeting heat shock protein 90. As heat shock protein 90 functions to stabilize many of the oncogenes and growth promoting proteins in cancer cells, such drugs have broad specificity in many types of cancer cell and offer the possibility of evading the development of resistance through point mutation or use of compensatory pathways. Heat shock proteins have a further property that makes them tempting targets in cancer immunotherapy. These proteins have the ability to induce an inflammatory response when released in tumors and to carry tumor antigens to antigen presenting cells. They have thus become important components of anticancer vaccines. Overall, heat shock proteins are important new targets in molecular cancer therapy and can be approached in a number of contrasting approaches to therapy.

Book excerpt: The hydrolyzable tannin, gallotannin (GT), is a polyphenol that possesses intere sting anti-carcinogenic properties in colon cancer. In the human colon cancer c ell lines HCT116 (p53+/+) and HCT116 (p53-/- ), GT induced cell cycle arrest and apoptosis, respectively. However, the mechanisms underlying these anti-carcinog enic effects are not well defined. The signal transducer and activator of transcription 3 (STAT3) is constitutively activated in HCT116 cell lines, where it promotes cell survival and proliferati on. In this study, we show that GT totally inhibited the phosphorylation of STA T3 and JAK2, an upstream regulator of STAT3 and reduced the phosphorylation leve ls of STAT1. GT downregulated the expression levels of STAT3, STAT1, and JAK2 p roteins. It also downregulated the downstream targets of p-STAT3, Bcl-xL, c-Myc . GT also decreased the protein levels of Bcl-2, another downstream target of p -STAT3, in the two human colon cancer cell lines, HCT116 (p53-/- ) and HCT116 (p2 1-/- ). Bcl-2 decrease led to a significant increase in the Bax/Bcl-2 ratio, ind icating induction of apoptosis. In contrast, in HCT116 (p53+/+) cells, Bcl-2 was not modulated. Apoptosis in HCT116 (p53-/- and p21-/- ) cells occurred in caspa se-independent manner and was associated with PARP cleavage. Moreover, the three cell lines exhibited differential sensitivity to GT, with HC T116 (p21-/- ) cells being most sensitive and HCT116 (p53+/+) cells that express p21 proteins, being least sensitive. In HCT116 (p53+/+) cells, GT induced senesc ence and a concomitant decrease in phosphorylated retinoblastoma (p-Rb) protein levels and increase in the active form of retinoblastoma (Rb). These effects we re not observed in the other two cell lines, suggesting a role for p21 in mediat ing senescence induction. At low concentrations, GT induced senescence in a STA T3-independent manner. GT-induced cell cycle arrest and apoptosis in the human colon cancer cell lines was associated with the production of reactive oxygen species (ROS). Increase i n ROS levels was associated with induction of HO-1 and p-ERK protein levels, whi ch suggests that these proteins are at least partially involved in mediating the oxidative effects of ROS.
