Book excerpt: Ultrasmall nanoparticles have seen growing interest in several fields such as medicine, optics, and catalysis due to their unique size-dependent properties. Among these are fluorescent core-shell silica nanoparticles synthesized in water known as Cornell prime dots (C' dots), which are composed of a silica core and a poly(ethylene glycol) (PEG) shell. These C' dots are poised to become promising diagnostic and therapeutic tools in medicine, particularly for cancer applications. While spherical C' dots have already been in FDA-approved human clinical trials, exploration has expanded into different topologies of silica nanoparticles such as rings and cages, providing higher aspect ratios and higher surface area to volume ratios. For clinical translation of such nanostructures, quantitative assessments of their size and size dispersity remain parameters for further investigation and are critical to understanding and evaluating their effectiveness and safety in nanomedicine applications. Here, solution small-angle X-ray scattering was employed to characterize 3D structural details of ring-type silica nanostructures and their size and size dispersity. A hollow cylinder model provided consistent structural and dispersity parameters that were supported by transmission electron microscopy investigations.

This book was released on 2011 with total page 156 pages. Available in PDF, EPUB and Kindle. Book excerpt: Ordered mesoporous silica materials are characterized by uniform and tunable pore size, high surface area and large pore volume. In particular, nano-sized ordered mesoporous silica particles have drawn interest from several fields, including biorelated areas, because silica is benign, possesses chemical stability and can be integrated with other materials. Structural aspects, such as pore connectivity, geometry and pore size are known to govern materials performance. Extensive efforts have been devoted to synthesize mesoporous silica particles with different structures, functionalities and sizes. In contrast, only a small number of studies so far have concentrated on the formation mechanism of these particles. This is hence the focus of the present dissertation. The first part reports on the synthesis and characterization of ordered mesoporous silica nanoparticles with and without embedded magnetic nanoparticles. The formation mechanism of silica nanocomposites is investigated by capturing particle formation at different time points during the synthesis. A combination of transmission electron microscopy (TEM) and small angle x-ray scattering (SAXS) is used to characterize the structure evolution of resulting materials. Incorporating organic moieties into the silica matrix provides additional functionalities to ordered mesoporous silica nanoparticles. However, it often leads to disordered pore structure or pore blockage. The second part demonstrates the preparation of aminated and ordered mesoporous silica nanoparticles using a cocondensation method. Increasing the amount of aminosilane in the synthesis feed causes a structural transition of organically modified particles from hexagonal to cubic. Pore size of ordered mesoporous silica and aminated ordered mesoporous silica nanoparticles can be tailored by the addition of a swelling agent during the synthesis. The structural transformation from hexagonal to cubic is also observed in the latter case, albeit at different amino silane concentrations. The final part reports on the internalization of nanoparticles into cells. Fluorescent aminated mesoporous silica nanoparticles are first prepared and then coated with poly(ethylene glycol) to improve particle stability and lower protein adsorption. Dye-labeled aminated mesoporous silica nanoparticles are spontaneously internalized by cells.

Book excerpt: This volume is based on lectures given at the NATO-Advanced Study Institute on Structure and Dynamics of Polymer and Colloid Systems held in Les Houches, France from September 14-24, 1999. The meeting arose from a perceived need to bring together scientists studying the polymer and colloid fields. Although these fields are intertwined and share many techniques (e. g. , light, neutron and x-ray scattering), it is remarkable how little the approaches and concepts used by the one field penetrate the other. For instance, the theory of spherical colloids is very highly developed and many of the concepts developed for these systems can be extended to those with non-spherical morphology, such as solutions of rigid rod polymers. In addition, mixtures of polymers and colloids, both in the bulk and at interfaces, are the basis for many industrial products. Methods are now rapidly being developed for understanding the structure and dynamics in polymer/colloid mixtures at the molecular level, but the point of view of the colloid scientist is often rather different from that of the polymer scientist. The NATO-ASI brought together polymer and colloid scientists, including many young researchers, who presented and discussed recent developments in these fields and the possibilities for cross-fertilization This volume contains articles on a wide variety of topics at the research forefront of the polymer and colloid fields by some of the world's foremost experts at a level accessible to graduate students, post-docs and researchers.

Book excerpt: Small-angle scattering of X rays and neutrons is a widely used diffraction method for studying the structure of matter. This method of elastic scattering is used in various branches of science and technology, includ ing condensed matter physics, molecular biology and biophysics, polymer science, and metallurgy. Many small-angle scattering studies are of value for pure science and practical applications. It is well known that the most general and informative method for investigating the spatial structure of matter is based on wave-diffraction phenomena. In diffraction experiments a primary beam of radiation influences a studied object, and the scattering pattern is analyzed. In principle, this analysis allows one to obtain information on the structure of a substance with a spatial resolution determined by the wavelength of the radiation. Diffraction methods are used for studying matter on all scales, from elementary particles to macro-objects. The use of X rays, neutrons, and electron beams, with wavelengths of about 1 A, permits the study of the condensed state of matter, solids and liquids, down to atomic resolution. Determination of the atomic structure of crystals, i.e., the arrangement of atoms in a unit cell, is an important example of this line of investigation.
