Initially a novel grafting-to procedure for the production of high molecular weight and high grafting density PEO brushes will be presented. This procedure will allow the accurate control of the grafting density of PEO brushes. The crystallisation and oxidation of PEO brushes is also briefly investigated. Ex situ ellipsometry experiments will show that the adsorbed amount of protein is related to the surface coverage of polymer. supporting the random sequential adsorption theory (RSA-model) of Katira et al. However. through in situ QCM-D experiments it will be shown that RSA-model cannot fully describe the protein adsorption kinetics and may need to be revised to take these observations into account. We will show through use of an exponential model that the adsorption of protein at PEO brushes is a site blocking process and not an entropic process on the time scales measured here. Neutron reflectivity experiments on the INTER reflectometer instrument at ISIS show that the lysozyme protein is most likely adsorbing in the primary position at the gold- brush interface. The experiments presented in this thesis support the conclusion that the protein resistive nature of PEO brushes is that of a site blocking barrier.

Book excerpt: The ultimate goal of this dissertation research is to enhance the protein resistant nature of poly(ethylene oxide) (PEO) or poly(ethylene glycol) by introduction of a siloxane linker and to subsequently prepare coatings which prevent surface-induced thrombosis. The hydrophobicity and flexibility of the siloxane tether should impart both amphiphilicity and conformational mobility to the PEO chain to further decrease protein adhesion. Because adsorption of plasma (blood) proteins initiates the clotting process, coating surfaces based on these new PEO-silanes should prevent or significantly diminish thrombosis. Thus, these coatings would be extremely useful for bloodcontacting medical devices such as stents, grafts, arteriorintravenous shunts, and biosensors. Novel amphiphilic PEO-silanes were prepared with systematic variations to several key structural features, including: siloxane tether length, PEO segment length, and PEO architecture. Thus, PEO-silanes were prepared having the general formulas: [alpha]-(EtO)3Si(CH2)2-oligodimethylsiloxanen-block-[PEO-OCH3] (n = 0, 4, and 13; linear architecture) and [alpha]-(EtO)3Si(CH2)2-oligodimethylsiloxanen-block-[PEOm-OCH3]2 (n = 0, 4, and 13; m = 6 and 12 branched architecture). The reactive triethoxysilane [(EtO)3Si-] group serves as the crosslinking or grafting moiety. The PEO segment is distanced from the (EtO)3Si- group by an oligodimethylsiloxane tether which is both hydrophobic and exhibits a high degree of chain flexibility. Crosslinked silicone coatings and surfacegrafted coatings were prepared with amphiphilic linear PEO-silanes (a-c). Crosslinked silicone coatings were also prepared with branched PEO-silanes (1a-3a and 1b-3b). All coatings showed improved resistance to common plasma proteins compared to silicone coatings. Furthermore, protein adsorption generally decreased with siloxane tether length. For crosslinked PEO-modified silicone coating systems based on linear (a-c) and branched PEO-silanes (1a-3a and 1b-3b), longer tethers enhanced PEO reorganization to the film-water interface to enhance protein resistance. In the absence of surface reorganization for surface grafted coatings prepared with linear PEO-silanes, longer siloxane tethers better inhibited protein adsorption despite a moderate decrease in graft density (sigma) and decrease in surface hydrophilicity. This indicates that longer siloxane tethers enhance the configurational mobility of the PEO segments to better repel proteins.

Book excerpt: Protein adsorption onto foreign material is the beginning of a biological process that results in induced thrombosis that leads to a response that may be detrimental to the body. The prevention of blood plasma protein adsorption is a major factor in promoting biocompatibility of biomaterials with blood plasma. The application of coating biomaterial surfaces with poly(ethylene oxide) (PEO) has been very popular in biocompatibility studies. Theoretical studies have created models that have based protein adsorption on surfaces coated with polymers on free energies (van der Waals, hydrophobic interactions, and steric repulsions). The random sequential adsorption (RSA) model is a theoretical model that postulates that randomly distributed polymers and their obstruction of adsorption sites is a major factor that affects the random placement of protein adsorption based on the availability of the remaining unobstructed adsorption sites. This work examines the simulation of how the arrangement of polymer layers prepared by self-assembled monolayers (SAM) and physical adsorption affects the diffusion of the arrangement of proteins on the surface based on the RSA model. The model in this work extends the RSA model by considering the exact arrangement of polymers on the surface as well as dynamic rearrangements of adsorbed proteins that lead to a more closely packed layer. The modified RSA models are compared to previously reported experimental protein adsorption on PEO grafted surfaces using SAMs as well as physically adsorbed copolymers of PEO. Our results show that accounting for underlying polymer arrangement and protein rearrangements post-adsorption significantly improves the predictions of the RSA model. The simulations are compared with experimental observations for the polymers: EG1OH, EG2OH, EG4OH, EG6OH, EG17OCH3, EG23OH, EG46OH, and EG115OH. The effect of polymer grafting density and polymer chain length of PEO on protein adsorption are also investigated. These results provide a more robust model for predicting protein surface adsorption on hydrophilic polymer coated surfaces as a function of the polymer chain length, grafting density, protein size, protein surface sub-diffusion, and polymer layer structure. This paves a path for the design of high performance anti-fouling surface coatings.
