Book excerpt: The submersed cultivation of organisms in sterile containments or fermenters has become the standard manufacturing procedure, and will remain the gold standard for some time to come. This book thus addresses submersed cell culture and fermentation and its importance for the manufacturing industry. It goes beyond expression systems and integrally investigates all those factors relevant for manufacturing using suspension cultures. In so doing, the contributions cover all industrial cultivation methods in a comprehensive and comparative manner, with most of the authors coming from the industry itself. Depending on the maturity of the technology, the chapters address in turn the expression system, basic process design, key factors affecting process economics, plant and bioreactor design, and regulatory aspects.

This book was released on 2015-08-28 with total page 224 pages. Available in PDF, EPUB and Kindle. Book excerpt: Amongst the mammalian producer cell lines, the Chinese hamster ovary (CHO) cell lines are of predominant importance in biopharmaceutical production. Thus, novel factors increasing overall productivity are sought and bear the potential to reduce the unit costs of a production process. Furthermore, the current patent situation for several therapeutic proteins demands innovative tools to at least maintain or preferentially increase the cost-effectiveness of their production processes. In this thesis, hitherto unknown factors were revealed by next generation sequencing of chemically mutated and selected CHO-K1 suspension cell lines. Two factors were proven to improve CHO-based production processes: cgrSnord78 and cgrTtc36. The Cricetulus griseus Ttc36 increases the integral as well as the maximal viable cell density and abolishes the cell-cell aggregation whilst cgrSnord78 improves the specific as well as volumetric productivity without significant impact on cell growth. Based on the present results and discussion, foundations for future research on these functionally unrevealed factors are laid. Hence, this work represents the first step towards the application of the genuine biomolecules cgrTtc36 and cgrSnord78 in biopharmaceutical protein production.

Book excerpt: The huge potential for gene therapy to cure a wide range of diseases has led to high expectations and a great increase in research efforts in this area, particularly in the study of delivery via viral vectors, widely considered to be more efficient than DNA transfection. In Viral Vectors for Gene Therapy: Methods and Protocols, experts in the field present a collection of their knowledge and experience featuring methodologies that involve virus production, transferring protocols, and evaluating the efficacy of gene products. While thoroughly covering the most popular viral vector systems of adenovirus, retrovirus, and adeno-associated virus, this detailed volume also explores less common viral vector systems such as baculovirus, herpes virus, and measles virus, the growing interest in which is creating a considerable demand for large scale manufacturing and purification procedures. Written in the highly successful Methods in Molecular BiologyTM series format, many chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and vital tips on troubleshooting and avoiding known pitfalls. Comprehensive and practical, Viral Vectors for Gene Therapy: Methods and Protocols provides basic principles accessible to scientists from a wide variety of backgrounds for the development of gene therapy viral products that are safe and effective.

Book excerpt: This book is the lasting product, a resource of up-to-date information in the scientific literature for the field of animal cell technology, as it was presented during a pleasant and stimulating meeting in Tylösand, Sweden, in June 2001. The title of the meeting, From Target to Market, indicates the usefulness of Animal Cell Technology during all steps in the pharmaceutical development process. Following the biotech products reaching the market, it shows an upward trend in the contribution of biotech products to total New Molecular Entity output in the nineties, which continued until 1996 when biotech represented 25% of the annual output. Since then the proportion has been decreasing. A perceived hurdle from a market perspective is that a protein per definition is biodegradable and thus requires intravenous, or for some drugs subcutaneous administration. New promising administration technologies such as pulmonary delivery were highlighted at this meeting. The emphasis on project selection prior to entry in the development phase has triggered a portfolio management using more extensive preclinical data before a development decision is taken. Animal cells have become a very important tool in the drug discovery process. The next generation of products will evolve from applications such as gene therapy, novel vaccines, cell therapy, and gene regulation. Animal cell technology has a major role to play in the post-sequence era.

Available in PDF, EPUB and Kindle. Book excerpt: The completion of the Human Genome Project and the rapid progress in cell bi- ogy and biochemical engineering, are major forces driving the steady increase of approved biotech products, especially biopharmaceuticals, in the market. Today mammalian cell products (“products from cells”), primarily monoclonals, cytokines, recombinant glycoproteins, and, increasingly, vaccines, dominate the biopharmaceutical industry. Moreover, a small number of products consisting of in vitro cultivated cells (“cells as product”) for regenerative medicine have also been introduced in the market. Their efficient production requires comprehensive knowledge of biological as well as biochemical mammalian cell culture fundamentals (e.g., cell characteristics and metabolism, cell line establishment, culture medium optimization) and related engineering principles (e.g., bioreactor design, process scale-up and optimization). In addition, new developments focusing on cell line development, animal-free c- ture media, disposables and the implications of changing processes (multi-purpo- facilities) have to be taken into account. While a number of excellent books treating the basic methods and applications of mammalian cell culture technology have been published, only little attention has been afforded to their engineering aspects. The aim of this book is to make a contribution to closing this gap; it particularly focuses on the interactions between biological and biochemical and engineering principles in processes derived from cell cultures. It is not intended to give a c- prehensive overview of the literature. This has been done extensively elsewhere.
