Book excerpt: Author's abstract: The Eastern Oyster, Crassostrea virginica, is an ecologically important species that plays a role in protecting shorelines, water filtration, and providing habitat and food for various fish species. Perkinsus marinus and Haplosporidium nesloni are two pathogens currently affecting oyster populations. These pathogens have both been documented as causes of mass mortality events along the East coast. They greatly diminish the health of oysters, resulting in the degradation of oyster tissue, reduction in growth and reproductive ability, and may cause death. In this study, wild oysters were collected from eleven locations along the Georgia coast to investigate the presence, intensity, and impact on the health of local populations. The condition of each oyster was calculated during processing, and qPCR was used to determine the presence and intensity of both P. marinus and H. nelsoni. Additionally, population genetic structure and kinship within and among populations was examined using 20 microsatellite loci. Results showed that >90% of oysters tested were infected by one or both pathogens, and 48.8% of these were cases of co-infection. Book excerpt: Author's abstract: The eastern oyster, Crassostrea virginica, is both an ecologically and economically important bivalve species that has experienced drastic population declines do to overharvest and habitat degradation. Populations that are impacted by anthropogenic stressors are further driven down by infections with protists like Haplosporidium nelsoni (MSX) and Perkinsus marinus (Dermo). In order to determine the prevalence and intensity of these detrimental infections, a survey was performed in Oyster Creek, Georgia. Of the oysters collected, 80% of them were infected with a single protist or both at relatively low intensities. The infection intensity was variable throughout the creek, whereas the infection prevalence was not variable. Microsatellites were used in order to determine kinship within Oyster Creek and it was found that oysters that were closely related (i.e. full/half/quarter siblings) were more likely to be located on the same reef patch than on different patches.

Book excerpt: Because of the continued development and production of a variety of nanomaterials and nanoparticles, their uptake and effects on the biota of marine ecosystems is a concern. In estuarine environments, where these nanomaterials are likely to accumulate, agglomeration rates, interaction with organics, and response to UV radiation are critical in determining the extent of their biological impact. Bivalve molluscs are filter feeders and are highly adapted to internalize nano and micro-sized particles through endocytosis. Because of their filtering abilities and abundance they are commonly used as indicator species for marine contamination. In this study adult eastern oysters, Crassostrea virginica, were exposed to a range of titanium dioxide nanoparticle (Ti02-NP) concentrations (5, 50, 500, and 5000 pg/L) in conjunction with natural UV radiation. In addition, isolated hepatopancreas, gill, and mantle tissues were isolated and exposed to the same Ti02-NP concentrations under similar UV conditions. Analysis of particle size using dynamic light scattering (DLS) reveled agglomeration rates increased with an increase in concentration. Concentration had a significant impact on agglomeration rates, which was in agreement with the dose dependent decrease in lysosomal stability in both the adult oysters as well as in the isolated tissue. Isolated tissues showed accumulation of titanium mainly in the hepatopancreas, which demonstrated a direct correlation to lysosomal stability. Measurements of lipid peroxidation (LPx) as an indicator of oxidative stress did not support the hypothesis that an increase in toxicity will result from exposure to UV radiation. It did however show decreased LPx in the gill and mantle tissue of the adult oysters, suggesting a possible increase in antioxidants in response to the TiC2-NPs. Generally, this study illustrates the possibility of low concentrations of TiO2-NPS to demonstrate sublethal toxicity on oysters, which might be enhanced due to UV radiation.

While cadmium (Cd) has been shown to alter specific immune defense mechanisms, the direct effect of Cd on the inactivation and degradation of bacteria in this animal is unknown. Furthermore, the roles of individual tissues in the distribution, inactivation and degradation of live bacteria introduced into the host remain unclear. First, we determined the effect of Cd on the immune responses of C. virginica at the organismal, cellular and molecular level. Following a chronic Cd exposure, oysters were injected in the adductor muscle with 105 live Vibrio campbellii. Quantitative real-time polymerase chain reaction (Q-PCR) was used to quantify the number of intact bacteria, while selective plating was used to quantify the number of injected bacteria that remained culturable within the tissues and fluids of the whole animal at 10, 30 and 60 min after injection. While Cd did not alter the oyster's ability to inactivate bacteria, Cd exposure induced a significant decrease in the numbers of circulating hemocytes and significant changes in the expression of selected genes as determined by microarray analysis. The gills, digestive gland and hemocytes mounted unique responses to chronic Cd exposure, yet the functional integrity of immune defense at the organismal level was maintained under these conditions. Second, we determined the relative contribution of individual tissues to the antibacterial response of C. virginica following injections of V. campbellii. Numbers of intact bacteria were used to identify sites of bacterial accumulation, while percentages of culturable bacteria were quantified to determine sites of bacteriostasis within the digestive gland, mantle, gills, adductor, gonadal tissues, labial palps and hemolymph at 10, 30, 60 and 120 min after injection. The gonadal tissues contained the greatest concentration of intact bacteria and the lowest percentage of culturable V. campbellii compared to all other tissues. We suggest that the gonadal tissues, when present, are the main site of bacterial accumulation and bacteriostasis. Overall, this study demonstrates the resilience of oysters to bacterial infections by their ability to inactivate bacteria.
