Studies of the cerebellum are now a central focus in neuroscience. During the last four decades, many laboratories worldwide have dedicated their research activities to understanding the roles of the cerebellum in motor control, cognitive processes and biology of mental processes, behavioral symptoms, and emotion. It is now accepted that the cerebellum acts as a cognitive operator in learning, perception, and attention. Moreover, major improvements in our assessment of in vivo cerebellar architecture using imaging techniques have occurred. A typical example is the accurate description of cerebellar anatomy during fetal development with MRI, a progress which has direct impacts on patient care. These advances have been associated with discoveries of new clinical disorders, in particular in the field of genetic ataxias. More than 20 new genes have been identified these last 10 years. Only for dominant ataxias, more than 30 diseases have now been unravelled. The number of ataxic disorders will increase with aging, the cerebellum being the structure of the brain with the most important loss of neurons with age. More than 300 different cerebellar disorders are encountered during daily practice, but we are missing a single source of information explaining their pathogenesis. Despite the immense amount of knowledge acquired about the cerebellar circuitry these last years, a large book covering the neuroscience of the cerebellum is missing. The goal of this endeavour is to bring up to date information relevant for basic science and also for clinical activities. The cerebellar cortex was from the first an object of close study in the electron microscope, repeating in modern cytology and neuroanatomy the role it had in the hands of RAMON y CAJAL at the end of the nineteenth century. The senior author vividly remembers a day early in 1953 when GEORGE PALADE, with whom he was then working, showed him an electron micrograph of a cerebellar glomerulus, saying "That is what the synapse should look like. " It is true that the tissue was swollen and the mitochondria were exploded, but all of the essentials of synaptic structure were visible. At that time small fragments of tissue, fixed by immersion in osmium tetroxide and embedded in methacrylate, were laboriously sectioned with glass knives without any predetermined orientation and then examined in the electron microscope. After much searching, favorably preserved areas' were studied at the cytological level in order to recognize the parts of neurons and characterize them. Such procedures, dependent upon random sections and uncontrollable selection by a highly erratic technique of preservation, precluded any systematic investigation of the organization of a particular nucleus or region of the central nervous system. It was difficult enough to distinguish neurons from the neuroglia.

Although specialists in pharmacology and developmental biology had been studying this problem many years before, this catastrophic episode triggered the passage of legislation which required that information about the teratogenicity of drugs be produced before the drugs could be available to the general public. Gross deformities in man produced by drugs are frequently difficult to reproduce in experimental animals and the changes which are produced in other animals are frequently not translatable to humans. The problem of evaluating the potential that drugs have to produce gross malformations is small, however, compared to the evaluation of subtle but permanent behavioral effects which drugs may exert upon the developing organism. Nevertheless, many experimental studies in recent years indicate that subtle biochemical changes produced by drugs on brain tissue during critical periods of fetal or early post natal maturation may become manifest subsequently as behavioral deviations in early childhood or adolescence. Hyperkinetic disorders, epilepsies and other developmental disabilities may have a subtle biochemical imbalance, perhaps drug induced, as an underlying factor. This symposium was organized with the intent of bringing to gether prominent investigators who are working in different aspects of brain development and who are interested in the effects of drugs on the developing brain in order to discuss their findings, pro pose new theories, and open new avenues for future research.

In particular, we felt that as the discrete anatomy of the cerebellum is quite well known, only certain aspects of the structure should be discussed here. For example, the organization of the pontocerebellar pathways, we felt, would be particularly interesting given the enormity of the system in higher vertebrates. Also of interest is the distribution and development of the synaptology and neurotransmitter properties in this cortex.
