Book excerpt: These volumes present the results of two studies undertaken for the Directorate General of Employment, Social Affairs and Education of the Commission of the European Communities. The aim was to review available data concerning the metabolism of a selection of toxic chemi cals in man and to develop mathematical models representing this meta bolism. Volume 1 is the report from the first study and presents re views of the data for each of the substances and proposes models for their metabolic behaviour. Volume 2 is the report of the second study and contains a review of new data relevant to the development of meta bolic models for each substance. The modelling of uptake and retention by the two major routes of uptake (the respiratory system and the gas trointestinal tract) is also reviewed. The implementation of the models in a computer code is described and results are presented illus trating organ concentrations and rates of excretion for typical regimes of exposure. vi 1 Foreword This report contains the results of a study undertaken by Associated Nuclear Services (ANS) for the Directorate General of Employment, Social Affairs and Education of the Commission of the European Communities (CEC). This study was entitled "Models for the Metabolism of Chemical Carcinogens in Man". vii 2 Summary This report includes results of a review of the metabolism of selected actual and potential human carcinogens and proposals for pharmacokinetic models to represent that metabolism.

Book excerpt: This report contains the results of a study undertaken by Associated Nuclear Services (ANS) for the Directorate General of Employment, Social Affairs and Education of the Commission of the European Communities (CEC). This study was entitled "Dynamics of Selected Toxic Materials in Man". vii 2 Summary This report presents results from the second phase of a study on the metabolism of selected actual and potential human carcin ogens. In the first phase of this work a comprehensive review of literature relating to the metabolism of these materials was under taken and pharmacokinetic models were proposed to represent that meta bolism. It was suggested that these models, and the reviews on which they were based, would provide information relevant to the development of monitoring programmes and in the interpretation of dose-response data. In the second phase of the study, attention has been concen trated on the implementation of the models proposed for the various substances reviewed, i.e. arsenic, beryllium, cadmium, lead, nickel, chromium, asbestos, benzene, vinyl chloride monomer, benzidine and its congeners, carbon tetrachloride and methyl iodide. However, this programme has been supplemented by an on-going review of the relevant literature on these substances and a detailed analysis of the applica bili ty of the pharmacokinetic models used for the organs of intake, i.e. the lung and gastrointestinal tract.

Book excerpt: Trichloroethylene is a chlorinated solvent widely used as a degreasing agent in industrial and manufacturing settings. It is also used as a chemical intermediate in making other chemicals and is a component of products such as typewriter correction fluid, paint removers, adhesives, and spot removers. In 2001, EPA issued a draft health risk assessment and proposed exposure standards for trichloroethylene. PA's Scientific Advisory Board (SAB) reviewed the draft and it was issued for public comment. A number of scientific issues were raised during the course of these reviews. Assessing the Human Health Risks of Trichloroethylene identifies and assesses the key scientific issues relevant to analyzing the human health risks of trichloroethylene, considering pertinent toxicologic, epidemiologic, population susceptibility, and other available information, including relevant published scientific literature, EPA's 2001 draft health risk assessment of trichloroethylene, scientific and technical comments received by EPA from public and private sources, and additional relevant information to be provided by the sponsoring agencies. This report highlights issues critical to the development of an objective, realistic, and scientifically balanced trichloroethylene health risk assessment. Guidance for hazard characterization of trichloroethylene is presented in Chapters 2 through 10. Chapter 2 provides guidance for evaluating large sets of epidemiologic data. In Chapter 3, the committee applies this guidance as an example in its evaluation of the epidemiologic data on trichloroethylene and kidney cancer, and this example should help guide evaluations of other cancer risks. Chapter 3 also assesses new information on the kidney toxicity of trichloroethylene and its metabolites and potential modes of action. Chapters 4, 5, 6, 7, and 8 evaluate the key issues regarding liver toxicity and cancer, reproductive and developmental toxicity, neurotoxicity, respiratory tract toxicity and cancer, and immunotoxicity, respectively. However, the committee's review focused on mode-of-action information to understand how trichloroethylene might affect certain processes differently in different species. Chapter 9 discusses susceptibility to trichloroethylene and its metabolites, and Chapter 10 describes important factors in considering trichloroethylene in mixtures. Physiologically based pharmacokinetic models are evaluated in Chapter 11, and guidance is provided on future directions for model development. Finally, Chapter 12 considers issues related to dose-response assessment and quantitative assessment of risk.

This book was released on 2004-06-04 with total page 226 pages. Available in PDF, EPUB and Kindle. Book excerpt: The EPA commissioned The National Academies to provide advice on the vexing question of whether and, if so, under what circumstances EPA should accept and consider intentional human dosing studies conducted by companies or other sources outside the agency (so-called third parties) to gather evidence relating to the risks of a chemical or the conditions under which exposure to it could be judged safe. This report recommends that such studies be conducted and used for regulatory purposes only if all of several strict conditions are met, including the following: The study is necessary and scientifically valid, meaning that it addresses an important regulatory question that can't be answered with animal studies or nondosing human studies; The societal benefits of the study outweigh any anticipated risks to participants. At no time, even when benefits beyond improved regulation exist, can a human dosing study be justified that is anticipated to cause lasting harm to study participants; and All recognized ethical standards and procedures for protecting the interests of study participants are observed. In addition, EPA should establish a Human Studies Review Board (HSRB) to evaluate all human dosing studiesâ€"both at the beginning and upon completion of the experimentsâ€"if they are carried out with the intent of affecting the agency's policy-making.
