This thesis will describe efforts in two areas that are consistent with this broad objective, concerning the synthesis of natural products, and the development of methods for the synthesis of designed molecules. Our desire to better utilise the Diels-Alder reaction - a powerful means for the formation of carbon-carbon bonds, sits as a guiding principle behind these investigations.Part 1 of this thesis concerns the synthesis of the ramonanin natural products: four dimeric structures formed biosynthetically through a Diels-Alder dimerisation reaction. Chapter 1 will review the prior art on natural products formed using a Diels-Alder dimerisation as a key step. The area has not been reviewed substantively thus far - something overdue given the rapid development in this area over the past 15 years. Biomimetic synthesis is a recurrent theme of the chapter, with the vast majority of syntheses covered to some extent inspired by an understanding of the target's biosynthetic origin.Chapter 2 will cover original research performed in the total synthesis of the ramonanin family of the natural products. The chapter begins with a discussion of the possible biosynthetic origins of the ramonanins, and the ways this might be applied in a biomimetic synthesis. After this, an account is given of the successful forward synthesis of the ramonanin natural products. This begins with an account of the synthesis of the ramonanin monomeric subunit, followed by a detailed discussion of the dimerisation event from which the natural products were formed. Part 2 concerns the use of tricarbonyliron-diene complexes as building blocks in the synthesis of reactive cross-conjugated hydrocarbons.Chapter 5 concerns a methods-driven investigation, aimed at establishing the basic means by which cross-conjugated hydrocarbons might be assembled from a tricarbonyliron-diene starting material. A range of 1-[3]dehydrodendralenes and 1-(Z)-[3]dendralenes were prepared using this general method - the first general method for the synthesis of either class of hydrocarbon.Chapter 6 concerns the repurposing of the methods of chapter 5 to greater ends. In the first part of the chapter, the synthesis of higher and greatly more reactive polyenes using these general methods is detailed. The second part of the chapter details the use of a 1-(Z)-[3]dendralenes in a Diels-Alder cascade - a model for future applications in target-driven synthesis that might follow.

Book excerpt: Natural product synthesis and methodology development have long enjoyed a symbiotic relationship with the structural complexity observed in natural products inspiring chemists to invent new reactions, as well as providing a proving ground for the utility of existing methods in complex settings. Chapter 1 of this dissertation provides a recent perspective on catalytic methodology developed to enable the synthesis of complex natural products. A focus on the last 20 years of organic chemistry demonstrates the state-of-the-art in methods development and its direct application to accessing complex natural products efficiently. Chapter 2 details our efforts to synthesize three sulfolipid natural products isolated from algae Ochromonas danica. Key features of our synthesis of danicalipin A include the application of our catalytic, enantioselective dichlorination of allylic alcohols and a diastereoselective boron allylation of the dichloroaldehyde fragment. Subsequent syntheses of two biosynthetic precursors of danicalipin A are presented. We also describe the results of an interdisciplinary collaboration enabled by the synthetic sulfolipids to investigate the biophysical properties of these exotic molecules. Chapter 3 describes our efforts towards the total synthesis of napyradiomycin natural product azamerone. Our synthesis required the development of a catalytic, enantioselective chlorocyclization reaction to access a key chlorinated benzopyran intermediate. Subsequent B-alkyl Suzuki cross-coupling between two enantioenriched fragments, followed by the late-stage installation of the pyridazine heterocycle via a tetrazine [4+2] cycloaddition yielded the natural product. This work constitutes the first enantioselective total synthesis of azamerone.

The development of a stereoselective iron (III)-mediated intramolecular cascade dearomatization of phenol allowed access to various spirocyclohexadienones and tropons in one step using potassium ferricyanide via single electron transfer (SET). Using this oxidative dearomatization reaction, the thesis focuses on three main parts. The first part is the synthetic efforts toward total synthesis of Harringtonolide. The second part focused on the synthesis of spiro[4.5]decane, a core structure in multiple natural products. The spiro compounds were prepared from ortho hydroxyphenyl nitro alkanes using SET reaction with good yield and diastereoselectivity. In third part, we worked towards the total synthesis of both Magellanine and Megalleninone by applying this novel methodology. The fourth part was the first total synthesis of Phaeocauliine D, and L which were achieved from commercially available methyl-4-methyl salicylate in 9-13 simple steps.

Mechanistic Studies, Optimization, and Further Applications of the Organocatalytic [alpha]-Hydroxymethylation of Aldehydes: Further developments in studies of the direct [alpha]-hydroymethylation of aldehydes employing the [alpha],[alpha]-diarylprolinol trialkylsilyl ether class of organocatalysts are described. This synthetic transformation has proven effective in expedient access to [beta]-hydroxycarboxylic acids and [delta]-hydroxy-[alpha],[beta]-unsaturated esters from aldehydes in moderate to excellent yields, excellent enantioselectivity, and compatibility across a wide range of funtional groups in the starting aldehyde. These studies have also been successful in identifying the critical reaction variables that influence the yield and enantioselectivity in the [alpha]-hydroxymethylation process including catalyst structure, pH of the reaction medium, purity of the reagents employed, nature of the buffer, along with standard reaction variables including the solvent, time, temperature, and mixing efficiency. In addition, the previously identified lactol intermediate has been further characterized and examined. The culmination of these studies has translated directly into an improved substrate scope, reproducibility, enantioselectivity, and yields of the described method. Finally, the described [alpha]-hydroxymethylation protocol has been applied to a multi-gram scale synthesis of the southern hemisphere of the apoptosis inducer, (-)-rasfon. Part II. Studies Toward the Total Synthesis of FK-506: An approach to the total synthesis of the immunosuppressant FK-506 is described. The synthetic strategy features the division of the macrocycle into four subunits allowing for a maximum degree of convergence in the forward synthesis. Improvements to the synthesis of a key intermediate vinyl bromide are described that have allowed for the multi-gram throughput of the southern portion of the macrocycle. Improvements have been made to the synthesis of the northern hemisphere of the macrocyle synthesis, again allowing for better material throughput. Finally, several coupling strategies have been explored and the culmination of these efforts have led to promising preliminary results on the unification of the northern and southern hemispheres, bringing the molecules synthesis to near completion"--Pages vii-viii.
