By 1978, it had become evident that the coronavirus genomic RNA was infectious (i. e. , positive strand), and by 1983, at least the framework of the coronavirus replication strategy had been per ceived. Subsequently, with the application of recombinant DNA techniques, there have been remarkable advances in our understanding of the molecular biology of coronaviruses, and a mass of structural data concerning coronavirus genomes, mRNAs, and pro teins now exists. More recently, attention has been focused on the role of essential and accessory gene products in the coronavirus replication cyde and a molecular analysis of the structure-function relation ships of coronavirus proteins. Nevertheless, there are still large gaps in our knowledge, for instance, in areas such as the genesis of coronavirus subgenomic mRNAs or the function of the coronavirus RNA-dependent RNA polymerase. The diseases caused by coronaviruses have been known for much longer than the agents themselves. Possibly the first coronavirus-related disease to be recorded was feline infectious peritonitis, as early as 1912. The diseases associ ated with infectious bronchitis virus, transmissible gastroenteritis virus, and murine hepatitis virus were all well known before 1950.

Normal cell growth and division, tis sue differentiation, and organism development all depend on a strictly ordered progres sion of specific events. Perturbation of the control of these processes, for example by ex posure to harmful chemicals or infection with viruses leads to aberrant forms of meta bolism, often resulting in malignancies and cell death. One of the most challenging problems in biology is to derme at the molecular level the mechanisms that govern gene function in higher organisms, including ultimately man. This goal serves to unify the diverse efforts of many investigators, whether studying the precise patterns of embryo genesis, the loss of control that occurs during neoplastic growth or the redirection of biosynthetic pathways in virus-infected cells. Recently there has been remarkable and exciting progress toward understanding the molecular biology of eukaryotic expression. Much of this rapidly increasing new infor mation has come from studies of animal virus systems. Just as investigations of the relatively simple, rapidly assayed, and easily manipulated bacteriophages lead to basic discoveries about prokaryotic cells, analyses of animal viruses and their interactions with host cells have provided fundamental information about how eukaryotic nucleic acids are organized for regulated replication, transcription, and translation. For example, the small genome of SV , like cellular DNA in chromatin, is associated with histones to 40 form nucleosomal arrays (Griffin 1975).

It also became apparent that there was a need for an international assembly of this sort at regular intervals. These workshops not only encourage stim ulating discussion and constructive thinking but also result in increased collaboration and productive liaison between laboratories with common interests. Hence a ten-day advanced studies institute course was organised in Edinburgh from 11-21 July 1979, and in this volume we have published the contributions given by the invited speakers. The subjects discussed covered most areas of plant molecular biology and the lecturers were asked to balance a review of their chosen subject with the results of their own recent research and likely future advances. Probably the most important technical advance since the previous meeting of this group in Strasbourg, was the application of restriction enzyme analysis and cloning techniques. This is illustrated in many of the published lectures and was the basis for many of the more informal discussion sessions.
