Available in PDF, EPUB and Kindle. Book excerpt: Laser desorption mass spectrometry has been considered as a potential new method for fast DNA sequencing. Our approach is to use matrix-assisted laser desorption to produce parent ions of DNA segments and a time-of-flight mass spectrometer to identify the sizes of DNA segments. Thus, the approach is similar to gel electrophoresis sequencing using Sanger's enzymatic method. However, gel, radioactive tagging, and dye labeling are not required. In addition, the sequencing process can possibly be finished within a few hundred microseconds instead of hours and days. In order to use mass spectrometry for fast DNA sequencing, the following three criteria need to be satisfied. They are (1) detection of large DNA segments, (2) sensitivity reaching the femtomole region, and (3) mass resolution good enough to separate DNA segments of a single nucleotide difference. It has been very difficult to detect large DNA segments by mass spectrometry before due to the fragile chemical properties of DNA and low detection sensitivity of DNA ions. We discovered several new matrices to increase the production of DNA ions. By innovative design of a mass spectrometer, we can increase the ion energy up to 45 KeV to enhance the detection sensitivity. Recently, we succeeded in detecting a DNA segment with 500 nucleotides. The sensitivity was 100 femtomole. Thus, we have fulfilled two key criteria for using mass spectrometry for fast DNA sequencing. The major effort in the near future is to improve the resolution. Different approaches are being pursued. When high resolution of mass spectrometry can be achieved and automation of sample preparation is developed, the sequencing speed to reach 500 megabases per year can be feasible.

Book excerpt: The USP–NF is a combination of two compendia, the United States Pharmacopeia (USP) and the National Formulary (NF). It contains standards for medicines, dosage forms, drug substances, excipients, biologics, compounded preparations, medical devices, dietary supplements, and other therapeutics. The current version of USP–NF standards deemed official by USP are enforceable by the U.S. Food and Drug Administration for medicines manufactured and marketed in the United States.

Book excerpt: MALDI-TOF-MS has the ability to streamline identification of microorganisms in the microbiology lab. In this project, we focused on two main aims. First, the implementation of this technology for direct blood culture identification. Within this aim we determined the limit of detection for the protein purification method we planned on using in our laboratory, determined handling conditions for the samples, and evaluated the MALDI-TOF-MS for concordance with our current workflow and reports. Second, customization of the commercial library for Cryptococcus gattii was performed. Within this aim, we improved representation of the species in the MALDI-TOF-MS library, and also explored ways to determine the subspecies based on gathered spectra.

Download or read book Time of Flight Spectrometry of DNA for Rapid Sequence Determination. Final Technical Report written by and published by . This book was released on 2001 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: The objective of this project was to exploit the demonstrated capability to ablate intact DNA of very high mass from thin frozen films of aqueous solutions to achieve time-of-flight mass spectrometric readout of Sanger dideoxy-terminated sequencing mixtures. The authors also pursued a more conventional approach to DNA mass spectrometry, using a matrix-assisted laser desorption/ionization (MALDI) approach in which the desorption is driven by pulsed laser ablation of an organic matrix which encapsulates the DNA and strongly absorbs the laser energy. Efforts in this area were directed largely at understanding fundamental issues in the desorption process, because in all such approaches it has been difficult to ablate intact DNA beyond[approximately] 100-mer with acceptable resolution and sensitivity. In addition the authors looked into clean-up technologies, which will be mandatory for any mass spectrometric approach, and demonstrated a feasible clean-up scheme using affinity capture techniques. In this approach to affinity cleanup first gained experience using protein substrates, and this led to side-projects which included the first demonstration of a mass spectrometric immunoassay and subsequent development of an ultra-rapid endoprotease mapping technology using proteases covalently coupled to the mass spectrometer probe surface.
