This book was released on 2018-02-13 with total page 230 pages. Available in PDF, EPUB and Kindle. Book excerpt: ODE/PDE Alpha-Synuclein Models for Parkinson’s Disease discusses a mechanism for the evolution of Parkinson’s Disease (PD) based on the dynamics of the protein a-synuclein, a monomer that has been implicated in this disease. Specifically, a-synuclein morphs and aggregates into a polymer that can interfere with functioning neurons and lead to neurodegenerative pathology. This book first demonstrates computer-based implementation of a prototype ODE/PDE model for the dynamics of the a-synuclein monomer and polymer, and then details the methodology for the numerical integration of ODE/PDE systems which can be applied to computer-based analyses of alternative models based on the reader's interest. This book facilitates immediate computer use for research without the necessity to first learn the basic concepts of numerical analysis for ODE/PDEs and programming algorithms Includes PDE routines based on the method of lines (MOL) for computer-based implementation of ODE/PDE models Offers transportable computer source codes for readers, with line-by-line code descriptions relating to the mathematical model and algorithms Authored by a leading researcher and educator in ODE/PDE models

Available in PDF, EPUB and Kindle. Book excerpt: Parkinson's disease (PD) results primarily from the death of dopaminergic neurons in the substantia nigra. Current PD medications treat symptoms; none halt or retard dopaminergic neuron degeneration. The main obstacle to developing neuroprotective therapies is a limited understanding of the key molecular mechanisms that provoke neurodegeneration. The discovery of PD genes has led to the hypothesis that misfolding of proteins and dysfunction of the ubiquitin-proteasome pathway are pivotal to PD pathogenesis. Previously implicated culprits in PD neurodegeneration, mitochondrial dysfunction, and oxidative stress may also act in part by causing the accumulation of misfolded proteins, in addition to producing other deleterious events in dopaminergic neurons. Neurotoxin-based models have been important in elucidating the molecular cascade of cell death in dopaminergic neurons. PD models based on the manipulation of PD genes should prove valuable in elucidating important aspects of the disease, such as selective vulnerability of substantia nigra dopaminergic neurons to the degenerative process.

Book excerpt: Parkinson’s disease is the second most prevalent neurodegenerative disease and is characterized by the irreversible loss of dopamine neurons. Despite its high prevalence in society and many decades of research, the origin of the pathogenesis and the molecular determinants involved in the disorder has remained elusive. Confounding this issue is the lack of experimental models that completely recapitulate the disease state. The identification of a number of genes thought to play a role in the cell death, and development of both toxin and genetic models to explore the function of the genes both in unaffected and diseased cells are now providing new insights into the molecular basis of the neurodegeneration, as well as therapeutic approaches. In this reference, we will describe the advances and the advantages that various invertebrates, cell culture, rodents, and mammals provide in the identification of the molecular components and mechanisms involved in the cell death, and outline the opportunities that these systems provide in drug discovery. Comprehensive and critical assessment of the utility of various model systems to identify the molecular components and pathways involved in Parkinson’s disease Describes the power of toxin and genetic models to identify novel therapeutic targets and compounds that can be used in PD Current overviews of current status of PD research and discovery from bench-to-bedside Provides novel insights and views on where the future of PD research may lead Provides a powerful teaching tool and template to explore the utility of model systems to identify molecular pathways, molecular targets, and therapeutics that are applicable to a variety of neurological diseases

Book excerpt: Despite being described about 200 years ago, the pathophysiology of Parkinson’s disease (PD) is not well understood due to its complex etiology and heterogeneity of symptoms and progression. Animal models provide a great opportunity to follow the progression of neurodegeneration and to screen potent therapeutic molecules, knowledge of which can be critical for different levels of clinical trials. Contemporary health technologies are coming in handy to accelerate the pace of developing novel and refined therapeutic strategies for PD. This book, Parkinson’s Disease - Animal Models, Current Therapies and Clinical Trials, provides a comprehensive overview of PD, presenting information on animal models of PD and contemporary therapeutic strategies, health technologies, clinical trials, and their influence on the quality of life of patients with PD.

Book excerpt: Download PDF Download EPUB Parkinson's disease is a neurodegenerative disorder that affects 1.5% of the global population over 65 years of age. The hallmark feature of this disease is the degeneration of dopamine neurons in the substantia nigra pars compacta and a consequent striatal dopamine deficiency. The pathogenesis of Parkinson's Disease remains unclear. Despite tremendous growth in recent years in our knowledge of the molecular basis of Parkinson's Disease and the molecular pathways of cell death important questions remain regarding why are substantia nigra cells especially vulnerable, which mechanisms underlie progressive cell loss or what do Lewy bodies or alpha-synuclein reveal about disease progression. Understanding the different vulnerability of the dopaminergic neurons from midbrain regions and the mechanisms whereby pathology becomes widespread are primary objectives of basic and clinical research in Parkinson's Disease. This e-Book discusses the etiopathogenesis of Parkinson's Disease, presenting a series of papers that provide up-to-date, state-of-the-art information on molecular and cellular mechanisms involved in the neurodegeneration process in the disease, the role of activation of functional anatomical organization of the basal ganglia and in particular habitual vs goal directed systems as a factor of neuronal vulnerability, the possibility that Parkinson's Disease coulb be a prion disease and how genetic factors linked to familial and sporadic forms of PD. We hope that this e-Book will stimulate the continuing efforts to understand the cell and physiological mechanisms underlying the origin of Parkinson's Disease.
