Dans le but d'apporter notre contribution au développement de ce type de supports, nous avons préparé une famille de supports à base de silice passivée avec une phase stationnaire de type polymère greffée par la bêta-cyclodextrine. Les cyclodextrines appartiennent à la famille des molécules dites cages car elles comportent au sein de leur structure une cavité indéformable et hydrophobe qui leur permet d'emprisonner ou d'encapsuler d'autres molécules. Notre choix s'est porté sur les poly(alkylamines) comme polymères. Ces polymères sont des polybases de structure relativement simple, qui présentent de fortes interactions avec les gels de silice. Nous avons utilisé la poly(éthylèneimine), la poly(allylamine) et la poly(vinylamine). Ce dernier polymère n'étant pas commercial, a été synthétisé. Ces polymères ont été fonctionnalisés par la bêta-cyclodextrine. Ces polymères greffés ont alors servi de matériaux de base dans la préparation de nouveaux supports pour la chromatographie liquide haute performance. Les phases stationnaires préparées nous ont permis de séparer des dérivés disubstitués du benzène (nitrophénol, acide nitrobenzoïque, crésol) et d'autres molécules telles que la caféine, la théophylline, l'indométhacine le mécanisme de rétention est basé non seulement sur la formation de complexe d'inclusion, mais aussi sur des interactions de type acide-base et des interactions stériques. Nous avons tenté d'établir une corrélation entre les caractéristiques physique et chimique de nos supports et leur comportement chromatographique. La quantité et la nature du polymère déposé, la teneur en cyclodextrine, ainsi que certains paramètres chromatographiques (la nature et la proportion de la phase éluante, longueur de la colonne...) ont été étudiés.

Les synthèses organiques décrites dans ce manuscrit permettent d’obtenir des dérivés de cyclodextrine possédant une ou deux unités glucosidiques différentes des autres. Ces composés sont préparés selon deux stratégies distinctes : la voie directe par dé-O-méthylation de l’α-cyclodextrine perméthylée pour obtenir des composés mono et di hydroxylés, ou après dérivatisation, des composés acétylés et trifluoroacétylés ; et la voie indirecte, par homologation de l’α-cyclodextrine perméthylée (ouverture du cycle de la cyclodextrine, insertion d’une ou deux unités glucosidiques différentes et macrocyclisation). Ces nouvelles molécules, ainsi synthétisées, sont caractérisées par résonance magnétique nucléaire et chromatographie liquide haute performance couplée à un spectromètre de masse. La dernière partie de ce travail présente l’application de ces molécules, diluées dans un polysiloxane (OV-1701-OH), comme nouvelles phases stationnaires chirales pour la chromatographie en phase gazeuse énantiosélective. L’analyse de ces sélecteurs chiraux déposés dans des capillaires de silice fondue nous a permis d’évaluer leur énantiosélectivité vis à vis de solutés chiraux en étudiant les phénomènes d’inclusion. Cette étude préliminaire, par injections de séries homologues de dérivés de l’hydantoïne, du 1-phényléthanol, de γ-lactones et des solutés de structures variées, nous a permis d’apporter une première évaluation des mécanismes de reconnaissance chirale. Les résultats chromatographiques ont permis de mettre en évidence qu’une des phases, la 3-monotrifluoroacétyl-α-cyclodextrine perméthylée, présente une énantiosélectivité significativement différente de l’α-cyclodextrine perméthylée prise comme phase de référence.
