The results of this investigation were summarized in 1959 in the book, On Neural Mechanisms of Spatial Orientation in Higher Vertebrates. In the last ten years we have set ourselves the goal of a multifaceted study of the origin of image memory in different representatives of the vertebrate series, from fish to monkeys inclusively.

Given the temperament of those who work in the field of color vision there seems little doubt that heated debates will continue, but for the present at least, the subject seems to be as close to settled as such things can be in science." How the brain represents color remains one of the most controversial topics in neurophysiology. We know that color is represented through an opponent mechanism, demonstrated by the fact that some colors are exclusive of others. Yet how these antagonistic chromatic axes are represented in the cortex has been a mystery. Dr. Conway mapped the spatial and temporal structure of the cone inputs to single neurons in the primary visual cortex of the alert macaque. Color cells had receptive fields that were often Double-Opponent, an organization of spatial and chromatic opponency sufficient to form the basis for color constancy and spatial color contrast. Almost all color cells gave a bigger response to color when preceded by an opposite color, suggesting that these cells also encode temporal color contrast. In sum, color perception is likely subserved by a subset of specialized neurons in the primary visual cortex. These cells are distinct from those that likely underlie form and motion perception. Color cells establish three color axes sufficient to describe all colors; moreover these cells are capable of computing spatial and temporal color contrast - and probably contribute to color constancy computations - because the receptive fields of these cells show spatial and temporal chromatic opponency.

How does sensory information get transformed into a spatially organized representation about our current state in the world and how is this abstract representation utilized when producing motor commands that lead to successful navigation? When navigating in a complex environment, all animals must encode information about their position and orientation in a rich sensory environment. In vertebrates this may occur by means of distributed activity across several navigation circuits located in the hippocampal formation. Arthropods, however, lack a hippocampal formation and thus it is unclear what circuits mediate navigation. A wide range of studies indicate that the central complex (CX), is not only involved in directional sensory information processing and the control of motor commands, but also plays a role in orientation coding in polarized light guided navigation and landmark orientation. All of these neural mechanisms point in the direction that single neurons in the CX might be directly involved in head direction coding, as well as other aspects of adaptive navigation. In the work described in this dissertation I used multi-channel extracellular recording techniques to uncover the neural correlates of head direction coding and spatial context cues in the cockroach CX. Specifically, I used tetrodes to record the activity of single neurons in the CX while the animal was passively rotated around on a platform surrounded by a circular arena (Chapter 2). In the same setting I also recorded local field potentials (LPFs) in the CX to uncover how navigational information modulates the network’s activity in a more global manner (Chapter 3). I found that single units, as well as LFPs in the cockroach CX encode the animal’s head direction relative to a salient visual cue. However, when landmarks are not available to the animal, both single neuron and network-level activity can rely upon idiothetic motion cues to update the animal’s relative heading in a landmark-free setting. In addition to these results, I found that a subpopulation of single neurons and some of the LFP frequency bands encoded the rotation direction history of the animal, a common spatial context cue. These results suggest that the CX navigation circuit is involved in environmental context discrimination processes that might be utilized by spatial memory circuits in the insect brain.Taken together, these results provide a solid foundation for future studies on the neural basis of adaptive navigation in insects. By placing these results in a wider context of adaptive navigation in all animals and by comparing them to the mechanisms described in mammalian navigation circuits, these data also contribute to a broad comparative approach to understand the general principles of navigation, as well as the diversity of the neural substrates of navigation across evolutionarily distinct animals.

Book excerpt: Human and non-human primates (NHPs) have the ability to see in three-dimensional (3D) space, allowing for successful navigation and interaction with objects in the environment. The robust (i.e., accurate and precise) 3D visual perception is achieved by the reconstruction of 3D scene information from two-dimensional (2D) retinal projections. However, it remains unclear where in the brain this reconstruction occurs and how the 2D signals were transformed into robust 3D representations. This dissertation assessed perceptual and neuronal representations of 3D vision in macaque monkeys using a planar surface orientation discrimination task with different spatial poses (i.e., orientation and distance). First, I evaluated if NHPs dynamically integrate visual cues for 3D surface orientation perception. The results showed that the perceptual bias was small across the surface poses, whereas sensitivity varied with the pose-dependent reliabilities. The prediction of perceptual sensitivity by an optimal integration model using the cue-isolated data matched the observed sensitivity to combined-cue stimuli. These findings demonstrate that NHPs optimally integrate stereoscopic and perspective cues to achieve robust 3D perception by dynamically reweighting all available information. Second, I evaluated the neural mechanism of dynamic cue reweighting with reliability in the caudal intraparietal (CIP) area. Neuronal responses were recorded using laminar probes while monkeys performed the discrimination task. The results showed that neural responses to combined-cue were well described by the linear weighted summation of the cue-isolated responses. The neural weight ratio between isolated cues was related to the ratio of perceptual sensitivity between the object poses. Finally, I investigated the relationships between the robustness of 3D sensory representations, choice-related activity, and motor-related activity in CIP. The results showed that the neurons with more robust 3D representations preferentially coupled with choice-related activity. Neurons with choice-related activity further increased the robustness of 3D selectivity after the onset of choice activity. Furthermore, I found that the sensorimotor associations between 3D visual representation and saccade-related activity were experience-dependent and stronger for neurons that carried choice-related activity. In sum, these results uncovered a neural basis for visual cue integration as well as systematic links between the high-level sensory representations, choice-related activity, and motor-related activity in CIP.
