Book excerpt: ABSTRACT: N-Thiolated beta-lactams are a new class of anti-MRSA and anti-Bacillus agents that have recently been reported by our laboratories. From previous studies performed in our laboratories, it is believed that the N-thiolated beta-lactams exert their antimicrobial activity through a unique mode of action that is completely unlike that of classical beta-lactam antibiotics. In the first chapter of this dissertation, a review of previously prepared N-thiolated beta-lactam analogues and their mode of action is presented. In the second chapter, the synthesis of seven different C3-oxygenated derivatives is described. These analogues were tested for antibacterial activity against Staphylococcus aureus, nine different strains of MRSA, and seven different species of Bacillus. The results of the antibacterial testing will be discussed in relation to the differences in the structures of the analogues. In chapter 3, the design and synthesis of two new, functionalized caprolactone monomers are presented. These monomers were subsequently cooligomerized with epsilon-caprolactone, as described in chapter 4. N-thiolated beta-lactams were attached to the functionalized oligomers. These antibiotic containing oligomers were then screened for activity against MSSA, MRSA, and Bacillus. The results of these biological tests and their implications for future experiments are discussed.

The synthesis and structure-activity relationship of these analogues is discussed in Chapter III. Moreover, microscopy and 3H pulse-labeling studies, which are described in Chapter IV, demonstrate that N-methylthio beta-lactams appear to be inhibitors of protein biosynthesis.

Book excerpt: ABSTRACT: Work in the Turos group over the past five years has focused on the development of N-alkylthio beta-lactams, which show antibacterial activity against Staphylococcus (including MRSA), Bacillus, and others. These compounds do not function in the manner of the traditional beta-lactam antibiotics, but were thought to undergo an intracellular thiol-transfer to coenzyme A. In expanding the SAR of these novel compounds, it was found that N-alkylthio-2-oxazolidinones also exhibit antibacterial activity. Although CoA acts as the thiol-redox buffer in the genera most susceptible to the N-alkylthio beta-lactams, studies on Coenzyme A disulfide reductase (CoADR) show that the redox buffer is not affected by these compounds. However, the recent finding that fatty acid synthesis is affected by the N-alkylthio beta-lactams led to the discovery that these compounds act as prodrugs, and that the asymmetric CoA disulfides produced by in vivo thiol transfer are potent inhibitors of beta-ketoacyl-acyl carrier protein synthase III (FabH) through a novel thiol-disulfide exchange with the active site cysteine. Lactams 2a and 2g were also found to be potent inhibitors of this enzyme. In an effort to produce a CoA mixed-disulfide mimic which could cross the cell membrane, a series of simple aryl-alkyl disulfides were synthesized and tested against E. coli, S. aureus, and B. subtilis. Several of these compounds were found to be very potent antibacterials both in vitro and in vivo, with MICs less than 0.125 micrograms/mL. Comparison of the activities of these disulfides with those of acyl-CoA analogs and CoA mixed disulfides support the assertion that FabH is indeed the cellular target of these potent new compounds.

Book excerpt: These results led to the idea about examining a fO-lactam conjugated polymer as a possible pro-drug delivery method, which is the focus of Chapter 3. To synthesize the initial drug-polymer candidate, microemulsion polymerization of an acrylate-substituted lactam was done in aqueous solution to form hydrophilic polymeric nanoparticles containing the highly water-insoluble solid antibiotic, N-methylthio fO-lactam. This method has advantages over the conventional emulsion polymerization methods because a solid co-monomer (fO-lactam drug) can be utilized. SEM studies show that these polymeric nanoparticles have a microspherical morphology with nano-sizes of 40-150 nm. The N-thiolated fO-lactam containing nanoparticles display potent anti-MRSA activity at much lower drug amounts compared with free lactam drug, penicillin G or vancomycin.

We hypothesized that active N-thiolated beta-lactams can damage DNA and induce apoptosis in human breast cancer cells in nude mice. In this final report. we have first evaluated potencies of many novel synthetic beta-lactams to inhibit proliferation and induce apoptosis in human cancer cell. We then determined whether several of these beta-lactam, L1, HY2, HY14, HY15 and SC4, could damage breast tumor cell DNA and inhibit breast tumor growth in vivo. We have found that these beta-lactams inhibited growth of implanted MDA-MB-231 breast tumors in a concentration-dependent manner, associated with their DNA-damaging activities. Our studies have provided strong support for proof-of-concept of the potential use of these N-thiolated beta-lactams in breast cancer prevention and treatment.
