Recent advances in the understanding of the molecular biology and genomics of MPNs have led to the development of novel targeted treatment approaches; however, the high rate of life-threatening vascular events and the high risk of disease progression with myelofibrotic and leukemic transformation continue to present challenges in the management of MPNs. Thereby, there remains a need for novel treatment strategies. Recently, research has revealed a role for clonal hematopoiesis, driver mutations and inflammation in thrombosis risk, which may provide a basis for novel thrombosis-prevention strategies.
