Book excerpt: We seek to identify the phases in the cell cycle during which steroid-activated estrogen and androgen receptors are normally transcriptionally active and to determine whether this cell cycle regulation of receptor activity is maintained when cancer-inducing non- steroidal agents activate the receptors. Our hypothesis is that the activities of steroid-induced ER and AR are controlled by cell cycle regulators and that cancer-inducing, non-steroidal activators bypass or alter this regulation of receptor activity giving rise to aberrant ER and AR function. Through these studies we are gaining understanding of how estrogen and androgen receptors may contribute to the onset of tumors and to the development of hormone independent growth. We have found that the activity of the androgen and estrogen receptors is indeed regulated throughout the cell cycle with no activity of the androgen receptor in the G2/Phase phase and low activity of the estrogen receptor in cells which are mainly in S phase. Furthermore, our data indicate that non-steroidal activators of the receptors may act in a cell type and promoter dependent manner as we do not detect receptor activation by growth factors in our system.

Book excerpt: The overall goal of our laboratory is to elucidate the mechanism of signal transduction and transcriptional regulation by the estrogen receptor (ER), a ligand- regulated transcription factor that plays a critical role in the development, progression and hormone responsiveness of breast cancer cells. Combining genetic, molecular and biochemical approaches, we have identified the Hsp9O-associated co-chaperone, p23, as a key regulator of ER action and have also determined that ER is phosphorylated and regulated by the cyclin A/Cdk2 complex. Understanding of the communication between ER and these regulator factors is fundamental to understanding the mechanism of ER- regulated gene expression and may reveal novel points of intervention to be exploited in the development of new therapies for breast cancer.

We provide evidence that alteration in the regulation of cyclinA/Cdk2 complex lead to changes in hormone-dependent and hormone-independent transcriptional enhancement by ER This is accomplished, at least in part, through the direct phosphorylation of ER S104, S106 and S118 by the cyclinA/Cdk2 complex. Since alterations in cydin dependent kinase regulatory proteins are a common feature in breast cancer, these findings provide a framework for understanding the mechanism by which CDK dysregulation can directly affect ER signaling through alterations in receptor phosphorylation.

Furthermore, we show that a second protein, Apurinic/apyrimidinic endonuclease 1 or redox factor-1 (Ape1/Ref-1), interacts with ERalpha, promotes the ERalpha-ERE interaction, influences ERalpha-mediated transactivation, and selectively associates with endogenous, estrogen-responsive genes in MCF-7 cells. Our findings suggest that NM23-H1 and Ape1/Ref-1 are instrumental in modulating expression of estrogen-responsive genes. Interestingly, we demonstrate that Ape1/Ref-1 and NM23-H1, as well as the oxidative stress proteins Cu/Zn superoxide dismutase (SOD1), thioredoxin (Trx) and protein disulfide isomerase (PDI) are overexpressed in human breast cancer tissues. These studies provide a novel link between DNA repair, redox regulation and modulation of ERalpha-mediated transcription.

In addition, estrogen has been shown to cause breast cancer cell proliferation. A common form of breast cancer treatment involved selective estrogen receptor modulation. A molecular explanation of how this works is that estrogen regulates and binds to estrogen receptor (ER), a ligand-dependent transcription factor. ER associated with estrogen induces gene transcription by translocating into the nucleus and binding to estrogen response element. ER also recruits cofactor proteins, which results in chromatin remodeling and gene expression regulation through interacting with histone acetylases or transcriptional machinery. Most studies have focused on the study of how ER can activate gene transcription. Recently, ER has been shown to also repress gene transcription. my research has two parts. The first part was to find genes that were down regulated by estrogen in order to increase the data pool of genes down-regulated by estrogen. Four target genes, ARGN, MGC16169, CALML5, and NFIB are suspected to be involved in down-regulation by ER. However, after conducting validation tests, these genes were determined to not be repressed. The second part includes characterizing the specific effects of co-repressors NCoR, NRIP1, and SMRT. Removal of these co-repressors and subsequent effect of their removal on following four ER target sites, HES1, PSCA, SLC35A1, and MME were studied. A knock down of a single co-repressor did not affect the majority of transcriptional activity in ER repressed target genes. A triple knock down was also conducted in hope that removal of multiple co-repressors might affect repression. However, the triple knock down was a failure and future experiments need to be done. Understanding the mechanisms of ER transcriptional repression would significantly aid the creation of effective treatments for breast cancer.
